Purpose.: Familial exudative vitreoretinopathy (FEVR) is an inherited disorder that disrupts the development of the retinal vasculature and can result in blindness. FEVR is genetically heterogeneous and mutations in four genes, NDP, FZD4, LRP5, and TSPAN12, encoding components of a novel ligand-receptor complex that activates the Norrin-β-catenin signaling pathway, account for approximately 50% of cases. Catalytic reduction of 5-nitroimidazoles 4 in dioxane solution gives 5-aminoimidazoles 2 in good yield. The derivatives 2d–f were isolated as stable, crystalline compounds which undergo slow decomposition on exposure to air. In a similar manner, solutions of 4-aminoimidazoles 1 were generated from the corresponding 4-nitroimidazoles 3 but attempts to isolate the amines were unsuccessful. Reaction of the novel derivatives (2) with amidines gives 5-(2-quinolyl)-1,2,4-thiadiazoles (8) and reaction with anionic nucleophiles results in substitution at sulphur.

Cytidine deamination that is guided by clustered regularly interspaced short palindromic repeats (CRISPR) can mediate a highly precise conversion of one nucleotide into another — specifically, cytosine to thymine — without generating breaks in DNA. Thus, genes can be base-edited and rendered inactive without inducing translocations and other chromosomal aberrations. The use of this technique in patients with relapsed childhood T-cell leukemia is being investigated.
