THE term "molecular disease" was coined almost 30 years ago by Linus Pauling and his associates to describe their analysis of the abnormality in sickle-cell anemia.1 Since that time tremendous progress has been made in the understanding of the structure and function of this mutant hemoglobin molecule and the conditions under which it aggregates when deoxygenated to form a gel. Indeed, in the last decade, particularly in the last few years, there has been an explosive growth in knowledge of the structure ...

Red Blood CellsContained within the normal erythrocyte is a 5 mM solution of hemoglobin, constituting 97.5 per cent of the cytoplasmic protein by weight.121 The other 2.5 per cent is primarily made up of protein systems that can be considered to support and regulate the function of hemoglobin as well as maintain the integrity of the cell. These proteins provide energy to maintain the ionic composition and water content of the cell, supply a reducing potential to keep hemoglobin in the ferrous form ...

Jurrien Dean, Alan N. Schechter,
An anti-hemoglobin antiserum obtained from a sheep immunized with human carboxyhemoglobin A0 demonstrated little difference in its reactivity with deoxy- or carboxyhemoglobin A0. However, a subpopulation of this antiserum isolated by synthetic peptide affinity chromatography clearly distinguished between these two hemoglobin species. This subpopulation, designated alpha(129-141) anti-hemoglobin antibodies, represents less than 1% of the total anti-hemoglobin antibodies. They are nonprecipitating ...

Abstract The human leukemia K562 cell line can be induced by 20 micro M hemin to reversibly accumulate embryonic and fetal hemoglobins without any change in the rate of cell division. When we reduced the rate of cell division by glutamine starvation or addition of hydroxyurea, the cells increased by tenfold the basal hemoglobin level of 0.3–0.5 pg Hb/cell. The combined effects of hemin and inhibitors of cell division permitted K562 cells to attain levels of hemoglobin (26–34 pg Hb/cell) close to ...

A male case of centronuclear myopathy is reported, with severe weakness at birth and death at 7 weeks. In all the muscles studied the fibres, despite their immature appearances, showed normal histochemical differentiation into type I and type II moieties. In contrast to the extrafusal fibres, the intrafusal fibres seemed to be normal in their development. Although the small centrally-nucleated muscle fibres were equipped with motor end-plates, the EMG revealed profuse fibrillation activity. The ...

A putative protective Plasmodium knowlesi antigen, recognized by two monoclonal antibodies which prevent invasion of erythrocytes by merozoites in vitro, is synthesised only by schizonts with 7 or more nuclei, during the last 1.5-2 h of the 24 h erythrocytic cycle of parasite development. The 66 000 MW antigen, which constitutes a minor parasite protein, is processed at the time of schizont rupture and merozoite release, to give rise to two smaller molecules of 44 000 and 42 000 MW. The 44 000 and ...

A cluster of conserved histidines and arginines (His-62, His-167, Arg-21, Arg-38, and Arg-168) in 3-phosphoglycerate kinase (PGK) has been implicated as possibly involved in the binding of 3-phosphoglycerate (3-PG) and/or stabilization of the negatively charged transition state. The role of these residues in the catalytic function of yeast PGK and in the substrate- and sulfate-dependent activation was investigated by site-directed mutagenesis. The following substitutions, R21A, R21Q, H62Q, H167S, and ...
