Manfred Renz, Peter Nehls, John Hozier,
There now seems to be little doubt that DNA in the nucleus of eukaryotic cells is in the form of very long, continuous molecules, probably thousands of times as long as the largest dimension of the nucleus. All of this DNA must be folded in such a way that it fits within the interphase nucleus and yet maintains its functional capacity. Since the degree of condensation is so large, there are most likely several “levels” of folding of the DNA molecule. The two basic levels of folding — the formation ...

Manfred Renz, Peter Nehls, John Hozier,
At high ionic strength (e.g., physiological salt concentrations) chromosome fibers are 200 A in diameter and composed of discrete globular structures that are held together by histone H1. At low ionic strength the fibers unfold and appear as the familiar chains of nucleosomes (80 A in diameter). The unfolding of chromosome fibers occurs within a narrow salt range. It results from a change in the mode of the interaction between histone H1 and the chromosome fiber and is very likely the consequence ...

DNA lengths in the structural repeat units of Chinese hamster ovary (CHO) and chicken erythrocyte chromatin were compared by analyzing the sizes of DNA fragments produced after treatment of nuclei with staphylococcal nuclease. The repeat length of CHO chromatin (173 ±4 BP) is about 20 base pairs (BP) smaller than that of chicken erythrocyte chromatin (194 ± 8 BP). Repeat lengths of rat liver and calf thymus chromatin were found to be about 10 BP shorter than that of chicken erythrocyte chromatin. Thus ...

The time course of the fragmentation of calf thymus chromatin by DNase II (deoxyribonucleate 3'-oligonucleotidohydrolase, EC 3.1.4.6) has been examined by sedimentation of chromatin digests through linear (5-20%) sucrose gradients. The action of nuclease is decidedly nonrandom, ultimately producing roughly equal amounts of acid-soluble oligonucleotides and 11S nucleoprotein particles. The 11S particles contain double-stranded DNA that is approximately 400 A or 120 base-pairs long, as measured by electron ...

Digestion of calf thymus chromatin with micrococcal nuclease produces a mixture of apparently well defined nucleoprotein fragments which have been partially resolved by sedimentation on linear (5-20%) sucrose gradients. Sedimentation patterns reveal a predominant peak at the 11S position, three slower components, which have not previously been reported, at the 3.4S, 5.3S and 8.6S positions, and three faster components at the 17S, 22S and 26S positions. DNA isolated from the 3S to 12S region of gradients ...

John Hozier, J. Herbert Taylor,
The distribution of lengths of single-strand DNA in Chinese hamster ovary cells in the G1 phase of the cell cycle has been observed for various conditions of cell lysis and incubation of the lysates. The method of analysis was band sedimentation through a self-generating density gradient, a technique developed originally for the analytical ultracentrifuge, but modified here for the preparative ultracentrifuge so that measurements of sedimentation coefficients could be made under conditions that ...
