The classic nonhomologous end-joining (c-NHEJ) pathway is largely responsible for repairing double-strand breaks (DSBs) in mammalian cells. XLF stimulates the XRCC4/DNA ligase IV complex by an unknown mechanism. XLF interacts with XRCC4 to form filaments of alternating XRCC4 and XLF dimers that bridge DNA ends in vitro, providing a mechanism by which XLF might stimulate ligation. Here, we characterize two XLF mutants that do not interact with XRCC4 and cannot form filaments or bridge DNA in vitro. ...

XRCC4 and DNA Ligase 4 (LIG4) form a tight complex that provides DNA ligase activity for classical non-homologous end joining (the predominant DNA double-strand break repair pathway in higher eukaryotes) and is stimulated by XLF. Independently of LIG4, XLF also associates with XRCC4 to form filaments that bridge DNA. These XRCC4/XLF complexes rapidly load and connect broken DNA, thereby stimulating intermolecular ligation. XRCC4 and XLF both include disordered C-terminal tails that are functionally ...

Abstract DNA double strand breaks (DSBs) are induced by external genotoxic agents (ionizing radiation or genotoxins) or by internal processes (recombination intermediates in lymphocytes or by replication errors). The DNA ends induced by these genotoxic processes are often not ligatable, requiring potentially mutagenic end-processing to render ends compatible for ligation by non-homologous end-joining (NHEJ). Using single molecule approaches, Loparo et al. propose that NHEJ fidelity can be maintained ...

Phlebotomines of the genus Lutzomyia are important vector for species of Leishmania, the etiological agent of leishmaniasis. The aim of this study was to assess the diversity of sandflies in an endemic area for Visceral Leishmaniasis (VL). Sampling was performed in four localities on the coast of the municipality of Goiana, state of Pernambuco, Northeastern Brazil. Specimens were collected monthly on three consecutive nights, from November 2017 to October 2018. CDC light traps were installed at a ...

The presence of insects and mice in the hospital environment offer risks to patients and health professionals, as they can transmit diseases. The present work aims emphasize the importance of controlling pests and disease vectors in hospital settings, identifying the most common pests and vectors, the measures of prevention and control and expose sanitary legislation. Was performed an exploratory research, literature and documents on the theme. After collecting data, we describe some aspects of ...

This paper deals with a system composed of a three-level Neutral-Point- Clamped inverter fed by photovoltaic panels. The system is able to operate adequately even under partial shading operation by controlling the dc-link neutral point injected current. System model, pulse width modulation strategy, maximum power point tracking, and control logic for balancing the dc-link capacitors voltages are presented. Analysis of harmonic distortion, thermal stress, limits of control and power losses are included. ...

The Visceral leishmaniasis (LV) is widely distributed in the territory of Pernambuco, although data on the dynamics and epidemiological characteristics of the disease in some areas are still fragmented and incipient; this is the case of the municipality of Goiana, where cases of visceral leishmaniasis in dogs and in humans have been recorded. The present study aims to analyze the behavior of the disease in the municipality of Goiana – PE in the period from 2007 to 2015 through the use of epidemiological ...

This paper presents a capacitor voltage balancing method for the five-level diode-clamped inverter using level-shifted pulse-width modulation (LS-PWM), including the voltage balancing of DC-link capacitors. The proposed system has a reduced number of components for mitigation of losses. The DC-Link capacitor voltage is regulated by adjusting the switching duty cycles of DC/DC converter. Simulation results are presented to verify the validity of this method.
