(±)-5-([4-[2-Methyl-2(pyridylamino)ethoxy]phenyl]methyl) 2,4-thiazolidinedione (BRL 49653) is a new potent antidiabetic agent that improves insulin sensitivity in animal models of NIDDM. In C57BL/6 obese (ob/ob) mice, BRL 49653, included in the diet for 8 days, improved glucose tolerance. The half-maximal effective dose was 3 μmol/kg diet, which is equivalent to ∼0.1 mg/kg body wt. Improvements in glucose tolerance were accompanied by significant reductions in circulating triacylglycerol, nonesterified ...

Geoffrey Holder,

The performance of methods used for the measurement of urinary-free cortisol (UFC) has been monitored in the UK by an external quality assessment scheme (EQAS). Bias and variation of the bias (VAR) from the all laboratories trimmed mean (ALTM) show that mean bias ranged from −15·5% to + 83·1% and mean VAR from 9·3% to 53·2%. The dominant kit methods had a bias range of −4·2% and + 56·9% and VAR between 11·4% and 18·4%. Recovery of added cortisol ranged from 74·9% to 207·2%. The precision of the estimate of ...

Conventional insulin concentration units (IU/mL or just U/mL) are bioefficacy based, whereas the Système International (SI) units (pmol/L) are mass based. In converting between these two different approaches, there are at least 2 well-accepted conversion factors, where there should be only 1. The correct value is not the most-used or well-accepted using online calculators, some journal styles, laboratory reports, and published articles. In short, an incorrect insulin conversion factor is widely used ...

A Venturi tube ( D1=15mm,D2=10mm ) connected to a differential pressure sensor (SDP816-125 Pa) allows accurate measurement of flow between 5-75L·min-1 , with Pearson Correlation over 4 min at 50Hz⩾0.97 , and distance correlation ⩾0.96 . The pressure measurement was similarly accurate using a MPVZ4006GW7U. ...

An account is given of an investigation into the interaction between the boundary layer on a flat plate and a shock wave produced either externally, by a wedge in the supersonic mainstream, or from within the boundary layer, by a wedge held in contact with the plate. A wide range of free-stream Mach numbers, boundary-layer Reynolds numbers, and shock strengths has been covered, shock strength being defined as the ratio of the static pressure downstream of the shock to the static pressure upstream ...

Immune responses against an introduced transgenic protein are a potential risk in many gene replacement strategies to treat genetic disease. We have developed a gene delivery approach for hemophilia B based on lentiviral expression of human factor IX in purified hematopoietic stem cells. In both normal C57Bl/6J and hemophilic 129/Sv recipient mice, we observed the production of therapeutic levels of human factor IX, persisting for at least a year with tolerance to human factor IX antigen. Respiratory model-based methods require datasets containing enough dynamics to ensure model identifiability for development and validation. Rapid expiratory occlusion has been used to identify elastance and resistance within a single breath. Currently accepted practice for rapid expiratory occlusion involves a 100 ms occlusion of the expiratory pathway. This article presents a low-cost modular rapid shutter attachment to enable identification of passive respiratory mechanics. Shuttering faster than ...
