The thiation of methyl substituted oxindoles (I) with phosphorous pentasulfide gave the corresponding 2-indolinethiones (II). The 2-indolinethione (II) having more than one hydrogen at 3-position, however, were sensitive towards phosphorous pentasulfide to produce the indolic compounds such as III, IV, V and VI. On the other hand the 2-indolinethione (VIII) prepared from VII with phosphorous pentasulfide, was stable towards the reagent. Reactivity of 2-indolinthione (IIb) was compared with the oxindole ( ...

2-Aminoindoles (2 and 4) have been prepared by the reaction of 2-indolinethiones (1) with primary and secondary aliphatic amines. The reaction of 1b with ethyleneimine produced the S-alkylated product 5, and with phenylhydrazine compound 9. The reaction 1b with aniline and 10 with piperidine failed to give an 2-aminoindole derivative or 12. The UV and NMR spectra of the free base 2 shows the presence of a tautomeric mixture of 13 and 14 in bases 2c, 2g, 2h and 2i. The free base 2b in CDCl3 exists ...

Several lysosomal enzyme activities in cultured lymphoid cell lines were studied during 3 phases of cell culture; logarithmic growth phase, stationary phase and decline phase. Enzyme induction during cell growth was found in N-acetyl-hexosaminidase, beta-galactosidase and alpha-L-fucosidase, but no induction in alpha-D-mannosidase, alpha-glucosidase and beta-glucuronidase. The latter two enzymes were unchanged during all cell culture phases. A drop in alpha-L-fucosidase and alpha-D-mannosidase activity ...

ABSTRACT A new procedure of assaying interferon (IF) has been developed. Cell suspension was dispensed into liquid scintillation counting vials together with IF sample. During an overnight incubation, the cells adhered sufficiently to the bottom of the vials and all the subsequent procedures were carried out without transfer of the cells from the vials. Vesicular stomatitis virus was inoculated and virus‐specific RNA was labeled by adding 3 H‐uridine and actinomycin D to the medium. Incubation was ...

Cysteamine at 0.5-5 mM effectively kills ${\rm HeLa} {\rm S}_{3}$ cells, while it is much less toxic in higher concentration (30 mM). The toxicity develops gradually with time as peroxide generates in the medium. Addition of catalase and peroxidase to the culture inhibits the toxic effect of cysteamine. The results suggest that the paradoxical cell-killing action of cysteamine can be ascribed to the generation of the peroxide by the compound ( $2{\rm RSH}+{\rm O}_{2}={\rm RSSR}+{\rm H}_{2}{\rm O}_{2}$). ...

The activity of RNase P (EC 3.1.26.5) from veal heart can be abolished by pretreatment of the enzyme preparation with micrococcal nuclease, pancreatic RNase A, or RNase T1. This indicates that veal heart RNase P contains an RNA component essential for function of the enzyme as has also been shown for E. coli RNase P (1–3). Additionally, veal heart RNase P has a buoyant density in Cs2SO4 of 1.33 g/cm3, which is intermediate between that of protein and nucleic acid.

Radical formation in D- and L-alanines was studied using ESR after internal3H-β-irradiation under the situation in which the contribution of Bremsstrahlung to form the radicals is assumed to be considerably less than the case of9 0Y-β-irradiation. It was demonstrated that the relative radical concentration by the β-rays was distinguishably higher in D-alanine than in L-alanine. Thus the asymmetry found in these experiments in the radical formation of the alanines may be attributed to the different ...

The nucleotide sequence of a 2.8-kilobase fragment of the Escherichia coli chromosome containing the umuDC genes has been determined. The DNA sequence specifies two open reading frames of 417 and 1266 nucleotides encoding proteins with calculated molecular weights of 15,063 and 47,677, respectively. From these and the previous results of genetic and biochemical studies on the cloned genes, we conclude that the former is the umuD and the latter is the umuC gene. In vitro transcription of the regulatory ...
