Clostridium (Clostridioides) difficile is the main cause of nosocomial diarrhoea. Ribotype 018 (RT018) has been recognized as the predominant strain responsible for C. difficile infection (CDI) in Italy, whereas in most other European countries only sporadic RT018 cases occur. Between August and October 2015, a suspected C. difficile outbreak at two associated hospitals in Southern Germany was investigated by comprehensive molecular typing. Surprisingly, RT018 was detected in 9/82 CDI patients, which ...

Even after more than three decades of intensive research in the field of contact mechanics, there is still a great need for improving the numerical methods. Recent investigations on the 2D Hertzian contact problem clearly showed significantly varying results for different finite element versions. In this paper we will compare the analytical solution of the 2D Hertzian contact problem with the numerical results of the classical h-version with uniform and locally refined meshes, as well as with the ...

In this article the p-version finite element method is applied to thin-walled structures. Two different hierarchic element formulations are compared, a shell approach as well as a shell-like, solid formulation. Both approaches are compared for linear elastic and elastoplastic problems. Special emphasis is placed on the efficiency as well as on determining the area of application for both formulations.

Cellular contractility and, thus, the ability to alter cell shape are prerequisites for a number of important biological processes such as cytokinesis, movement, differentiation, and substrate adherence. The contractile capacity of vascular smooth muscle cells (VSMCs) is pivotal for the regulation of vascular tone and thus blood pressure and flow. Here, we report that conditional ablation of the transcriptional regulator Junb results in impaired arterial contractility in vivo and in vitro. This was ...

The emergence of methicillin-resistant Staphylococcus aureus (MRSA) in animals such as horses, pet animals and productive livestock has raised questions of a probable human origin and in more general of host specificity of S. aureus. Particular clonal lineages are obviously specific for humans (e.g. ST15, ST25, ST45) and other for ruminants (e.g. ST151). MRSA associated with veterinary nosocomial infections (e.g. ST8 and ST254 in horses, ST22 in small animals) very likely have their origin in health ...

ESBL-/AmpC-producing Escherichia coli from organic fertilizers were previously detected on soil surfaces of arable land and might be emitted by wind erosion. To investigate this potential environmental transmission path, we exposed ESBL-/AmpC-positive chicken litter, incorporated in agricultural soils, to different wind velocities in a wind tunnel and took air samples for microbiological analysis. No data exist concerning the airborne tenacity of ESBL-/AmpC-producing E. coli. Therefore, we explored ...

Background Since 2013, over 100 cases of Mycobacterium chimaera prosthetic valve endocarditis and disseminated disease were notified in Europe and the USA, linked to contaminated heater–cooler units (HCUs) used during cardiac surgery. We did a molecular epidemiological investigation to establish the source of these patients' disease. Methods We included 24 M chimaera isolates from 21 cardiac surgery-related patients in Switzerland, Germany, the Netherlands, and the UK, 218 M chimaera isolates from ...

Specific climate adaptation and resilience measures can be efficiently designed and implemented at regional and local levels. Climate and environmental databases are critical for achieving the sustainable development goals (SDGs) and for efficiently planning and implementing appropriate adaptation measures. Available federated and distributed databases can serve as necessary starting points for municipalities to identify needs, prioritize resources, and allocate investments, taking into account often ...

During a field experiment applying broiler manure for fertilization of agricultural land, we detected viable Clostridioides (also known as Clostridium) difficile in broiler faeces, manure, dust and fertilized soil. A large diversity of toxigenic C. difficile isolates was recovered, including PCR ribotypes common from human disease. Genomic relatedness of C. difficile isolates from dust and from soil, recovered more than 2 years after fertilization, traced their origins to the specific chicken farm ...

Clostridium (Clostridioides) difficile is a major cause of nosocomial diarrhoea. A first inter-laboratory ring trial was performed in four European countries to evaluate the genotyping and antibiotic susceptibility testing (AST) accuracy. Six C. difficile isolates representing the epidemiologic important ribotypes (RT), RT001, RT002, RT010, RT014, RT027, and RT078 were blinded and send to 21 participating laboratories. Participants tested the samples with their genotyping and AST methods in use for ...

The Wnt-pathway, with nuclear beta-catenin as a main effector protein, can induce Epithelial-Mesenchymal Transition (EMT) and indirectly chromosomal instability (CIN). Here, we investigated for disseminated tumor cells (DTC) expressing nuclear beta-catenin and if this phenotype is associated with specific genomic alterations. Therefore, we established a double-immunfluorescence labelling protocol to determine the staining pattern of beta-catenin in single disseminated tumor cells. Single CK-positive ...

Myxococcota represents a small bacterial phylum comprising seven families, which have been shown to be exceptionally rich producers of pharmaceutically interesting natural products. Because of the challenging microbiology of most Myxococcota, their cultured biodiversity is scarcely explored. This study introduces a divergent myxobacterial family, called Pendulisporaceae, with distinct sporulation behavior. Strikingly, 90% of their biosynthetic gene clusters have not been assigned to their respective ...

Clostridioides difficile is the primary infectious cause of antibiotic-associated diarrhea. Local transmissions and international outbreaks of this pathogen have been previously elucidated by bacterial whole-genome sequencing, but comparative genomic analyses at the global scale were hampered by the lack of specific bioinformatic tools. Here we introduce a publicly accessible database within EnteroBase ( http://enterobase.warwick.ac.uk ) that automatically retrieves and assembles C. difficile short- ...

Enterococcus faecalis is a multifaceted microorganism known to act as a beneficial intestinal commensal bacterium. It is also a dreaded nosocomial pathogen causing life-threatening infections in hospitalised patients. Isolates of a distinct MLST type ST40 represent the most frequent strain type of this species, distributed worldwide and originating from various sources (animal, human, environmental) and different conditions (colonisation/infection). Since enterococci are known to be highly recombinogenic ...
