Abstract The amino acid derivative reactivity assay (ADRA) is an in chemico alternative to animal testing for skin sensitization that solves certain problems found in the use of the direct peptide reactivity assay (DPRA). During a recent validation study conducted at multiple laboratories as part of the process to include ADRA in an existing OECD test guideline, one of the nucleophilic reagents used in ADRA— N ‐(2‐(1‐naphthyl)acetyl)‐ l ‐cysteine (NAC)—was found to be susceptible to oxidation in much ...

Laser scattering signals that indicate the presence of small dust particles (diameter ⩽ 2 μm) have been occasionally observed in the JIPPT-IIU tokamak chamber. This phenomenon was reproduced deliberately by spreading carbon dust from the top of the vacuum chamber. No noticeable effect on the plasma was observed for dust falls of up to at least 106 particles (10 μg) in 20 ms during discharges. Dust falling just before plasma startup seemed to be confined but it was soon ejected (in less than 30 ms)

BACKGROUND It has been reported that the c-erbB-2 protooncogene is frequently amplified and overexpressed in many types of cancers, except sarcomas and hematological malignancies. METHODS Expression of ErbB-2 in the tumors of 26 patients with conventional osteosarcoma was evaluated by immunoblotting. DNA from osteosarcoma tissues that expressed ErbB-2 were analyzed by Southern blot hybridization to examine gross rearrangement of the gene. The DNA was also surveyed for the presence of genetic mutation ...

The current paradigm is that inflammatory pain passively resolves following the cessation of inflammation. Yet, in a substantial proportion of patients with inflammatory diseases, resolution of inflammation is not sufficient to resolve pain, resulting in chronic pain. Mechanistic insight into how inflammatory pain is resolved is lacking. Here, we show that macrophages actively control resolution of inflammatory pain remotely from the site of inflammation by transferring mitochondria to sensory neurons. ...

The influence of basal-plane dislocation (BPD) depth on the expansion of single Shockley-type stacking faults (1SSFs) was investigated during the forward-current degradation of 4H–SiC p–i–n diodes. The 1SSF expanded from the BPD converted into a threading edge dislocation (TED) in the substrate. The hole density that was injected into the substrate increased with the stress-current density; therefore, BPDs converted into TEDs at a greater depth caused the expansion of 1SSFs under higher stress-current ...

The origin of expanded single Shockley-type stacking faults in forward-current degradation of 4H-SiC p–i–n diodes was investigated by the stress-current test. At a stress-current density lower than 25 A cm−2, triangular stacking faults were formed from basal-plane dislocations in the epitaxial layer. At a stress-current density higher than 350 A cm−2, both triangular and long-zone-shaped stacking faults were formed from basal-plane dislocations that converted into threading edge dislocations near ...

Stacking faults expanded by the application of forward current to 4H-SiC p–i–n diodes were observed using a transmission electron microscope to investigate the expansion origin. It was experimentally confirmed that long-zonal-shaped stacking faults expanded from basal-plane dislocations converted into threading edge dislocations. In addition, stacking fault expansion clearly penetrated into the substrate to a greater depth than the dislocation conversion point. This downward expansion of stacking ...

The present study experimentally investigated the performance and emission characteristics of the diesel engine with hydrogen added to the intake air at late diesel-fuel injection timings. The diesel-fuel injection timing and the hydrogen fraction in the intake mixture were varied while the available heat produced by diesel-fuel and hydrogen per second of diesel fuel and hydrogen was kept constant at a certain value. NO showed minimum at specific hydrogen fraction. The maximum rate of incylinder ...

Abstract MCP1 is upregulated by various stimuli, including LPS, high glucose, and hyperosmolality. However, the molecular mechanisms of transcriptional regulation of the MCP1 gene under hyperosmolar conditions are poorly understood. Treatment of NRK52E cells with NaCl or mannitol resulted in significant elevation of MCP1 mRNA and protein in a time- and dose-dependent manner. Treatment with a p38MAPK inhibitor (SB203580), an ERK inhibitor (PD98059), or an MEK inhibitor (U0126), suppressed the increase ...

Granulocyte colony stimulating factor (G-CSF) has been shown to have neuroprotective and anti-inflammatory effects in cerebral damage models. In addition, bone-marrow-derived hematopoietic cells, which can be mobilized with G-CSF, have a neuroprotective effect in hereditary retinal cell death. The present study was conducted to investigate whether G-CSF protects photoreceptors from light-induced cell death.G-CSF or vehicle was systemically injected before the light exposure and for four consecutive ...

4H-SiC homoepitaxial layers were grown on on-axis (0001̄) C-face substrates at an inclination of 0.5° or less by horizontal hot wall chemical vapor deposition. After H2 etching, the surface morphology of the on-axis (0001̄) C-face substrate showed a straight step structure that had the same height as a 4H-SiC lattice constant along the c-axis. Specular surface morphology of a wide area of up to 80% of a 2-in wafer was obtained at a low C/Si ratio of 0.6. It was found that that appearance of basal ...

Additionally, the correlation between HPV-38 infection and expressions of proliferating cell nuclear antigens (PCNA) or p53 protein was analyzed immunohistochemically. To obtain information about the early changes of experimentally induced cerebral aneurysms in rats, the luminal surface of branching areas of their cerebral arteries was examined with a scanning electron microscope. At the branching sites of major cerebral arteries in the control animals, the intima just distal to the apex markedly protruded into the lumen forming a linear bank-like intimal pad. Along and distal to this pad, there was a shallow long groove (juxta-apical groove). Such grooves were ...

Liquid Telocity distribution was measured in a bubble column 5.5 m in diameter and approximately 7 m in liquid height. Liquid flow was upward in the column center and downward near the wall. The upward velocities were from 52 to 115 cm/sec in the superficial gas velocity range from 2.4 to 8.5 cm/sec. The liquid velocity distribution is analyzed by a circulating flow model. Values of the turbulent viscosity-the parameter of the model-are in the range from 3000 to 4000 g/cm • sec. These values agree ...

Factor VII is a vitamin K-dependent zymogen that plays a key role in the initiation of the extrinsic pathway. A severe factor VII deficiency was identified in a 45-year old male whose plasma factor VII antigen was less than 60 ng/ml and expressed 5.2% of normal factor VII activity. DNA sequence analysis of the patient's factor VII gene showed a thymidine to guanine transversion at nucleotide 10968 in exon VIII that results in a novel amino acid substitution of His348 to Gln. The patient was homozygous ...

Next generation risk assessment (NGRA) is an exposure-led, hypothesis-driven approach that has the potential to support animal-free safety decision-making. However, significant effort is needed to develop and test the in vitro and in silico (computational) approaches that underpin NGRA to enable confident application in a regulatory context. A workshop was held in Montreal in 2019 to discuss where effort needs to be focussed and to agree on the steps needed to ensure safety decisions made on cosmetic ...

According to the International Agency for Research on Cancer classification, formaldehyde is a human carcinogen that targets the nasal cavity. In humans and rats, inhaled formaldehyde is primarily deposited in the nasal cavity mucosa, metabolized to the less toxic formic acid, and finally excreted into the urine or exhaled. Thus, formaldehyde-induced nasal carcinogenicity may be a direct effect of formaldehyde itself, although the underlying mechanisms remain unclear. With regard to cytotoxicity, ...
