Summary Background Faricimab is a bispecific antibody that acts through dual inhibition of both angiopoietin-2 and vascular endothelial growth factor A. We report primary results of two phase 3 trials evaluating intravitreal faricimab with extension up to every 16 weeks for neovascular age-related macular degeneration (nAMD). Methods TENAYA and LUCERNE were randomised, double-masked, non-inferiority trials across 271 sites worldwide. Treatment-naive patients with nAMD aged 50 years or older were randomly ...

We propose a new algorithm for sampling the N-body density ∣Ψ(R)∣2∕∫R3N∣Ψ∣2 in the variational Monte Carlo framework. This algorithm is based upon a modified Ricci-Ciccotti discretization of the Langevin dynamics in the phase space (R,P) improved by a Metropolis-Hastings accept/reject step. We show through some representative numerical examples (lithium, fluorine, and copper atoms and phenol molecule) that this algorithm is superior to the standard sampling algorithm based on the biased random walk ( ...

Abstract Background Oxidized low‐density lipoproteins play key roles in atherosclerosis. Their toxicity is at least in part due to 7‐ketocholesterol (7KC), which is a potent inducer of apoptosis. In this study on human promonocytic U937 cells, we determined the effects and the interactions of 7KC with cellular lipids during 7KC‐induced apoptosis. Methods Morphologic and functional changes were investigated by microscopic and flow cytometric methods after staining with propidium iodide, 3,3′‐dihexyloxacarbocyanine ...

We have recently formulated a new approach, named the effective local potential (ELP) method, for calculating local exchange-correlation potentials for orbital-dependent functionals based on minimizing the variance of the difference between a given nonlocal potential and its desired local counterpart [V. N. Staroverov et al., J. Chem. Phys. 125, 081104 (2006)]. Here we show that under a mildly simplifying assumption of frozen molecular orbitals, the equation defining the ELP has a unique analytic solution ...

The equations of the temperature-accelerated molecular dynamics (TAMD) method for the calculations of free energies and partition functions are analyzed. Specifically, the exponential convergence of the law of these stochastic processes is established, with a convergence rate close to the one of the limiting, effective dynamics at higher temperature obtained with infinite acceleration. It is also shown that the invariant measures of TAMD are close to a known reference measure, with an error that ...

Linear induction variable detection is usually associated with the strength reduction optimization. For restructuring compilers, effective data dependence analysis requires that the compiler detect and accurately describe linear and nonlinear induction variables as well as more general sequences. In this article we present a practical technique for detecting a broader class of linear induction variables than is usually recognized, as well as several other sequence forms, including periodic, polynomial, ...

The oil sands industry usually leads to the production of large quantities of mineral waste, such as fluid fine tailings (FFT), whose disposal is often challenging. Electrokinetic geocomposites (eGCPs) installed into the FFT disposal area may improve in situ dewatering, as eGCPs can drain water expulsed during FFT consolidation as well as impose a voltage across FFT to displace water by electro-osmosis. This paper presents a laboratory device specifically developed to evaluate eGCP performance for ...

Hepatic cytochrome P450s play a critical role in the metabolism of hydrophobic xenobiotics. One of the major unsolved problems in xenobiotic metabolism is the molecular mechanism whereby phenobarbital induces hepatic enzymes, particularly CYP2B1 and CYP2B2 in rat liver. By using primary rat hepatocytes for transfection analyses, we previously identified in the CYP2B2 5′-flank a 163-base pair Sau3AI fragment that confers phenobarbital inducibility on a cat reporter gene and that has the properties ...

Summary Background To reduce treatment burden and optimise patient outcomes in diabetic macular oedema, we present 1-year results from two phase 3 trials of faricimab, a novel angiopoietin-2 and vascular endothelial growth factor-A bispecific antibody. Methods YOSEMITE and RHINE were randomised, double-masked, non-inferiority trials across 353 sites worldwide. Adults with vision loss due to centre-involving diabetic macular oedema were randomly assigned (1:1:1) to intravitreal faricimab 6·0 mg every 8 weeks, ...

Lentiviral-mediated integration of a CFTR transgene cassette into airway basal cells is a strategy being considered for cystic fibrosis (CF) cell-based therapies. However, CFTR expression is highly regulated in differentiated airway cell types and a subset of intermediate basal cells destined to differentiate. Since basal stem cells typically do not express CFTR, suppressing the CFTR expression from the lentiviral vector in airway basal cells may be beneficial for maintaining their proliferative ...

We present here a formulation of the electronic ground-state energy in terms of the second order reduced density matrix, using a duality argument. It is shown that the computation of the ground-state energy reduces to the search of the projection of some two-electron reduced Hamiltonian on the dual cone of N-representability conditions. Some numerical results validate the approach, both for equilibrium geometries and for the dissociation curve of N(2).

Mucus produced by submucosal glands is a key component of respiratory mucociliary transport (MCT). When it emerges from submucosal gland ducts, mucus forms long strands on the airway surface. However, the function of those strands is uncertain. To test the hypothesis that mucus strands facilitate transport of large particles, we studied newborn pigs. In ex vivo experiments, interconnected mucus strands moved over the airway surface, attached to immobile spheres, and initiated their movement by pulling ...

Significance Mucociliary transport (MCT) defends lungs by removing particulates, and defective MCT is hypothesized to contribute to the onset of lung diseases such as asthma, chronic bronchitis, and cystic fibrosis. However, testing those hypotheses has been limited by current MCT assays and mouse models of human disease. We developed an in vivo MCT assay in newborn pigs, which share physiological and anatomical features with humans. The X-ray–computed tomographic-based method provided high spatial ...

The Hartree-Fock exchange operator is an integral operator arising in the Hartree-Fock method and replaced by a multiplicative operator (a local potential) in Kohn-Sham density functional theory. This article presents a detailed analysis of the mathematical properties of various local approximations to the nonlocal Hartree-Fock exchange operator, including the Slater potential, the optimized effective potential (OEP), the Krieger-Li-Iafrate (KLI) and common energy-denominator approximations (CEDA) to ...

Significance In many lung diseases, increased amounts of and/or abnormal mucus impair mucociliary clearance, a key defense against inhaled and aspirated material. Submucosal glands lining cartilaginous airways secrete mucus strands that are pulled by cilia until they break free from the duct and sweep upward toward the larynx, carrying particulates. In cystic fibrosis (CF) pigs, progressive clearance of insufflated microdisks was repeatedly interrupted as microdisks abruptly recoiled. Aerosolizing ...
