Amyloid deposits with Arg124 mutated TGFBI protein have been identified in autosomal dominant blinding corneal dystrophies. We assessed in vitro the mechanisms determining TGFBI protein amyloid transformation involving mutations of Arg124. Eight peptides synthesized following the TGFBI protein sequence, centered on codon Arg124 holding the previously reported amyloidogenic mutations and the respective controls were studied. Cys124 and His124 mutated peptide preparations contained significantly higher ...

Purpose. To assess the morphologic differences of three types of lattice corneal dystrophies (LCDs) from histologic, immunohistochemical, and ultrastructural studies. Methods. Corneas from three patients, one LCD1, one His626Arg-LCD, and one LCD3A were processed for Congo red, βig-h3541–564 antibodies immunostaining, and electron transmission microscopy studies. Control tissues were submitted to identical analyses and consisted of one cornea from a patient not having LCD and one skin biopsy from ...

BetaIGH3 protein has been recently involved in the pathogenesis of blinding corneal diseases, some of which have characteristic amyloid corneal deposits. The 124 codon of the betaig-h3 gene seems to be crucial for the amyloidogenicity of the protein product. We presently report an in vitro system that reproducibly forms amyloid fibrils from betaIGH3((110-131)) derived peptides. We also assessed the differences in fibril formation of two 22-amino acid peptides centered on the 124 residue: the native ...

We screened family members of monozygotic twins with keratoconus to search for a corneal thinning disorder using clinical, videokeratographic, and pachymetric analyses. The parents had bilateral astigmatism of a symmetric bow-tie pattern with normal videokeratographic and pachymetric indices. The 8-year-old sister had slightly asymmetric astigmatism in both eyes with normal pachymetry values. No family member was suspected of a corneal thinning disorder. We discuss the mode of inheritance of keratoconus ...

ABSTRACT PURPOSE: Corneal thinning disorders weaken the mechanical strength of affected corneas, suggesting that photorefractive procedures may be contraindicated in keratoconus. Few cases have been reported to confirm this hypothesis. METHODS: A 45-year-old man had two laser in situ keratomileusis (LASIK) procedures and one photorefractive keratectomy (PRK) performed on his left eye, and three LASIK procedures on his right eye. After these surgeries, a dramatic corneal ectasia and gl'ade III haze ...

Motor neuron diseases (MNDs) are characterized by selective death of motor neurons and include mainly adult-onset amyotrophic lateral sclerosis (ALS) and spinal muscular atrophy (SMA). Neurodegeneration is not the single pathogenic event occurring during disease progression. There are multiple lines of evidence for the existence of defects in lipid metabolism at peripheral level. For instance, hypermetabolism is well characterized in ALS, and dyslipidemia correlates with better prognosis in patients. ...

Fatty acids are of paramount importance to all cells, since they provide energy, function as signaling molecules, and sustain structural integrity of cellular membranes. In the nervous system, where fatty acids are found in huge amounts, they participate in its development and maintenance throughout life. Growing evidence strongly indicates that fatty acids in their own right are also implicated in pathological conditions, including neurodegenerative diseases, mental disorders, stroke and trauma. ...

The progressive deterioration of the neuromuscular axis is typically observed in degenerative conditions of the lower motor neurons, such as amyotrophic lateral sclerosis (ALS). Neurodegeneration in this disease is associated with systemic metabolic perturbations, including hypermetabolism and dyslipidemia. Our previous gene profiling studies on ALS muscle revealed down-regulation of delta-9 desaturase, or SCD1, which is the rate-limiting enzyme in the synthesis of monounsaturated fatty acids. Interestingly, ...

The RPE65 protein appears late during the retinal development. To study the basis for this regulation, the rat RPE65 cDNA was sequenced and the mRNA subsequently quantitated at various stages by competitive RT‐PCR. RPE65 mRNA was detected as early as E18 (36 copies/ng of whole eye total RNA). It gradually accumulates up to P12 (27 000 copies/ng) at which point it reaches a steady state level. This increase is interrupted for 3 days (P2–P4) during which the levels of mRNA remain stable. This timing ...

The use of bicoronal incisions has been sufficiently described in neurosurgery and craniofacial surgery including osteotomies and injuries. This approach provides excellent surgical access for nasal reconstruction with a very low rate of morbidity. A series of 11 patients, together with three case reports, illustrates the advantages and possible complications of this type of incision.

Purpose: To assess and compare the accuracy of 2 methods of predicting visual acuity after phacoemulsification. Setting: Department of Ophthalmology, Montpellier, France. Methods: This prospective study evaluated 47 eyes of 47 patients having uneventful phacoemulsification over a 1-month period. All the patients had mild to moderate cataract. Visual acuity recovery was predicted using the white-light IRAS GT® interferometer on the 3- and 8-degree wide test area and the Guyton-Minkowski potential ...

A 25-year-old man sustained bilateral ocular trauma from an explosion, which resulted in ocular injuries from the blast, mineral projections, and heat. Before the accident, the patient had eight-incision radial keratotomy in the left eye followed 1 week later by photorefractive keratectomy in the right eye. After the accident, the left cornea had a full-thickness rupture of four incisions; the fellow cornea had a full-thickness laceration from a mineral projection. Five months after the accident, ...

Infrastructures create many problems in the security, economic or health sector. The high level of connectivity on Internet of Things (IoT) field is at the center of new networks and requires interoperable solutions that promote redundancy, and that fault tolerance. Many standard protocols currently allow extensive exchanges between different IoT groups. In this paper, we are interested in reconfigurable links management between sets of IoT linked by Message Queue Telemetry Transport (MQTT). This ...

High-fidelity numerical simulations are used to study flame root stabilization mechanisms of cryogenic flames, where both reactants (O2 and CH4) are injected in transcritical conditions in the geometry of the laboratory scale test rig Mascotte operated by ONERA (France). Simulations provide a detailed insight into flame root stabilization mechanisms for these diffusion flames: they show that the large wall heat losses at the lips of the coaxial injector are of primary importance, and require to solve ...

Rational design toward macrocellular silica scaffolds has been reached. The first technique employed is based on the use of concentrated direct emulsions as a macroscale pattern where the final monolith textures can vary upon the oil volumic fraction, emulsification process, and pH conditions. Also, in order to improve void spaces alignment we can make the use of external magnetic field. In this issue, an hydrophobic Ferro-fluid replaces the oily phase. The second approach is rather focussed on ...

This work reports a new evidence of the versatility of soft responsive microgels as stabilizers for Pickering emulsions. The organization of microgels at the oil–water interface is a function of the preparation pathway. The present results show that emulsification energy can be used as a trigger to modify microgel deformation at the oil–water interface and their packing density: high shear rates bring strong flattening of the microgels, whereas low shear rates lead to dense monolayers, where the ...
