Altered transcription is a cardinal feature of acute myeloid leukemia (AML); however, exactly how mutations synergize to remodel the epigenetic landscape and rewire three-dimensional DNA topology is unknown. Here, we apply an integrated genomic approach to a murine allelic series that models the two most common mutations in AML: Flt3-ITD and Npm1c. We then deconvolute the contribution of each mutation to alterations of the epigenetic landscape and genome organization, and infer how mutations synergize ...

A. D. Osborne, George Porter,

The rate constants of quenching of the triplet state of naphthalene by α -iodonaphthalene, t -butyl hydroperoxide and oxygen have been studied in several solvents as a function of viscosity and temperature. The diffusion controlled rate constants for α -iodonaphthalene in hydroxylic media agree with those predicted by the equation k calc. = 8 RT /2000 ƞ ( ƞ being viscosity). This is interpreted as evidence for the validity of the Stokes‒Einstein expression, Stoke’s law with ‘slip’ (i. e. coefficient of sliding ...

Since inhibition of myo-inositol monophosphatase (EC 3.1.3.25) by lithium ions and the resulting attenuation of phosphatidylinositol cycle activity may be the mechanism by which lithium exerts its therapeutic effect in the treatment of manic depression, it is of great interest to understand the mechanism of the enzyme and how lithium and other metals interact with it. Divalent magnesium is essential for enzyme activity, whereas Li+ and high concentrations of Mg2+ act as uncompetitive inhibitors with ...

Neville N. Osborne, George S. Richardson,

Isolated bovine retinas possess the ability to concentrate tryptamine from an external medium. The entry mechanism is independent of sodium and other cations, but is dependent on temperature. Kinetic analysis of the tryptamine uptake shows a single mechanism with Km and Vmax values of 2 × 10−5m and 50 pmol/mg/min respectively. A lack of specificity in the process is indicated by its insensitivity to a number of metabolic inhibitors and various analogues. Autoradiography has shown that specific neuronal ...

ADV is a non-defective parvovirus (single stranded DNA virus), which causes a persistent infection in mink and a slowly progressive lymphoproliferative disease, characterised by plasmacytosis, hypergammaglobulinaemia and immune complex-mediated glomerulonephritis and arteritis.Earlier vaccination efforts have failed so far, probably because the vaccines contained low amounts of inactivated virus. In one report (Porter et al., J. We have genotyped 53 individuals from three ethnic groups (Japanese, Chinese, Asian Indian) for an allotypic polymorphism of a widely expressed low affinity Fc receptor for IgG (Fc gamma RIIA). The method, requiring PCR amplification of genomic DNA and Southern analysis with allele specific oligonucleotide probes, detects a single nucleotide difference (G or A) at base 494 which results in an arginine (R) or histidine (H) at amino acid 131 of the Fc gamma RIIA protein. This polymorphism has been shown ...

Absolute photoabsorption cross-sections at 195±3 K are reported for dinitrogen pentoxide, N2O5, between 150 and 240 nm. The spectrum consists of a broad intense band with a maximum at 160 nm. The lack of sharp features indicates that excitation of N2O5 results in dissociation over the whole of the spectral range studied. At wavelengths above 200 nm, our results are up to 30% smaller than the current NASA recommendation, but are in generally good agreement with a more recent measurement. Below 200 ...
