The purpose of this study was to determine the efficacy of novel recombinant adeno-associated viral (AAV) vector constructs in correcting metabolic defects in the liver in two strains of ornithine transcarbamylase (OTC)-deficient mice (spf and spf-ash). AAV vectors expressing mouse OTC were produced with capsids from AAV2 and the novel serotypes AAV7, 8, and 9. OTC-deficient mice were infused with these vectors as well as a control AAV2/8 vector expressing LacZ. In vivo activity of OTC was assessed ...

Unlike core histones, the linker histone H1 family is more evolutionarily diverse, and many organisms have multiple H1 variants or subtypes. In mammals, the H1 family includes seven somatic H1 variants; H1.1 to H1.5 are expressed in a replication-dependent manner, whereas H1.0 and H1X are replication-independent. Using ChIP-sequencing data and cell fractionation, we have compared the genomic distribution of H1.0 and H1X in human breast cancer cells, in which we previously observed differential distribution ...

Abstract The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal patient-specific treatments. Here we report AstraZeneca’s large drug combination dataset, consisting of 11,576 experiments from 910 combinations across 85 molecularly characterized cancer cell lines, and results of a DREAM Challenge to evaluate ...

Abstract Background Whether worldwide increases in life expectancy are accompanied by a better health status is still a debate. People age differently, and there is a need to disentangle whether healthy-ageing pathways can be shaped by cohort effects. This study aims to analyse trends in health status in two large nationally representative samples of older adults from England and the USA. Methods The sample comprised 55 684 participants from the first seven waves of the English Longitudinal Study ...

The hydrogenation of nitrous oxide by PNP pincer ruthenium complexes supposes a promising way to functionalize a hazardous gas and reduce the greenhouse effect, generating dinitrogen and water. Here, by DFT calculations we describe not only the whole mechanism for such a green transformation but we unravel the fundamental role of water, without which the reaction could not go forward. Water assists mandatorily in the H transfer to generate the hydroxyl group together with the release of dinitrogen.

From the analysis of the polarizability of carbon nano-onions (CNOs), it was concluded that CNOs behave as near perfect nanoscopic Faraday cages. If CNOs behave as ideal Faraday cages, the reactivity of the C240 cage should be the same in Li+ @C240 and Li+ @C60 @C240 . In this work, the Diels-Alder reaction of cyclopentadiene to the free C240 cage and the C60 @C240 CNO together with their Li+ -doped counterparts were analyzed using DFT. It was found that in all cases the preferred cycloaddition is on bond [ ...

Abstract Deep brain stimulation ( DBS ) is a new investigational therapy that has generated positive results in refractory depression. Although the neurochemical and behavioral effects of DBS have been examined, less attention has been paid to the influence of DBS on the network dynamics between different brain areas, which could contribute to its therapeutic effects. Herein, we set out to identify the effects of 1 h DBS in the infralimbic cortex ( IL ) on the oscillatory network dynamics between hippocampus ...

Recent results of Concepcion’s group (Chem. To analyze the differences between the reaction mechanisms of this WOC at different pHs, we performed ...

Acceptorless dehydrogenative coupling (ADC) of alcohols and amines using a manganese-based catalyst is able to produce aldimines, which have a wide reactivity, without underestimating the generation of a clean fuel, since as a result of this coupling, molecular hydrogen is also obtained. Therefore, the aldimine synthesis represents an interesting reaction from chemical and clean energy points of view. In this work, the computational study, via density functional theory (DFT) calculations, of a manganese- ...

A recent study showed that a Mn-pincer could catalyze the acceptorless dehydrogenative coupling of nitriles and alcohols to yield acrylonitriles. The reaction mechanism proposed in that work contained some intermediates that, in most of the cases, were not characterized. Moreover, one of the intermediates involved a charged separation, which is unlikely in apolar solvents. To clarify the reaction mechanism of this critical reaction, we decided to perform a DFT study. Our results prove the existence ...

We study with density functional theory calculations the mechanism of the original example of a base-metal-catalyzed synthesis of amides from alcohols and amines. A preliminary proposal of the mechanism of this reaction was experimentally reported by Milstein and co-workers. Our aims were to develop a Spanish version of the short portable mental status questionnaire (SPMSQ) (Pfeiffer's test) and to validate the resultant Spanish version of the test among a general population aged 65 years or older.First, we developed the Spanish version of the SPMSQ by means of a cross-cultural adaptation methodology. Patients were the assessed by independent teams in two sessions the same day. The first team (two trained psychiatrists and one trained family physician) carried out a standardised ...

Charisma is a term commonly used in conservation biology to describe species. However, as the term "charismatic species" has never been properly defined, it needs to be better characterized to fully meet its potential in conservation biology. To provide a more complete depiction, we collected information from four different sources to define the species currently considered to be the most charismatic and to understand what they represent to the Western public. First, we asked respondents of two separate ...

Density functional theory (DFT) calculations have been used to investigate the behavior of different transition metals from Groups 8 (Fe and Ru) and 9 (Co and Rh) in an already well-known catalytic mechanism, which is based on an Ru(SIMes)(PPh3)Cl2=CH(Ph) complex. As expected, Ru has proven to perform better than their Fe, Co, and Rh counterparts. Even though the topographic steric maps analysis shows no difference in sterical hindrance for any of the metal centers, geometrically, the Fe-based species ...
