Abstract: Pituitary adenylate cyclase‐activating polypeptide (PACAP) is a pleiotropic neuropeptide that belongs to the secretin/glucagon/vasoactive intestinal peptide (VIP) family. The administration of systemic corticosteroids is partially effective, but continuous treatment causes severe side effects. PACAPによる海馬細胞死防御とその分子制御機構について,機能形態学的方法を用いて調べた.ラットおよびマウスの虚血-再灌流モデルでは,海馬領域の特にCA1の神経細胞が多数遅発性細胞死をおこす.超微量(pM, nMオーダー)のPACAPを脳室内および静脈内投与すると,この遅発性神経細胞死は著明に抑制されることが明らかになった.さらに,再灌流1日後からPACAPを脳室内および静脈内投与しても神経細胞死を有意に抑制することが明らかになった.また神経細胞死のおきる部位に一致して星状膠細胞にPACAP受容体が強く発現した.また,外傷モデルのEGFP-GFAPトランスジェニックマウスでも,虚血実験と同じく星状膠細胞にPACAP受容体が特異的に発現した.さらに細胞死のシグナル伝達系路として最も重要であると考えられているMAPキナーゼカスケードについて虚血モデル動物の海馬領域を採取して調べたところ,神経細胞死とともにこの系路が活性化しているのが明らかになった.PACAPはこの系路を介して遅発性神経細胞死を抑制することが明らかになった.また,PACAPによる虚血性神経細胞死抑制作用として,PACAPが星状膠細胞を刺激してIL- ...
