カブトガニ血球抽出物(LAL)を用いたリムルステストにおいて,大過剰のカルボキシメチル化カードラン(CM-カードラン)を反応系に共存させることによつてβ-グルカン自身によるLALの活性化を阻害し,エンドトキシンを特異的に検出する方法を開発した。LALの反応液に0.1mg/ml以上のCM-カードランを添加した場合,CM-カードラン自身によるLALの活性化は完全に阻害されたが,エンドトキシンの測定は影響を受けなかつた。1mg/mlのCM-カードランを含むLALの測定系を構成したところ,一般に用いられているリムルステストの手法であるゲル化転倒法,比濁時間分析法,合成基質法のすべてでβ-グルカンの影響を受けないエンドトキシンの測定が可能であつた。また,この測定系を用いてグラム陰性菌を特異的に検出できる可能性も示唆された。この方法は,試薬の調製が非常に簡便であり,従来通りの操作法でエンドトキシンに特異的なリムルステストが行えることから,薬剤のエンドトキシン試験のみならず,臨床分野でも有用と考える。
