This paper presents the construction of amperometric biosensors for the highly sensitive detection of carbamate insecticides based on the inhibition of acetylcholinesterase (AChE). This enzyme was immobilised by entrapment in an optimised sol–gel matrix on TCNQ-modified screen-printed electrodes. The enzyme activity was estimated by measuring the thiocholine produced by the enzymatic hydrolysis of the acetylthiocholine using TCNQ as mediator. Wild and genetically engineered AChEs from Drosophila ...

This work presents an automatic method able to identify the presence of neurotoxic insecticides using omethoate as a model compound. The acetylcholinesterase (AChE)-based biosensors used to detect the neurotoxic insecticides present two shortcomings: the weak limit of detection (LOD) and the lack of selectivity. An appropriate LOD is obtained in our work by using two sensitive AChEs from Drosophila melanogaster (Dm): the wild type (wt-Dm) and the E69W mutant. To discriminate between the insecticides ...

Development of new and more reliable methods to immobilise biomolecules has emerged rapidly due to a continuous need for more stable, sensitive and reliable biosensors. This paper reports a new method of acetylcholine-esterase (AChE) immobilisation based on the high affinity interaction between the glycoproteic enzyme and Concanavalin A (Con A). In order to establish the nature of the link formed between the glycoenzyme, lectin and support, three different configurations are presented. The optimum ...

We report two cases of aneurysm of the distal branches of the superior cerebellar artery presenting with cerebellar infarction. In both cases, the diagnosis required close correlation of the findings from different imaging techniques, as catheter angiography failed to opacify the lesions. In one patient endovascular parent vessel occlusion was performed, whereas in the second patient the aneurysm thrombosed spontaneously. We describe the clinical and radiologic presentation of these aneurysms and ...

Abstract Classifications of acute myeloid leukemia (AML) patients rely on morphologic, cytogenetic, and molecular features. Here we have established a novel flow cytometry-based immunophenotypic stratification showing that AML blasts are blocked at specific stages of differentiation where features of normal myelopoiesis are preserved. Six stages of leukemia differentiation-arrest categories based on CD34, CD117, CD13, CD33, MPO, and HLA-DR expression were identified in two independent cohorts of ...

Jean Danet,

Flavescence dorée (FD) is a European quarantine grapevine disease transmitted by the Deltocephalinae leafhopper Scaphoideus titanus. Whereas this vector had been introduced from North America, the possible European origin of FD phytoplasma needed to be challenged and correlated with ecological and genetic drivers of FD emergence. For that purpose, a survey of genetic diversity of these phytoplasmas in grapevines, S. titanus, black alders, alder leafhoppers and clematis were conducted in five European ...

The etiology and main pathways for the spread of lavender decline, an infectious disease affecting French lavender production since the 1960s, have remained unclear, hampering the development of efficient control strategies. An extensive survey of lavender fields led to the conclusion that “ Candidatus Phytoplasma solani” was chronically infecting declining lavenders and was associated with large infectious populations of Hyalesthes obsoletus planthoppers living on the crop itself. Lavender appeared ...

ABSTRACT Phytoplasmas are uncultivated plant pathogens and cell wall-less bacteria and are transmitted from plant to plant by hemipteran insects. The phytoplasma's circulative propagative cycle in insects requires the crossing of the midgut and salivary glands, and primary adhesion to cells is an initial step toward the invasion process. The flavescence dorée (FD) phytoplasma possesses a set of variable membrane proteins (Vmps) exposed on its surface, and this pathogen is suspected to interact with ...

We report the dual-frequency and dual-polarization emission of an optically-pumped vertical external-cavity semiconductor laser. Our laser source provides a high-purity optically-carried RF signal tunable in the GHz range, and is specifically designed for the coherent population trapping (CPT) of Cs atoms in compact atomic clocks. The laser spectrum is stabilized onto a Cs atomic transition at 852.1 nm, and the frequency difference is locked to a local oscillator at 9.2 GHz. Special attention has ...

Toward the next generations of compact atomic clocks, clocks based on coherent population trapping (CPT) offer a very interesting alternative. Thanks to CPT, a quantum interfering process, this technology has made a decisive step in the miniaturization direction. Fractional frequency stability of 1.5x10-10 at 1 s has been demonstrated in commercial devices of a few cm3. The laboratory prototype presented here intends to explore what could be the ultimate stability of a CPT based device. To do so, ...

The Dick effect can be a limitation of the achievable frequency stability of a passive atomic frequency standard when the ancillary frequency source is only periodically sampled. Here, we analyze the Dick effect for a pulsed vapor cell clock using coherent population trapping (CPT). Because of its specific interrogation process without atomic preparation nor detection outside of the Ramsey pulses, it exhibits an original shape of the sensitivity function to phase noise of the oscillator. Numerical ...

We report the development, absolute phase noise, and residual phase noise characterization of a 9.192 GHz microwave frequency synthesis chain devoted to be used as a local oscillator in a high-performance cesium vapor cell atomic clock based on coherent population trapping (CPT). It is based on frequency multiplication of an ultra-low phase noise 100 MHz oven-controlled quartz crystal oscillator using a nonlinear transmission line-based chain. Absolute phase noise performances of the 9.192 GHz output ...

We propose a constructive polarization modulation scheme for atomic clocks based on coherent population trapping (CPT). In this scheme, the polarization of a bichromatic laser beam is modulated between two opposite circular polarizations to avoid trapping the atomic populations in the extreme Zeeman sublevels. We show that if an appropriate phase modulation between the two optical components of the bichromatic laser is applied synchronously, the two CPT dark states which are produced successively ...

Vapor cell atomic clocks exhibit reduced frequency stability for averaging time between about one hundred and a few thousand seconds. Here we report a study on the impact of the main parameters on the mid-to-long term instability of a buffer-gas vapor cell Cs clock, based on coherent population trapping (CPT). The CPT signal is observed on the Cs D1 line transmission, using a double $\Lambda$ scheme and a Ramsey interrogation technique. The effects on the clock frequency of the magnetic field, the ...

The genetic diversity of three temperate fruit tree phytoplasmas 'Candidatus Phytoplasma prunorum', 'Ca. P. mali' and 'Ca. P. pyri' has been established by multilocus sequence analysis. Among the four genetic loci used, the genes imp and aceF distinguished 30 and 24 genotypes, respectively, and showed the highest variability. Percentage of substitution for imp ranged from 50 to 68 % according to species. Percentage of substitution varied between 9 and 12 % for aceF, whereas it was between 5 and 6 % for pnp ...

ABSTRACT Spiroplasma citri GII3 contains highly related low-copy-number plasmids pSci1 to -6. Despite the strong similarities between their replication regions, these plasmids coexist in the spiroplasma cells, indicating that they are mutually compatible. The pSci1 to -6 plasmids encode the membrane proteins known as S. citri adhesion-related proteins (ScARPs) (pSci1 to -5) and the hydrophilic protein P32 (pSci6), which had been tentatively associated with insect transmission, as they were not detected ...

Surface proteins play an important role in phytoplasma life cycle. The antigenic membrane protein (AMP) of ‘Candidatus Phytoplasma asteris’ has been shown to interact with the insect microfilaments. Due to the synteny of the groL-amp-nadE genetic locus between phytoplasma genomes, the gene stamp that encodes the antigenic membrane protein of stolbur phytoplasma has been cloned and characterized. It encodes a 157 amino acid-long protein with a predicted signal peptide and a C-terminal hydrophobic alpha- ...

The insect-transmissible strain GII-3 of Spiroplasma citri contains plasmids pSci1–6, five of which (pSci1–5) encode adhesin-like proteins and one (pSci6) encodes protein P32, which has been associated with insect transmissibility. In contrast, S. citri strains ASP-1 and 44, which cannot be transmitted via injection into the leafhopper vector Circulifer haematoceps , lack these proteins and also do not carry plasmids pSci1–6. To further study the apparent relationship between the presence of plasmids ...

Phytoplasmas are sieve tube restricted, wall-less phytopathogenic bacteria belonging to the class Mollicutes that have resisted cultivation. In previous studies we have shown that single-chain Fv antibodies (scFv) directed against an immunodominant membrane protein of the culturable plant pathogenic mollicute Spiroplasma citri inhibit its growth in vitro. Based on these observations, we considered a plantibody-based strategy for controlling phytoplasmas in plants. Different genetic constructs, in ...
