A previously unannotated, putative fliK gene was identified in the Campylobacter jejuni genome based on sequence analysis; deletion mutants in this gene had a 'polyhook' phenotype characteristic of fliK mutants in other genera. The mutants greatly overexpressed the sigma(54)-dependent flagellar hook protein FlgE, to form unusual filamentous structures resembling straight flagella in addition to polyhooks. The genome sequence reveals only one gene predicted to encode an orthologue of the NtrC-family ...

Summary Consistent with its role as a nitric oxide (NO)‐detoxifying globin in Campylobacter jejuni , Cgb ( Campylobacter globin) expression is strongly and specifically induced following exposure to nitrosative stress, suggesting a previously unrecognized capacity for NO‐related stress sensing in this food‐borne pathogen. In this study, Fur and PerR have been eliminated as major regulators of cgb , and NssR (Cj0466), a member of the Crp‐Fnr superfamily, has been identified as the major positive regulatory ...

ABSTRACT Explorative approaches such as DNA microarray experiments are becoming increasingly important in microbial research. Despite these major technical advancements, approaches to study multifactor experiments are still lacking. We have addressed this problem by using rotation testing and a novel multivariate analysis of variance (MANOVA) approach (50-50 MANOVA) to investigate interacting experimental factors in a complex experimental design. Furthermore, a new rotation testing based method was ...

The quality of bioanalytical data is dependent upon selective, sensitive, and reproducible analytical methods. With evolving technologies available, bioanalytical scientists must assess which is most appropriate for their molecule through proper method validation. For an early stage PEGylated insulin program, the characteristics of four platforms, ELISA, ECL, Gyrolab, and LC–MS/MS, were evaluated using fit-for-purpose method development and validation, while also evaluating costs. Methods selected ...

A survey was undertaken to determine the number of steps taken by individuals during their normal activities of daily life. Shortcomings in previous survey techniques led to the design and construction of a. small, battery-powered, electromagnetic counter. This complete unit, including battery, weighs less than five ounces arid measures 2-75 X 1’6 X 1 in. It is capable of recording 75,000 counts during the life of one battery. Details of this instrument and the plantar switch in the shoe are described. ...

Madelyn G. Sorrentino, Taylor Stewart, J. Ellen Marsden, Jason D. Stockwell,

After 45 years of stocking, lake trout in Lake Champlain have started to exhibit strong natural recruitment, suggesting a recent change in limiting factors such as prey availability or overwinter survival. The abundance of juvenile wild lake trout varies among regions of Lake Champlain which suggests the prey base, or foraging success, may vary geographically within the lake. One metric that can indicate differences in resources across regions is lake trout lipid content, which reflects the availability ...

John Janssen, J. Ellen Marsden, Thomas R. Hrabik, Jason D. Stockwell,

Abstract Hjort's insights on marine fish recruitment and larval fish advection are presumed not applicable to freshwater lakes because most freshwater lakes are small. The Laurentian Great Lakes (LGL), however, are large enough for certain oceanic-type hydrodynamics, such as strong currents and upwelling, to affect the distribution and survival of larval fish and thus fall under Hjort's purview. However, there are evolutionary constraints because LGL species underwent an evolutionary bottleneck during ...

Jason Gush, Adam B. Jaffe, Victoria Larsen, Athene Laws,

We estimate the impact of the NZ Marsden Fund on research success, by comparing the subsequent performance of funded and unfunded researchers. We control for selection bias using the Marsden panel rankings. We find that funding is associated with a 6%–15% increase in publications and an 11%–22% increase in citation-weighted papers for research teams. We attempt to pin down the dynamic structure of this effect by looking at the research trajectories of individual researchers, but this model yields ...

Kenneth Tran, Kyle Feliciano, Weiguang Yang, Erica L. Schwarz, Alison L. Marsden, Ronald L. Dalman, Jason T. Lee,

The durability of fenestrated endovascular aneurysm repair (fEVAR) has been threatened by thrombotic complications. In the present study, we used patient-specific computational fluid dynamic (CFD) simulation to investigate the effect of the endograft diameter on hemodynamics after fEVAR and explore the hypothesis that diameter-dependent alterations in aortic hemodynamics can predict for thrombotic events. A single-institutional retrospective study was performed of patients who had undergone fEVAR ...

ABSTRACT We previously identified and characterized a two-component regulatory system in the meningococcus with homology to the phoP-phoQ system in salmonella and showed that allele replacement of the NMB0595 regulator gene led to loss of virulence, sensitivity to antimicrobial peptides, perturbed protein expression, and magnesium-sensitive growth. On the basis of these findings we proposed that the system should be designated the meningococcal PhoPQ system. Here we further characterized the NMB0595 ...
