The uropathogenic Escherichia coli strain 536 (O6:K15: H31) carries two large chromosomal pathogenicity islands (Pais). Both Pais are flanked by tRNA genes. Spontaneous deletion of Pai II results in truncation of the leuX tRNA Leu 5 gene. This tRNA is required for the expression of type 1 fimbriae (Fim) and other virulence factors. Transcription of fimA , encoding the major type 1 fimbrial subunit is controlled by an invertable DNA switch. The inversion is catalysed by two recombinases, FimB and FimE. ...

We have measured the equilibrium constant for the denaturation transition of the engineered fluorescein-binding lipocalin FluA as a function of pressure and temperature, taking advantage of the fact that the ligand's fluorescence is almost fully quenched when complexed with the folded protein, but reversibly reappears on denaturation. From the equilibrium constant as a function of pressure and temperature all of the involved thermodynamic parameters of protein folding, in particular the changes ...

The gene locus nuo of the proton-translocating NADH : ubiquinone oxidoreductase in Escherichia coli was identified by means of a DNA probe made by the polymerase chain reaction. The primers used for the reaction were derived from consensus sequences of the NAD(H)-binding subunit of mitochondrial NADH : ubiquinone oxidoreductase and the NAD+-reducing hydrogenase of Alcaligenes eutrophus. The nuo locus lies between minutes 49·2 and 49·6 on the E. coli chromosome and contains a cluster of 14 genes. They ...

Multiple reaction monitoring (MRM)-based mass spectrometric quantification of peptides and their corresponding proteins has been successfully applied for biomarker validation in serum. The option of multiplexing offers the chance to analyze various proteins in parallel, which is especially important in obesity research. Here, biomarkers that reflect multiple comorbidities and allow monitoring of therapy outcomes are required. Besides the suitability of established MRM assays for serum protein quantification, ...

A pig-skin preparation enriched in epidermal G1-chalone when administered to cells of the rat tongue epithelial line RTE2 at concentrations of 3–300 μg/ml (dry mass) caused a 60% reduction in cell number. Three other cell lines showed essentially no growth inhibition during chalone treatment. The kinetics of chalone inhibition were similar to those observed in mouse epidermis in vivo. Five hours after the addition of chalone preparation in fresh medium a decrease in the rate of DNA synthesis was ...

To date, there is a lack of satisfactory inferential techniques for the analysis of multivariate data in factorial designs, when only minimal assumptions on the data can be made. Presently available methods are limited to very particular study designs or assume either multivariate normality or equal covariance matrices across groups, or they do not allow for an assessment of the interaction effects across within-subjects and between-subjects variables. We propose and methodologically validate a ...

Host protection upon vaccination usually results from the complex interplay of humoral and cellular components of the immune system. Exploring hepatitis B surface antigen (HBsAg)-specific T cell responses and their correlation with humoral responses under immunosuppression, we analyzed 51 renal transplant recipients, differing in HBV vaccine-specific antibody titers (non [NRs]-, low [LRs]-, and high responders [HRs]) and in 22 healthy controls (HCs) in a cross-sectional study. HBsAg-specific T cells were ...

Oral streptococci are a heterogeneous group of human commensals, with a potential to cause serious infections. Activation of plasminogen has been shown to increase the virulence of typical human pathogenic streptococci such as S. pneumoniae. One important factor for plasminogen activation is the streptococcal α-enolase. Here we report that plasminogen activation is also common in oral streptococci species involved in clinical infection and that it depends on the action of human plasminogen activators. ...

ABSTRACT Viridans streptococci (VS) are responsible for several systemic diseases, such as endocarditis, abscesses, and septicemia. Unfortunately, species identification by conventional methods seems to be more difficult than species identification of other groups of bacteria. The aim of the present study was to evaluate the use of cell matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-MS) for the rapid identification of 10 different species of VS. A total of ...

Of cryogenic liquid hydrogen tanks for future airliners, their volumetric and gravimetric efficiencies, their robustness and their environmental adaptability are all strengthened via a novel thermal insulation concept proposed in this work. A conventional cryogenic tank is insulated either purely by a layer/layers of Polyurethane (PU) foam or by a vacuum-based multilayer insulation (MLI). In the new concept, an extra layer is inserted into the PU foam. The intermediate layer can be filled with liquid ...

Underwater acoustic networks (UANs) are an emerging technology for a number of oceanic applications, ranging from oceanographic data collection to surveillance applications. However, their reliable usage in the field is still an open research problem, due to the challenges posed by the oceanic environment. The UAN project, a European-Union-funded initiative, moved along these lines, and it was one of the first cases of successful deployment of a mobile underwater sensor network integrated within ...

Mapping of exact tree positions can be regarded as a crucial task of field work associated with forest monitoring, especially on intensive research plots. We propose a two-stage density clustering approach for the automatic mapping of tree positions, and an algorithm for automatic tree diameter estimates based on terrestrial laser-scanning (TLS) point cloud data sampled under limited sighting conditions. We show that our novel approach is able to detect tree positions in a mixed and vertically structured ...

Most animal tissues and organ systems are comprised of highly ordered arrays of varying cell types. The development of external sensory organs requires complex cell-cell communication in order to give each cell a specific identity and to ensure a regular distributed pattern of the sensory bristles. This involves both long and short range signaling mediated by either diffusible or cell anchored factors. In a variety of processes the heterophilic Irre Cell Recognition Module, consisting of the Neph- ...

ABSTRACT Wildfires in the sub‐alpine belt of the Austrian Limestone Alps sometimes cause severe vegetation and soil destruction with increased danger of secondary natural hazards such as avalanches and debris flows. Some of the affected areas remain degraded to rocky slopes even decades after the fire, raising the question as to whether the ecosystems will ever be able to recover. The mean fire interval, the duration of recovery and the role of geomorphic processes for vegetation regeneration are ...

Background. Polyomavirus BK virus (BKV) infection represents a serious complication leading to BKV-associated nephropathy (BKVAN) and subsequent kidney graft loss in up to 10% of transplant patients. Cellular immunity is known to play a crucial role in the control of BKV replication. However, the knowledge on the BKV-T-cell response is limited: only two (VP1 and large T antigen) of six known BKV proteins were evaluated for their antigenicity so far. Methods. By using 10-color flow cytometry and newly ...

We investigated the thickness and composition of five small alpine mires in Tyrol (Austria), performing a combination of conventional techniques (telescope rods, drilling) and geophysical methods. GPR surveys were carried out using 100 and 200 MHz antennas. Geoelectrical profiles supplemented the GPR at two sites; the 2D-surveys were enhanced by additional 1D-profiling. The aim was to achieve data on mire thickness, to cross-check the methods and to find reasons for different performance at the five ...

Mammalian thioredoxin reductase (TrxR) catalyzes reduction of thioredoxin and many other substrates, and is a central enzyme for cell proliferation and thiol redox control. The enzyme is a selenoprotein and can therefore, like all other mammalian selenoproteins, not be directly expressed in Escherichia coli, since selenocysteine-containing proteins are synthesized by a highly species-specific translation machinery. This machinery involves a secondary structure, SECIS element, in the selenoprotein- ...

Employing an agent–principal perspective on EU conflict resolution and focusing on the politics of discretion, we argue that the context in which the agent acts affects the principal–agent relation. Our conceptualization draws on the agent's actions as a source of discretion, which can be either an unintentional product of the characteristics of the external environment in which the agent performs (structure-induced discretion) or a product of intentionally pursued agent action (interest-induced discretion). ...

: One of the most challenging problems in developmental neurosciences is to understand the establishment and maintenance of specific membrane contacts between axonal, dendritic, and glial processes in the neuropils, which eventually secure neuronal connectivity. However, underlying cell recognition events are pivotal in other tissues as well. Recruitment patterns of Baltic Sea sprat (Sprattus sprattus) were correlated to time series of (i) month- and depth-specific temperature conditions and (ii) larval drift patterns inferred from long-term Lagrangian particle simulations. From the latter, we derived an index that likely reflected the variable degree of annual larval transport from the central, deep spawning basins to the shallow coastal areas of the Baltic Sea. The drift index was significantly (P < 0.001) correlated to sprat recruitment ...
