The cAMP pathway activates p38-MAPKs in the FRTL-5 rat thyroid cell line, contributing to the increased expression of the Na+/I- symporter (NIS) mRNA. This study investigates the cAMP-dependent expression and transcriptional activity of the p38-MAPK substrate CCAAT/enhancer-binding protein-homologous protein (CHOP). CHOP is expressed in the rat thyroid gland and in confluent PCCL3 and FRTL-5 cells. In FRTL-5 cells, TSH withdrawal induced a rapid down-regulation of CHOP that could be prevented by forskolin ( ...

The spectrum of the clinical presentation and severity of malaria infections is broad, ranging from uncomplicated febrile illness to severe forms of disease such as cerebral malaria (CM), acute lung injury (ALI), acute respiratory distress syndrome (ARDS), pregnancy-associated malaria (PAM) or severe anemia (SA). Rodent models that mimic human CM, PAM and SA syndromes have been established. Here, we show that DBA/2 mice infected with P. berghei ANKA constitute a new model for malaria-associated ALI. ...

Abstract Background Infection with Plasmodium is the cause of malaria, a disease characterized by a high inflammatory response in the blood. Dendritic cells (DC) participate in both adaptive and innate immune responses, influencing the generation of inflammatory responses. DC can be activated through different receptors, which recognize specific molecules in microbes and induce the maturation of DC. Methods Using Plasmodium yoelii , a rodent malaria model, the effect of Plasmodium -infected erythrocytes ...

Dendritic cells (DCs) have been proposed as mediators of immunity against malaria parasites, as well as a target for inhibition of cellular responses. Here we describe the transcriptomic analysis of spleen DCs in response to Plasmodium infection in a rodent model. We identified a high number of unique transcripts modulated in DCs upon infection. Many cellular functions suffer extensive genomic regulation including the cell cycle, the glycolysis and purine metabolism pathways and also defence responses. ...

Hemozoin (Hz) is released into the blood stream after rupture of infected red blood cells (iRBCs) at the end of each parasite replication cycle. This free Hz is ingested by circulating and resident phagocytes. The presence of Hz in tissues after clearance of infection has been previously reported. Still, little is known about the kinetics of Hz in vivo , during and after Plasmodium infection. It is particularly important to understand Hz kinetics after malaria infections as it has been reported that ...

Antibodies that neutralize infectivity of malaria sporozoites target the central repeat region of the circumsporozoite (CS) protein, which in Plasmodium falciparum is comprised primarily of 30-40 tandem NANP tetramer repeats. We evaluated immunogenicity of an alum-adsorbed (NANP)6 peptide conjugated to an outer membrane protein complex (OMPC) derived from Neisseria meningitidis, a carrier protein used in a licensed H. influenzae pediatric vaccine. Mice immunized with alum-adsorbed (NANP)6-OMPC, with ...

Immunization with Plasmodium sporozoites can elicit high levels of sterile immunity, and neutralizing antibodies from protected hosts are known to target the repeat region of the circumsporozoite (CS) protein on the parasite surface. CS-based subunit vaccines have been hampered by suboptimal immunogenicity and the requirement for strong adjuvants to elicit effective humoral immunity. Pathogen-associated molecular patterns (PAMPs) that signal through Toll-like receptors (TLRs) can function as potent ...

Intranasal (IN) immunization with a Plasmodium circumsporozoite (CS) protein conjugated to flagellin, a Toll-like receptor 5 agonist, was found to elicit antibody-mediated protective immunity in our previous murine studies. To better understand IN-elicited immune responses, we examined the nasopharynx-associated lymphoid tissue (NALT) in immunized mice and the interaction of flagellin-modified CS with murine dendritic cells (DCs) in vitro. NALT of immunized mice contained a predominance of germinal ...

After mosquito bite, the malaria parasite first infects the liver, where it is thought to be undetected by the host immune system as it develops into the blood-stage pathogen. Maria Mota and her colleagues now report that Plasmodium RNA is detected by hepatocytes, triggering an interferon response that controls the parasite burden in the liver and blood of infected mice. Before they infect red blood cells and cause malaria, Plasmodium parasites undergo an obligate and clinically silent expansion ...

Dryland pastures provide the basis for animal sustenance in extensive production systems in Iberian Peninsula. These systems have temporal and spatial variability of pasture quality resulting from the diversity of soil fertility and pasture floristic composition, the interaction with trees, animal grazing, and a Mediterranean climate characterized by accentuated seasonality and interannual irregularity. Grazing management decisions are dependent on assessing pasture availability and quality. Conventional ...

Dendritic cells (DC) and macrophages phagocytose pathogens and degrade them in their phagosomes to allow for proper presentation of foreign antigens to other cells of the immune system. The Plasmodium parasite, causative agent of malaria, infects RBC that are phagocytosed by DC and macrophages during the course of infection. Under specific conditions, the functionality of these cells can be affected by phagocytosis of Plasmodium-infected RBC. We investigated whether phagosomal maturation and degradation ...

To study this problem, we use the one dimensional (1D) nine-directors Cosserat theory approach which reduces the exact three-dimensional equations to a system depending only on time and on a single spatial variable. A one-dimensional model for the flow of a viscous fluid with axisymmetric swirling motion is derived in the particular case of a straight tube of variable circular cross-section. The model is obtained by integrating the Navier–Stokes equations over cross section the tube, taking a velocity field approximation provided by the Cosserat theory. This procedure yields a one-dimensional system, depending only on time and a single spatial variable. The velocity field approximation satisfies exactly both ...

This one-dimensional system is obtained by integrating the linear momentum equation over the cross-section of the tube, taking a velocity field approximation provided ...

In this work, we present sufficient conditions in order to establish different types of Ulam stabilities for a class of higher order integro-differential equations. In particular, we consider a new kind of stability, the σ-semi-Hyers-Ulam stability, which is in some sense between the Hyers–Ulam and the Hyers–Ulam–Rassias stabilities. These new sufficient conditions result from the application of the Banach Fixed Point Theorem, and by applying a specific generalization of the Bielecki metric.

A finite strain gradient model for the 3D analysis of materials containing spherical voids is presented. A two-scale approach is proposed: a least-squares methodology for RVE analysis with quadratic displacements and a full high-order continuum with both fourth-order and sixth-order elasticity tensors. A meshless method is adopted using radial basis function interpolation with polynomial enrichment. Both the first and second derivatives of the resulting shape functions are described in detail. The aim of this paper, is to analyze the axisymmetric unsteady flow of an incompressible second order fluid in a slender body of revolution with circular cross section with variable radius. From this 1D system, we obtain for a flow in a rigid and impermeable tube, without swirling ...
