Recent 13C nuclear magnetic resonance (13C-NMR) studies in the anesthetized rat and perfused liver suggest that hepatic glycogen is simultaneously synthesized and degraded, even during combined hyperglycemia and hyperinsulinemia. When methods were used that permit inactivation of α-amylase immediately after breaking the cells, the amount of maltosyloligosaccharides in liver was found to be 30- to 40-fold smaller than previously reported. With these procedures no change in maltosyloligosaccharide content was detected either under normal conditions or in some pathological situations. The evidence presented indicates that a major proportion of the oligosaccharides found in previous investigations was formed during homogenization.
