Abstract Hyphal interactions between two antagonistic binucleate Rhizoctonia isolates (BNR) and the seedling dampingoff pathogen, Pythium ultimum var. sporangiiferum, were observed by both light‐and scanning electron microscopy (SEM), on agar media and on capsicum seeds in sterilized potting mix. Three techniques for making agar films are compared, and two methods of estimating hyphal lengths are discussed. Hyphal length was less variable in agar films made using coverslip wells than those made using hemacytometers or micropipettes. Examples are given showing the use of variance estimates in optimizing experimental designs.
