The mechanism of ATP modulation of E2P dephosphorylation of sarcoplasmic reticulum Ca2+-ATPase wild type and mutant forms was examined in nucleotide binding studies of states analogous to the various intermediates of the dephosphorylation reaction, obtained by binding of metal fluorides, vanadate, or thapsigargin. Wild type Ca2+-ATPase displays an ATP affinity of 4 μm for the E2P ground state analog, 1 μm for the E2P transition state and product state analogs, and 11 μm for the E2 dephosphoenzyme. ...

The nucleotide binding properties of mutants with alterations to Asp351 and four of the other residues in the conserved phosphorylation loop,351DKTGTLT357, of sarcoplasmic reticulum Ca2+-ATPase were investigated using an assay based on the 2′,3′-O-(2,4,6-trinitrophenyl)-8-azidoadenosine triphosphate (TNP-8N3-ATP) photolabeling of Lys492 and competition with ATP. Nine patients with recurrent or refractory epithelial ovarian carcinoma following previous chemotherapy were treated with high-dose carboplatin (300 mg/m2) and ifosfamide according to a dose escalation schedule (1.50, 1.75, 2.00 g/m2), each given intravenously daily for 5 days with autologous bone marrow support. Abstract 2',3'-O-(2,4,6-trinitrophenyl)-8-azido-AMP (TNP-8N3-AMP) and -ATP photolabel Lys-492 at the active site of the Ca(2+)-ATPase of sarcoplasmic reticulum (McIntosh, D. B., Woolley, D. G., and Berman, M. C. (1992) J. Biol. Chem. 267, 5301-5309).
