Over the last two decades, N-acylhydrazone (NAH) has been proven to be a very versatile and promising motif in drug design and medicinal chemistry. Herein, we discuss the current and future challenges in the emergence of bioactive NAH-based scaffolds and to developing strategies to overcome the failures in drug discovery. The NAH-related approved drugs nitrofurazone, nitrofurantoin, carbazochrome, testosterone 17-enanthate 3-benzilic acid hydrazine, nifuroxazide, dantrolene, and azumolene are already ...

Several studies have established an association between diabetes and alterations in bone metabolism; however, the underlying mechanism is not well established. Although zinc is recognized as a potential preventive agent against diabetes-induced bone loss, there is no evidence demonstrating its effect in chronic diabetic conditions. This study evaluated the effects of zinc supplementation in a chronic (90 days) type 1 diabetes-induced bone-loss model. Male Wistar rats were distributed in three groups: ...

A DRUG: THALIDOMIDE.Thalidomide, first synthesized in 1953, was widely prescribed for morning sickness of pregnant women from 1957 to 1961, when it was found to be seriously teratogenic, having caused serious birth defect.Nowadays, a quarter of a century later, it appears that it may be a miracle drug for such diseases as leprosy, AIDS, cancer and tuberculosis.

In this work, we present the results of two combined approaches, molecular docking and comparative molecular field analysis (CoMFA), to propose how the selective cyclooxygenase-2 inhibitor celecoxib could act as a p38 mitogen-activated protein (MAP) kinase inhibitor. The docking analysis revealed why celecoxib has a less favorable binding energy (ΔG = −12.4 kcal/mol) than the selective p38 MAP kinase (p38 MAPK) inhibitor, SB203580 (ΔG = −22.2 kcal/mol). The CoMFA results revealed unfavorable steric effects ...

This paper describes molecular docking studies of a series of classical NSAIDs with PPARgamma receptor, which has been pointed as a new target for the design of anti-cancer and anti-inflammatory drugs, and has been found to be responsible for some of the already established pharmacological effects observed for marketed drugs. The results show the molecular basis of PPARgamma activation by non-selective COX inhibitors.

This study aimed to synthesize and characterize nanoparticles (NPs) of poly(methyl methacrylate) (PMMA) and evaluate their ability to incorporate plant extracts with antitumor activity and low dissolution in aqueous media. The extract used was n-hexane partition of the methanol extract of Piper cabralanum (PCA-HEX). PMMA NPs were obtained using the mini-emulsion method, which was able to encapsulate almost 100% of PCA-HEX. The synthesized polymeric particles presented with a size of 200 nm and a negative ...

A new method for the determination of proton-transfer rate constants in the ground state of anthocyanins and related flavylium salts is described. The method is based on the well-known pKa shift of phenols on going from ground to the first excited singlet state coupled to the typically very fast excited state proton transfer and very short lifetimes (picosecond range) occurring in these compounds. Under these conditions, a nanosecond light pulse instantaneously shifts the ground-state equilibrium, ...

The effects of anionic, cationic, and nonionic micelles on the dynamics of proton transfer in the ground and excited state of anthocyanins have been examined employing the 4-methyl-7-hydroxyflavylium ion (HMF) as a model compound. Unlike other anthocyanins and anthocyanin analogues, HMF does not hydrate in water, allowing study of the acid−base (AH+/A) equilibrium in the absence of competing hydration or tautomerism reactions. Anionic sodium dodecyl sulfate (SDS) micelles are found to stabilize the ...

In particular, it was shown that the behavior of the equilibrium binding ...

Murine and bovine embryos were sexed by using polyclonal and monoclonal anti-H-Y antibodies in cytotoxic and immunofluorescence assays. The specificity of 3 different sources of antibodies are compared. Sexing was accomplished by complement-mediated cytotoxicity or by H-Y antigen detection using a anti-mouse Ig fluorescein conjugate. Cytogenetic ascertainment for correct sex identification yielded a score of about 85% in both species, while the immunofluorescence assay yielded a score of 83%.
