Concanavalin A-induced proliferation of rat T-lymphocytes is completely inhibited by 10(-5) M pyrazofurin, a potent inhibitor of pyrimidine de novo synthesis, as judged by cell viability and [3H]thymidine incorporation. Proliferation is completely restored by 5 X 10(-5) M uridine. 5′-Deoxy-5′-methylthioadenosine inhibits Concanavalin A induced rat T lymphocyte proliferation in a dose dependent manner (50 μM to 1000 μM). The extent of inhibition by MTA of lymphocyte proliferation was greatest when MTA was added to the cells at the same time as Concanavalin A. Vinylphosphine (1) has been synthesized from diethyl vinylphosphonate in three steps including a retro cycloaddition reaction using flash vacuum pyrolysis (overall yield ∼30%) and characterized by mass, i.r., and n.m.r. spectroscopy; it exhibits a remarkable stability at room temperature in neutral solution.

The intermediary metabolism of pyrimidine nucleosides was studied in a line of human B lymphoblasts (Raji) in which pyrimidine de novo synthesis deficiency was pharmacologically induced by pyrazofurin. It was found that Raji cells are cytidine deaminase deficient that cytidine has a synergistic effect on the toxicity of pyrazofurin towards these cytidine deaminase deficient cells, affecting both the proliferation and the viability of the cells. We studied the effect of staurosporine, a potent inhibitor of protein kinase C (PKC) activity, on the phorbol ester- or monoclonal antibody (mAb)-induced modulation of CD3 and CD4 surface antigens. Staurosporine (10(-5) M) completely inhibited phorbol ester-induced modulation but had no effect on that induced by mAb.
