We have cloned a mammalian (rat) homologue of Drosophila ether á go-go (eag) cDNA, which encodes a distinct type of voltage activated potassium (K) channel. The derived Drosophila and rat eag polypeptides share > 670 amino acids, with a sequence identity of 61%, exhibiting a high degree of similarity at the N-terminus, the hydrophobic core including the pore forming P region and a potential cyclic nucleotide binding site. Rat eag mRNA is specifically expressed in the central nervous system. In the Xenopus ...

Constitutive heterochromatin is considered as a functionally inert genome compartment, important for its architecture and stability. How such stable structure is maintained is not well understood. Here, we apply four different visualization schemes to label it and investigate its dynamics during DNA replication and repair. We show that replisomes assemble over the heterochromatin in a temporally ordered manner. Furthermore, heterochromatin undergoes transient decompaction locally at the active sites ...

The deployment of measurement equipment in passenger aircraft for the observation of atmospheric trace constituents is described. The package of automated instruments that is installed in a one-ton-capacity aircraft freight container positioned in the forward cargo bay of a Boeing 767 ER can register a vast amount of atmospheric data during regular long-distance flights. The air inlet system that is mounted on the fuselage directly below the container comprises an aerosol inlet, a separate inlet ...

The German Library is a new series of the major works of German literature and thought from medieval times to the present. The volumes have forewords by internationally known writers and introductions by prominent scholars. Here the English-speaking reader can find the broadest possible collection of poetic and intellectual achievements in new as well as great classic translations. Convenient and accessible in format, the volumes of The German Library will form the core of any growing library of ...

Pseudomonas xanthomarina S11 is an arsenite-oxidizing bacterium isolated from an arsenic-contaminated former gold mine in Salsigne, France. This bacterium showed high resistance to arsenite and was able to oxidize arsenite to arsenate at concentrations up to 42.72 mM As[III]. The genome of this strain was sequenced and revealed the presence of three ars clusters. One of them is located on a plasmid and is organized as an “arsenic island” harbouring an aio operon and genes involved in phosphorous ...

Number concentrations of submicrometer aerosol particles at altitudes of 8.5–11.3 km were measured along a single flight route between Germany (∼50°N, ∼10°E) and the Indic (∼5°N, ∼80°E) over 3 years using a commercial aircraft platform (project Civil Aircraft for Regular Investigation of the Atmosphere Based on an Instrument Container (CARIBIC)). During 41 intercontinental flights (∼380 flight hours), more than one million individual particle concentration measurements were made, yielding a comprehensive ...

Simultaneous measurements of O 3 , CO, and ultrafine aerosol particles (UFP), conducted on board of a Boeing 767‐ER passenger aircraft flying from Sri Lanka to Germany (project CARIBIC), are used to study two‐way cross‐tropopause mixing near a subtropical tropopause fold. On the equatorward side of the fold, downward mixing of stratospheric air into the upper troposphere is identified by enhanced concentrations of O 3 and 14 CO. Very high UFP number concentrations of up to 1.5×10 4 cm −3 (STP) were encountered ...

Aside from the fully licensed herbal medicines there are products on the European pharmaceutical market which are registered by virtue of their longstanding traditional use. The normal registration procedure does not apply to them because presently they do not meet the legal requirements for a full license as set out in the relevant European Union Directive. One of these requirements, "proof of tradition", has so far been dealt with in different ways and fails to meet the criteria of good practice. ...

Methyl-CpG binding protein 2 (MECP2) is a protein that specifically binds methylated DNA, thus regulating transcription and chromatin organization. Mutations in the gene have been identified as the principal cause of Rett syndrome, a severe neurological disorder. Although the role of MECP2 has been extensively studied in nervous tissues, still very little is known about its function and cell type specific distribution in other tissues. Using immunostaining on tissue cryosections, we characterized ...

Rett syndrome is a neurological, X chromosomal-linked disorder associated with mutations in the MECP2 gene. MeCP2 protein has been proposed to play a role in transcriptional regulation as well as in chromatin architecture. Since MeCP2 mutant cells exhibit surprisingly mild changes in gene expression, we have now explored the possibility that Rett mutations may affect the ability of MeCP2 to bind and organize chromatin. We found that all but one of the 21 missense MeCP2 mutants analyzed accumulated ...

In situ trace gas (O 3 , NO, NO 2 , NO y ) and ultrafine aerosol particle data from a passenger aircraft (project CARIBIC) and the NOAA WP‐3D research aircraft are used to identify the origin of “ozone‐rich transients” that occasionally appear in aircraft ozone data sets along flight distances of 5–80 km. Evidence of ozone import from the stratosphere, once suggested as the most likely cause, cannot be found. Our data rather reveal that the majority of the recorded ozone transients are artifacts caused by ...

Rat eag potassium channels (r‐eag) were expressed in Xenopus oocytes. They gave rise to delayed rectifying K + currents with a strong Cole‐Moore effect. Deletions in the N‐terminal structure of r‐eag either shifted the activation threshold to more negative potentials and slowed the activation kinetics (Δ2–190, Δ2–12 and Δ7–12) or resulted in a shift to more positive potentials and faster activation kinetics (Δ150–162). The impact of the deletion Δ7–12 was investigated in more detail: it almost abolished ...

BACKGROUND Abiraterone Acetate (AA) represents a highly effective androgen-receptor (AR) axis targeted agent. Treatment with AA in castration-resistant prostate cancer (CRPC) may partly mediate neuroendocrine differentiation (NED) as an escape mechanism, which may have implications for the choice of sequential therapy in CRPC. We evaluated how treatment with AA influences circulating neuromediators chromogranin A (CGA), neuron-specific enolase (NSE), and pro-gastrin-releasing peptide (Pro-GRP) in chemotherapy- ...

Methyl CpG binding protein 2 (MeCP2) binds DNA, and has a preference for methylated CpGs and, hence, in cells, it accumulates in heterochromatin. Even though it is expressed ubiquitously MeCP2 is particularly important during neuronal maturation. This is underscored by the fact that in Rett syndrome, a neurological disease, 80% of patients carry a mutation in the MECP2 gene. Since the MECP2 gene lies on the X chromosome and is subjected to X chromosome inactivation, affected patients are usually ...

Sudden cardiac death due to out-of-hospital cardiac arrest is the third leading cause of mortality in industrialised nations.1–3 In Europe and in the USA, 800 000 people die every year despite the fact that the emergency medical services initially resuscitate them. The main problem is that emergency medical services take an average of 8 min – and often much more – before they arrive at the scene. The brain tissue, however, commences to die after 3 to 5 min. Therefore, resuscitation procedures initiated ...

Aim: Aim of the study was to detect small cell/neuroendocrine (SCNC) transformation in metastatic castration-resistant prostate cancer (mCRPC) that is a challenging procedure. We investigated the role of neuromediator dynamics as potential evidence of SCNC in patients undergoing docetaxel therapy. Patients and Methods: A multi-institutional, prospective observational study was conducted. Patients undergoing docetaxel treatment were included.
