Neonates have been shown to have a heightened sensitivity to the central depressive effects of opioids compared to older infants and adults. The limited development of P-glycoprotein (P-gp) may limit the ability of the neonate to efflux morphine from the brain back to the systemic circulation. The objective of the study was to determine the ontogeny of P-gp in the human brain. Postmortem cortex samples from gestational age (GA) 20–26 wk, GA 36–40 wk, postnatal age (PNA) 0–3 mo, PNA 3–6 mo, and adults ...

Abstract The second phase of the North American Monsoon Experiment (NAME) Model Assessment Project (NAMAP2) was carried out to provide a coordinated set of simulations from global and regional models of the 2004 warm season across the North American monsoon domain. This project follows an earlier assessment, called NAMAP, that preceded the 2004 field season of the North American Monsoon Experiment. Abstract As initial human gene therapy trials for β-thalassemia are contemplated, 2 critical questions important to trial design and planning have emerged. First, what proportion of genetically corrected hematopoietic stem cells (HSCs) will be needed to achieve a therapeutic benefit? Second, what level of expression of a transferred globin gene will be required to improve β-thalassemic erythropoiesis? These questions were directly addressed by means of a murine model of severe β-thalassemia. Generation ...

NaCl has been detected toward the circumstellar envelope of the post-AGB star CRL 2688 using the IRAM 30 m telescope, the first time this molecule has been identified in a source other than IRC +10216. The J = 7 → 6, 11 → 10, 12 → 11, and 18 → 17 transitions of NaCl at 1, 2, and 3 mm have been observed, as well as the J = 8 → 7 line of the 37Cl isotopomer. The J = 12 → 11 line was also measured at the ARO 12 m telescope. An unsuccessful search was additionally conducted for AlCl toward CRL 2688, although in the process ...

Patrick W. Kelly, David W. Pfennig, Sofia De La Serna Buzón, Karin S. Pfennig,

In a rapidly changing world, understanding the processes that influence a population's ability to respond to natural selection is critical for identifying how to preserve biodiversity. Two such processes are phenotypic plasticity and sexual selection. Whereas plasticity can facilitate local adaptation, sexual selection potentially impedes local adaptation, especially in rapidly changing or variable environments. Here we hypothesize that, when females preferentially choose males that sire plastic ...

There have been few studies of patients with renal allografts functioning for more than 20 years. We sought to identify clinical factors associated with ultra long-term (>20 year) renal allograft survival and to describe the clinical features of these patients. We performed a retrospective analysis of the Irish Renal Transplant Database and included 1174 transplants in 1002 patients. There were 255 (21.74%) patients with graft function for 20 years or more. Multivariate analysis identified recipient ...

Moroctocog alfa (AF-CC) (Xyntha, BDDrFVIII) is manufactured by a process designed to enhance the theoretical viral safety profile relative to ReFacto, its predecessor, and to provide alignment with clinical monitoring by the one-stage clotting assay. To evaluate the efficacy and safety of B-domain-deleted recombinant factor VIII (BDDrFVIII) was given as bolus injection (BI) or continuous infusion (CI) in haemophilia patients undergoing major surgery. BDDrFVIII was administered by BI or CI per investigator ...

Prophylaxis is increasingly prescribed in treatment of haemophilia and its benefit is believed to be most significant for the youngest patients as haemophilic arthropathy may be prevented if prophylaxis is initiated prior to recurrent haemarthroses. While clinical prophylaxis data are readily available for haemophilia A, analogous data for haemophilia B are relatively limited. A prospective clinical study of recombinant factor IX (BeneFIX; rFIX), designed to allow investigator prescribed prophylaxis ...

To present the long-term outcome of deceased-donor kidney transplantation in prune belly syndrome (PBS). All the PBS transplant patients had undergone extensive pretransplant urologic tract preparation. We also compared the patient and graft survival in an age-matched control group who underwent transplantation for end-stage renal disease due to a nonposterior urethral valve in our urology department. From 1988 to 2003, 11 kidneys were transplanted in 8 male patients with PBS. Hannu T. Aro, Patrick J. Kelly, David G. Lewallen, Edmund Y.S. Chao,

The effects of early physiologic dynamic compression on fracture healing were studied in the dog. Transverse midtibial osteotomies were performed bilaterally and stabilized with a relatively rigid external fixation system in a neutralization mode (800 μm) to prevent compression of the osteotomy ends during weight bearing. On the 15th day, one osteotomy in each animal was subjected to dynamic compression through weight bearing by release of the fixator-telescoping mechanism (axial dynamization), while ...

Jenifer Jowsey, B. Lawrence Riggs, Patrick J. Kelly, David L. Hoffman,

Calcitonin has been considered of therapeutic value in osteoporosis because of its effects in tissue culture. In the whole animal, however, the predominant result seems to be hypocalcemia, which might be expected to have the opposite effect of stimulating parathyroid hormone secretion and therefore resorption of bone. Indeed, in a short term study of 3- and 4-month duration in osteoporotic women, this was found to be so. A combination of calcium and calcitonin was therefore considered a more promising ...
