In 2006, we reported a mariner (Mos1)-transformed Aedes aegypti line, Carb77, which was highly resistant to dengue-2 virus (DENV2). Carb77 mosquitoes expressed a DENV2-specific inverted-repeat (IR) RNA in midgut epithelial cells after ingesting an infectious bloodmeal. The IR-RNA formed double-stranded DENV2-derived RNA, initiating an intracellular antiviral RNA interference (RNAi) response. However, Carb77 mosquitoes stopped expressing the IR-RNA after 17 generations in culture and lost their DENV2- ...

Abstract Transgenic Aedes aegypti were engineered to express a virus‐derived, inverted repeat (IR) RNA in the mosquito midgut to trigger RNA interference (RNAi) and generate resistance to dengue virus type 2 (DENV2) in the vector. Here we characterize genotypic and phenotypic stabilities of one line, Carb77, between generations G 9 and G 17 . The anti‐DENV2 transgene was integrated at a single site within a noncoding region of the mosquito genome. The virus resistance phenotype was strong until G 13 ...

Mosquitoes ( Aedes aegypti ) were genetically modified to exhibit impaired vector competence for dengue type 2 viruses (DENV-2). We exploited the natural antiviral RNA interference (RNAi) pathway in the mosquito midgut by constructing an effector gene that expresses an inverted-repeat (IR) RNA derived from the premembrane protein coding region of the DENV-2 RNA genome. The A. aegypti carboxypeptidase A promoter was used to express the IR RNA in midgut epithelial cells after ingestion of a bloodmeal. The ...

Trafficking of α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (AMPARs) is regulated by specific interactions with other proteins and by post-translational mechanisms, such as phosphorylation. We have found that the type II cGMP-dependent protein kinase (cGKII) phosphorylates GluA1 (formerly GluR1) at S845, augmenting the surface expression of AMPARs at both synaptic and extrasynaptic sites. Activation of cGKII by 8-Br-cGMP enhances the surface expression of GluA1, whereas its inhibition ...

The purpose of this work was to (1) establish an in vitro methodology to study dissolution and precipitation in early stages of drug development where low compound consumption and high throughput are necessary, (2) develop a mathematical model for a mechanistic explanation of generated in vitro dissolution and precipitation data, and (3) extrapolate in vitro data to in vivo situations using ...

Cryobanking skin samples permit preserving a maximum of genetic representation from the population biodiversity. This is a relevant aspect for threatened species, potentially menaced by an epizooty and from which it is difficult to obtain gametes. As a first step for properly cryobanking skin samples of a given species, the optimal conditions of culture and freezing have to be studied by covering a broad range of possibilities. This paper presents, for the first time, a systematic study of such ...

We study in detail the ratchetlike dynamics of topological solitons in homogeneous nonlinear Klein-Gordon systems driven by a biharmonic force. By using a collective coordinate approach with two degrees of freedom, namely the center of the soliton, X(t), and its width, l(t), we show, first, that energy is inhomogeneously pumped into the system, generating as result a directed motion; and, second, that the breaking of the time shift symmetry gives rise to a resonance mechanism that takes place whenever ...

We investigate the ratchet dynamics of nonlinear Klein-Gordon kinks in a periodic, asymmetric lattice of pointlike inhomogeneities. We explain the underlying rectification mechanism within a collective coordinate framework, which shows that such a system behaves as a rocking ratchet for point particles. Careful attention is given to the kink width dynamics and its role in the transport. We also analyze the robustness of our kink rocking ratchet in the presence of noise. We show that the noise activates ...

We study directed energy transport in homogeneous nonlinear extended systems in the presence of homogeneous ac forces and dissipation. We show that the mechanism responsible for unidirectional motion of topological excitations is the coupling of their internal and translation degrees of freedom. Our results lead to a selection rule for the existence of such motion based on resonances that explains earlier symmetry analysis of this phenomenon. The direction of motion is found to depend both on the ...

We study the dynamics of straight phi(4) kinks perturbed by an ac force, both with and without damping. We address this issue by using a collective coordinate theory, which allows us to reduce the problem to the dynamics of the kink center and width. We carry out a careful analysis of the corresponding ordinary differential equations, of Mathieu type in the undamped case, finding and characterizing the resonant frequencies and the regions of existence of resonant solutions. We verify the accuracy ...

We investigate the nonparametric, pure ac driven dynamics of nonlinear Klein-Gordon solitary waves having an internal mode of frequency Omega(i). We show that the strongest resonance arises when the driving frequency delta = Omega(i)/2, whereas when delta = Omega(i) the resonance is weaker, disappearing for nonzero damping. At resonance, the dynamics of the kink center of mass becomes chaotic. As we identify the resonance mechanism as an indirect coupling to the internal mode due to its symmetry, we ...

We study whether or not sine-Gordon kinks exhibit internal modes or ``quasimodes.'' By considering the response of the kinks to ac forces and initial distortions, we show that neither intrinsic internal modes nor ``quasimodes'' exist in contrast to previous reports. However, we do identify a different kind of internal mode bifurcating from the bottom edge of the phonon band which arises from the discretization of the system in the numerical simulations, thus confirming recent predictions.

The single X was paternal in origin (X p ) in 10 cases (22.2%) and maternal (X m ) in 35 cases (77.8%). Y chromosome material was detected in 1 out of the 35 cases with a 45, X m constitution. Analysis of parental ages and clinical data of the patients with respect to the origin of the single X revealed no significant differences between the origins.
