The complexity inherent in biological systems challenges efforts to rationally engineer novel phenotypes, especially those not amenable to high-throughput screens and selections. In nature, increased mutation rates generate diversity in a population that can lead to the evolution of new phenotypes. Here we construct an adaptive control system that increases the mutation rate in order to generate diversity in the population, and decreases the mutation rate as the concentration of a target metabolite ...

This study examines the dynamic co-localization of lipid-anchored fluorescent proteins in living cells using pulsed-interleaved excitation fluorescence cross-correlation spectroscopy (PIE-FCCS) and fluorescence lifetime analysis. Specifically, we look at the pairwise co-localization of anchors from lymphocyte cell kinase (LCK: myristoyl, palmitoyl, palmitoyl), RhoA (geranylgeranyl), and K-Ras (farnesyl) proteins in different cell types. In Jurkat cells, a density-dependent increase in cross-correlation ...

Synthetic biological pathways could enhance the development of novel processes to produce chemicals from renewable resources. On the basis of models that describe the evolution of metabolic pathways and enzymes in nature, we developed a framework to rationally identify enzymes able to catalyze reactions on new substrates that overcomes one of the major bottlenecks in the assembly of a synthetic biological pathway. We verified the framework by implementing a pathway with two novel enzymatic reactions ...

The performance of bilayer heterojunction, pentacene and perylene tetracarboxylic diimide organic solar cells (OSC) is dramatically enhanced by performing nanoimprinting on the hole transport layer in which poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) nanogratings are built. A standard OSC embeded with PEDOT:PSS gratings enables photocarriers to move toward electrodes. In particular, the visualization of morphologies for these heterojunction layers reveals pillar-like grains ...

Background: The aim of lower extremity reconstruction has focused on early wound coverage and functional recovery but rarely aesthetics. Free muscle flaps provide durable coverage; however, they require skin graft coverage and result in muscle atrophy limiting future revisions. Perforator-based flap reconstructions can be easily elevated to allow for both orthopedic and contouring procedures. The authors reviewed the role of secondary procedures in achieving improved functional and aesthetic results ...

C-C chemokine receptor-1 (CCR1) has been implicated in mediating a variety of inflammatory conditions including multiple sclerosis and organ rejection. Although originally referred to as the MIP-1alpha/RANTES receptor, CCR1 is quite promiscuous and can be activated by numerous chemokines. We used radioligand binding and [35S]-GTPgammaS exchange assays in membranes from a cell line transfected to express CCR1 (Ba/F3-hCCR1) to characterize a panel of chemokines (HCC-1, MIP-1alpha, MIP-1beta, MIP-1delta, ...

The efficiency of small-molecule solar cells critically depends on the match of the junction of the donor and acceptor semiconductors used in these devices to create charged carriers and on the mobility of individual components to transport holes and electrons. In the present study, a 2% efficient bilayer organic solar cell consisting of a p-type semiconductor, pentacene, and an n-type semiconductor, N,N′-diheptyl-3,4,9,10-perylenetetracarboxylic diimide (PTCDI-C7), is fabricated.
