Production of a 92-kDa gelatinase / type IV collagenase and tissue inhibitor of metalloproteinases (TIMP) was investigated with human sarcoma cell lines. Among the cytokines and growth factors examined, only human recombinant tumor necrosis factor α (TNFα) induced and stimulated the proteinase with concomitant increase in TIMP expression, but matrix metalloproteinase 2 (72-kDa gelatinase / type IV collagenase) expression was unchanged. These data suggest that gene expression of the two metalloproteinases ...

Metalloproteinases produced by connective tissue cells may play a key part in the destruction of joints in rheumatoid arthritis. Matrix metalloproteinase 3 (MMP-3; stromelysin) capable of degrading cartilage proteoglycans and type IX collagen and of activating procollagenase was immunolocalised in hyperplastic synovial lining cells in rheumatoid synovium, but not in the cells of normal synovium. Cells responsible for synthesis of MMP-3 have the phenotype of synovioblasts (B cells) by immunoelectron ...

Matrix metalloproteinase 9 (MMP-9) has been purified as an inactive zymogen of M(r) 92,000 (proMMP-9) from the culture medium of HT 1080 human fibrosarcoma cells. The NH2-terminal sequence of proMMP-9 is Ala-Pro-Arg-Gln-Arg-Gln-Ser-Thr-Leu-Val-Leu-Phe-Pro, which is identical to that of the 92-kDa type IV collagenase/gelatinase. The zymogen can be activated by 4-aminophenylmercuric acetate, yielding an intermediate form of M(r) 83,000 and an active species of M(r) 67,000, the second of which has a ...

Tea polyphenols (flavan-3-ol derivatives) suppressed the oxidative modification of low density lipoprotein (LDL) which is assumed to be an important step in the pathogenesis of atherosclerosis lesions. Inhibitory experiments on the oxidative impairment of porcine serum LDL by flavan-3-ols were carried out by incubating them at 37 degrees C in the presence of 5 microM Cu2+. The oxidation of LDL was monitored either by an absorption increase at 234 nm due to the conjugated diene formation, or the formation ...

Perforation is a major complication of endoscopic submucosal dissection (ESD) for early gastric cancer (EGC). However, there have been no reports on delayed perforation after ESD for EGC. We aimed to elucidate the incidence and outcomes of delayed perforation after ESD. Clinical courses in 1159 consecutive patients with 1329 EGCs who underwent ESD were investigated. Delayed perforation occurred in six patients (0.45 %). All these patients had complete en bloc resection without intraoperative perforation ...

From a practical point of view, some measures to reduce the thickness effect backed by a reasonable criterion are required for fabricating structures with heavy section plates. In this study, the thickness effect was investigated by systematic experiments on welded steel joints with thicknesses ranging from 10 to 80 mm. Cruciform joints and T-joints with improved weld by overall profiling or toe-grinding were tested under pulsating tension and under pulsating bending, respectively. These experimental ...

We have found extremely low efficiency of the elastomeric properties for SEBS (polystyrene-block-poly(ethylene-co-butylene)-block-polystyrene) triblock copolymers having short polystyrene (PS) block chains. Since the SEBS specimens form spherical PS microdomains embedded in the matrix of the rubbery poly(ethylene-co-butylene) (PEB) chains, they exhibit elastomeric properties (thermoplastic elastomer film). However, it was found that the stress was dramatically decreased with time when the specimens ...

There is evidence that abnormal formation of the sympathetic nervous system might be important for pathogenesis of hypertension. In the present study we analyzed nucleotide sequences of low affinity nerve growth factor receptor (LNGFR) genes in the spontaneously hypertensive rat and its stroke-prone substrain. There was a point mutation generating an amino acid substitution in signal peptide of these LNGFRs. This result suggested that the mutated LNGFR gene is one of the promising candidate genes ...

We have reported in our previous paper that several flavan-3-ol derivatives (tea polyphenols) inhibited the Cu2+-mediated low density lipoprotein (LDL) oxidation in vitro. (-)-Epigallocatechin gallate (EGCG), in particular, exhibited strong inhibition. In this study, we have compared the antioxidative effects of EGCG with those of other natural antioxidants, such as flavonols, sesaminol, curcuminoid derivatives, tocopherol analogues, and theaflavins. The antioxidative effects were monitored by conjugated ...

Type II collagen, a main structural ...

Premenopausal women are at a lower risk of coronary heart diseases relative to age matched men. However, the underlying mechanisms are not clearly understood. This article studies the effects of 17β-estradiol (17β-E2) at physiological concentrations on the cholesterylester metabolism in macrophages (J774 A.1 cells) with a particular focus on neutral cholesterol esterase (N-CEase). Cells were incubated with β-VLDL, [1-14C]oleic acid and 17β-E2 (0.25 and 2.5 nM). 17β-E2 dose-dependently reduced cholesteryl-[ ...

The decay of 56Ni is investigated to study lower excited states in the 56Co nucleus. From measurements of internal conversion electrons and angular-correlation functions for all coincident gamma-ray pairs, unique spin and parity assignments are obtained as follows: 163 keV level, 3+; 975 keV level, 2+; 1.45 MeV level, 1(2)−; 1.72 MeV level, 1+. Multipole mixtures of transitions between these levels are also determined. Results are discussed on the basis of the jj coupling shell model with p-n interactions.

Abstract A new ion exchanger, crystalline zirconium titanium phosphate with variable zirconium to titanium mole ratios has been prepared and the sodium ion exchange behavior studied. In the zirconium-rich exchanger a half-exchanged monosodium phase was produced. This differs from the titanium-rich exchanger where a disodium phase was formed without formation of the intermediate monosodium phase. The pH titration curves were complicated but a trend emerged with variation in the Zr/Ti ratio. On the ...

Forward and backward pH titration curves were measured for potassium-ion exchange on crystalline α-hafnium phosphate which had been prepared by the direct-precipitation method. Exchange in each direction proceeded in two steps, and the hysteresis was observed in the region of smaller loadings of the potassium ions. Exchange in the ternary H+–Li+–K+ system was also preliminarily investigated, the selectivity of the potassium and lithium ions was what would be expected from the results of the binary ...

2-Amino-3,4,8-trimethylimidazo[4,5-f]quinoxaline (4,8-DiMeIQx) was found in bacteriological-grade beef extract by liquid chromatographies with electrochemical detection and a photodiode array detector. 4,8-DiMeIQx was estimated to be present at a level of 10.0 ng per g of beef extract, and to account for 9% of the total mutagenicity of the extract.

A simple and sensitive method was developed for quantification of mutagenic/carcinogenic aminoimidazoquinoline and aminoimidazoquinoxaline compounds in heated materials. Samples were partially purified by blue-cotton treatment, 0.1 N HCl-methylene dichloride partition and separation in a SEP-PAK silica cartridge. The recoveries of aminoimidazoquinoline and aminoimidazoquinoxaline compounds at the step of partial purification were estimated by spiking with 14C-labeled compounds. The compounds in ...

A croaker (Nibea mitsukurii) has a high incidence of the skin melanoma, chromatophoroma, in a Pacific coastal area in Japan. A sea catfish (Plotosus anguillaris) bearing skin melanosis is also found in the same area. For elucidation of a correlation between these pigment cell neoplasms of the skin and environmental contaminants, an epidemiological survey was conducted to determine the distribution and prevalence of tumor-bearing fish. Based upon observations of a high prevalence of skin neoplasms ...
