S-nitrosylation of nuclear factor κB (NF-κB) on the p65 subunit of the p50/p65 heterodimer inhibits NF-κB DNA binding activity. We have recently shown that p65 is constitutively S-nitrosylated in the lung and that LPS-induced injury elicits a decrease in SNO-p65 levels concomitant with NF-κB activation in the respiratory epithelium and initiation of the inflammatory response. Here, we demonstrate that TNFα-mediated activation of NF-κB in the respiratory epithelium similarly induces p65 denitrosylation. ...

Richard J. Forrester, Harvey J. Greenberg,

In this paper we formulate four problems in computational mo lecular biology as 0-1 quadratic programs. These problems are all NP-hard, and the current solution methods u sed in practice consist of heuristics or approximation algorithms tailored to each problem. Using test problems fr scientific databases, we address the question, “Can a general-purpose solver obtain good answers in reasonable t ime?” In addition, we use the latest heuristics as incumbent solutions to address the question, “Can a ...

Ria Patel, Joel Harvey, Andrew Forrester,

There is a high prevalence of mental health need in prisons, much of which is currently unmet. Although considerable research has identified and described this mental health need, there has been limited research focussed on reviewing the delivery of mental health care in prisons. This study uses content analysis to review 36 unannounced prison inspection reports in England to establish whether mental health care was provided to an appropriate standard, and whether it is equivalent to services that ...

Right atrial pacing was performed in 41 subjects with coronary heart disease. Twenty developed angina pectoris during pacing, while 21 did not. The extent of coronary artery disease, as judged by selective cinearteriography, was similar in the two groups. Both had significant increases in heart rate and pressure-time per minute, but there was no significant difference in either of these parameters between groups. Among the hemodynamic parameters measured, the only statistically significant change ...

Dynamic control of thioredoxin (Trx) oxidoreductase activity is essential for balancing the need of cells to rapidly respond to oxidative/nitrosative stress and to temporally regulate thiol-based redox signaling. We have previously shown that cytokine stimulation of the respiratory epithelium induces a precipitous decline in cell S-nitrosothiol, which depends upon enhanced Trx activity and proteasome-mediated degradation of Txnip (thioredoxin-interacting protein). We now show that tumor necrosis factor- ...

The cytokine-inducible isoform of nitric oxide synthase (NOS2) is constitutively expressed in human respiratory epithelia and is upregulated in inflammatory lung disease. Here, we sought to better define the protein interactions that may be important for NOS2 activity and stability, as well as to identify potential targets of NOS2-derived NO, in the respiratory epithelium. We overexpressed Flag-tagged, catalytically-inactive NOS2 in A549 cells and used mass spectrometry to qualitatively identify ...

The effects of increased heart rate upon the physical and metabolic determinants of coronary blood flow in coronary heart disease provide a physiologic basis for the use of this investigative tool in cardiovascular studies. Heart rate increased by electrical pacing has minimal effects upon other hemodynamic indexes and arterial substrate concentrations, thus providing a considerable advantage over exercise and isoproterenol as a relatively uncomplicated myocardial stress. Atrial pacing represents ...

A review of Urban Dynamics [1] and additional simulation by the authors is presented. Structural choices of boundary conditions, major subsectors, and multiplier relationships in the Forrester model are examined. Some of their implications and shortcomings are noted. Forrester's policy conclusions and implicit evaluation criteria are discussed and related to our alternative assumptions and simulation.

An algorithm for converting signed digraphs to Forrester schematics is described and illustrated. A theoretical framework for the algorithm consisting of theorems is developed first. This theoretical framework is based upon discussions found in a companion paper [1]. Specifically, the algorithm uses the definitional properties of the various quantities and couplings, the consistency supposition, and related axioms and theorems as outlined in [1]. The algorithm is applied to an illustrative example.

Nitric oxide exerts a plethora of biological effects via protein S-nitrosylation, a redox-based reaction that converts a protein Cys thiol to a S-nitrosothiol. However, although the regulation of protein S-nitrosylation has been the subject of extensive study, much less is known about the systems governing protein denitrosylation. Most recently, thioredoxin/thioredoxin reductases were shown to mediate both basal and stimulus-coupled protein denitrosylation. We now demonstrate that protein denitrosylation ...

Our goals were to develop a rabbit model of subacute stent thrombosis and to evaluate the efficacy of a polymer stent coating with covalently bound heparin for its prevention. Bare metal stents (a nitinol wire coil with 3 stainless steel joints) were implanted in a carotid artery of 10 NZW rabbits with a 3.0 mm balloon catheter, inflated twice at 6 atm for 1 minute. Systemic antiplatelet and antithrombotic therapy was withheld. Animals were sacrificed at 24, 72 and 96 hours. Only the 96 hour group ...

While there are hundreds of predicted E3 ligases, characterizing their applications for targeted protein degradation has proved challenging. Here, we report a chemical biology approach to evaluate the ability of modified recombinant E3 ligase components to support neo-substrate degradation. Bypassing the need for specific E3 ligase binders, we use maleimide-thiol chemistry for covalent functionalization followed by E3 electroporation (COFFEE) in live cells. We demonstrate that electroporated recombinant ...
