Purpose Rare missense substitutions and in-frame deletions of BRCA1 and BRCA2 genes present a challenge for genetic counseling of individuals carrying such unclassified variants. We assessed the value of tumor immunohistochemical markers in conjunction with genetic and evolutionary approaches for investigating the clinical significance of unclassified variants. Patients and Methods We studied 10 BRCA1 and 12 BRCA2 variants identified in Australian families with breast cancer. Analyses assumed a ...

Small cell carcinoma of the lung has been shown to be exquisitely responsive to chemotherapy. Unfortunately, these responses are often short in duration and long-term disease-free survival is infrequent. This review of the records of all patients with small cell carcinoma of the lung treated on protocol at The Johns Hopkins Oncology Center from 1973 to 1982 showed that 25 of 225 (11.1 percent) survived two years or longer. Patients with limited disease (20 of 94) and patients with a complete response ( ...

The echocardiographic assessment of circulatory function in sick newborn infants has the potential to improve patient care. However, measurements are prone to error and have not been sufficiently validated. Phase-contrast magnetic resonance imaging (MRI) provides highly validated measures of blood flow and has recently been applied to the newborn population. The aim of this study was to validate measures of left ventricular output and superior vena caval flow volume in newborn infants.Echocardiographic ...

The primary objective of the game netball is to score more goals than the opposition. Subsequently, there is impetus on improving shooting accuracy to maximize scoring. Understanding the kinematic factors of shooting that lead to scoring success allows for iterative adjustment and optimization of technique. Three-dimensional retro-reflective motion capture has been used to assess kinematics; however, these systems are expensive, require substantial setup and postprocessing time, and cannot be used ...

It is often desirable or necessary to store collected seawater samples prior to analysis for dissolved inorganic nutrients. It is therefore important to establish preservation and storage techniques that will ensure sample integrity and will not alter the precision or accuracy of analysis. We have performed a series of experiments on the storage of nutrient samples collected at the oligotrophic North Pacific benchmark Station ALOHA, using both standard autoanalyses and low-level techniques. The estrogen receptors ERα and ERβ are recognized as important pharmaceutical targets for a variety of diseases including osteoporosis and breast cancer. A series of novel benzoxepin-derived compounds are described as potent selective modulators of the human estrogen receptor modulators (SERMs). We report the antiproliferative effects of these compounds on human MCF-7 breast tumor cells. These heterocyclic compounds contain the triarylethylene arrangement as exemplified by tamoxifen, conformationally ...

Lennox-Gastaut syndrome (LGS) is a severe developmental epileptic encephalopathy diagnosed in childhood that persists through adolescence and into adulthood. While the characteristics of LGS in pediatric patients are well defined, including "drop attacks", interictal slow spike and wave electroencephalogram (EEG) activity, and intellectual disability, these features can evolve over time, and different EEG activities may be present in adult patients with LGS. This may result in missed diagnoses in these ...

The most northwesterly outcrop of Lower Cretaceous Chaswood Formation is in a pit at Vinegar Hill, south of Sussex, New Brunswick. New mapping and boreholes show thick, fluvial, loosely lithified conglomerates and lesser sandstones unconformably overlying 12 m of mudstone in a 1 km 2 basin bounded to the northwest by the Clover Hill fault. Sparse paleocurrent indicators to the southwest parallel this fault. The tectonic setting is similar to that of the Chaswood Formation in the fault-bounded Elmsvale ...

The safety and immunogenicity of REMUNE, an HIV-specific immune based therapy for HIV infection, was evaluated in a cohort of 30 HIV infected subjects in Thailand. This therapy utilizes a gp120 depleted inactivated virus (HZ321), which exhibits a high degree of conservation with the core antigens of both type B' and E strains of HIV, the predominant Thailand isolates. The treatment was well tolerated, with no serious adverse events reported over the course of the 4-month trial. Treatment in which ...

Abstract BACKGROUND p53 is a transcription factor that regulates the cell cycle, DNA repair, and apoptosis. A variant at codon 72, rs1042522, results in altered activities for p53 and is, notably, differentially distributed among different ethnic populations. However, associations of this variant with cancer in men of African descent have not been explored. Herein, we tested the hypothesis that rs1042522 was associated with prostate cancer (PCa) risk. MATERIALS AND METHODS Genotypes were determined ...

Background Pediatric inflammatory bowel disease (IBD) is challenging to diagnose because of the non-specificity of symptoms; an unequivocal diagnosis can only be made using colonoscopy, which clinicians are reluctant to recommend for children. Diagnosis of pediatric IBD is therefore frequently delayed, leading to inappropriate treatment plans and poor outcomes. We investigated the use of 16S rRNA sequencing of fecal samples and new analytical methods to assess differences in the microbiota of children ...
