We report four patients (mean age 65 yr; range 40-77 yr) affected by acquired pure red cell aplasia (PRCA) complicating chronic lymphoid disorders and treated with anti-CD20 monoclonal antibody rituximab. Three out of four patients were given packed red cell transfusion. Steroids and recombinant erythropoietin (r-Epo) were also administered as first-line therapy without response. After a mean time of 57 d (range 23-62 d) from PRCA diagnosis, all patients received rituximab at a dosage of 375 mg/m(2)/ ...

This study analyses the evolution of the bibliometric indicators of productivity and repercussion of European Union (EU) research into the respiratory system during the period from 1987–1998, describing the geographical distribution. Using MedLine, a selection was made of those articles by EU authors published between 1987–1998 in 38 respiratory system journals (classification from the Institute for Scientific Information). The journals, country of origin, number of articles and the relation to socioeconomic ...

B-Myb is a transcription factor belonging to the myb family, whose activity has been associated with augmented DNA synthesis and cell cycle progression. We showed recently that B-Myb autoregulates its own expression through promoter transactivation. We report in this study that CDK9, the cyclin T associated kinase, which phosphorylates and activates RNA-Polymerase II, suppresses B-Myb autoregulation through direct interaction with the carboxyl-terminus of the B-Myb protein. Down-regulation of the ...

Catecholamine release from perfused cat adrenal glands has been continuously monitored by on-line connection of the perfusion fluid emanating from the gland to an electrochemical detector. This method allowed: the recording of basal levels and fluctuations of catecholamine release with good reliability; the study of the release of catecholamines in response to even submicromolar concentrations of nicotine or acetylcholine and 3 mM increments of the K+ concentration, perfused in pulses of a few seconds, ...

The peroxidase-catalyzed nitration of tyrosine derivatives by nitrite and hydrogen peroxide has been studied in detail using the enzymes lactoperoxidase (LPO) from bovine milk and horseradish peroxidase (HRP). The results indicate the existence of two competing pathways, in which the nitrating species is either nitrogen dioxide or peroxynitrite. The first pathway involves one-electron oxidation of nitrite by the classical peroxidase intermediates compound I and compound II, whereas in the second pathway ...

A methodology to develop a GIS-based system for the surface water risk assessment of agricultural chemicals is described. It is based on the integration of relational and spatial databases, GIS incorporating raster and vector, mass balance models, and pesticide risks indicators. Surface water pollution was modeled by taking into account two main processes: the load due to drift and the load due to a rainfall-runoff event. The former is immediately consequent to pesticide application; the second ...

The cyclopentenone 15-deoxy-Δ 12,14 -prostaglandin J 2 (15d-PGJ 2 ) induces cell proliferation and mitogen-activated protein kinase activation. Here, we describe that these effects are mediated by 15d-PGJ 2 -elicited H-Ras activation. We demonstrate that this pathway is specific for H-Ras through the formation of a covalent adduct of 15d-PGJ 2 with Cys-184 of H-Ras, but not with N-Ras or K-Ras. Mutation of C184 inhibited H-Ras modification and activation by 15d-PGJ 2 , whereas serum-elicited stimulation ...

The contribution of genetic variation in DNA repair genes to gastric cancer (GC) risk remains essentially unknown. The aim of this study was to explore the relative contribution of DNA repair gene polymorphisms to GC risk and severe chronic atrophic gastritis (SCAG). Method A nested case control study within the EPIC cohort was performed including 246 gastric adenocarcinomas and 1175 matched controls. Adult mammalian cells can be reprogrammed to a pluripotent state by forcing the expression of a few embryonic transcription factors. The resulting induced pluripotent stem (iPS) cells can differentiate into cells of all three germ layers. It is well known that post-natal cardiomyocytes (CMs) lack the capacity to proliferate. Here, we report that neonatal CMs can be reprogrammed to generate iPS cells that express embryonic-specific markers and feature gene-expression profiles similar to those of mouse ...

It was recently reported that B-MYB can overcome p107-induced growth arrest. Here we show that B-MYB autoregulation of its own transcription is specifically suppressed by p107 and transient transfection assays with p107 deletion constructs determined that the carboxyl terminus of the protein, containing the major pocket region, was associated with inhibition of B-MYB-dependent transactivation. Consistent with these results, co-immunoprecipitation studies showed that p107 interacted in vivo with ...

Abstract Chromaffin cells from the bovine adrenal medulla express α‐bungarotoxin‐sensitive acetylcholine receptors whose subunit composition is unknown. Northern blot analysis showed that the α7 subunit, a main component of these α‐bungarotoxin‐sensitive acetylcholine receptors in avian and rat brain, is expressed in chromaffin cells. The cDNA of this bovine α7 subunit was cloned by polymerase chain reaction amplification of adrenal medulla RNA for detailed characterization of structure and function. ...

In coarse-grained, unsaturated soils, heavy rainstorms may cause either shallow landslides or superficial soil erosion. The triggering of both of these phenomena is related to infiltration, runoff and overland flow, which are key processes requiring investigation. In particular, the quantitative estimates of the sediment yield at the outlet of mountain basins requires suitable physically based modelling. In this paper, the available approaches to soil erosion analysis are first reviewed, and the ...

Diabetes mellitus--whether driven by insulin deficiency or insulin resistance--causes major alterations in muscle metabolism. These alterations have an impact on nutrient handling, including the metabolism of glucose, lipids, and amino acids, and also on muscle mass and strength. However, the ways in which the distinct forms of diabetes affect muscle mass differ greatly. The most common forms of diabetes mellitus are type 1 and type 2. Thus, whereas type 1 diabetic subjects without insulin treatment ...

Aging is a complex phenomenon driven by a variety of molecular alterations. A relevant feature of aging is chronic low‐grade inflammation, termed “inflammaging.” In type 2 diabetes mellitus (T2DM), many elements of aging appear earlier or are overrepresented, including consistent inflammaging. T2DM patients have an increased death rate, associated with an incremented inflammatory score. The source of this inflammation is debated. Recently, the senescence‐associated secretory phenotype (SASP) has been ...

Abstract The optimal recognition of penicillin determinants, including amoxicillin (AX), by specific IgE antibodies is widely believed to require covalent binding to a carrier molecule. The nature of the carrier and its contribution to the antigenic determinant is not well known. Here we aimed to evaluate the specific-IgE recognition of different AX-derived structures. We studied patients with immediate hypersensitivity reactions to AX, classified as selective or cross-reactors to penicillins. Competitive ...

Recent clinical trials have demonstrated a strong cardiovascular (CV) protective effect of sodium/glucose cotransporter (SGLT) 2 inhibitors, a recently introduced class of hypoglycaemic agents. The improvement in glycated haemoglobin and other conventional risk factors explains only a portion of the observed reduction in CV risk. A relevant feature of SGLT2‐inhibitor‐treated diabetic patients is the increase in circulating levels of ketone bodies, which has been proposed to mediate part of the beneficial ...
