The Apicomplexan parasite, Toxoplasma gondii, is an obligate intracellular organism that must co-opt its host cell to survive. To this end, Toxoplasma parasites introduce a suite of effector proteins from two secretory compartments called rhoptries and dense granules into the host cells. Once inside, these effectors extensively modify the host cell to facilitate parasite penetration, replication and persistence. In this review, we summarize the most recent advances in current understanding of effector ...

RNA contains over 100 modified nucleotides that are created post-transcriptionally, among which pseudouridine (Ψ) is one of the most abundant. Although it was one of the first modifications discovered, the biological role of this modification is still not fully understood. Recently, we reported that a pseudouridine synthase (TgPUS1) is necessary for differentiation of the single-celled eukaryotic parasite Toxoplasma gondii from active to chronic infection. To better understand the biological role ...

New experimental data for gibbsite growth on powdered kaolinite and single crystal muscovite and published data for gibbsite growth on gibbsite powders at 80°C in pH3 solutions show that all growth rates obey the same linear function of saturation state provided that reactive surface area is evaluated for each mineral substrate. Growth rate (mol m−2 s−1) is expressed by Rateppt = (1.9 ± 0.2) × 10−10|ΔGr|/RT(0.90±0.01), which applies to the range of saturation states from ΔGr = 0 to 8.8 kJ mol−1, where ΔGr = ...

William Fairbrother and colleagues use a massively parallel splicing assay (MaPSy) to analyze 4,964 exonic, disease-causing mutations for splicing defects in vivo and in vitro. They find that 10% of these exonic mutations affect splicing, and they classify these alterations by the stage of spliceosome assembly that is disrupted. The lack of tools to identify causative variants from sequencing data greatly limits the promise of precision medicine. Previous studies suggest that one-third of disease- ...

Many such effectors must be translocated across the parasitophorous vacuole (PV), in which the parasites replicate, ultimately ending up in the host cytosol or nucleus. This translocation has previously been shown to be dependent on five parasite proteins: MYR1, MYR2, MYR3, ROP17, and ASP5. We report here the identification of several MYR1-interacting and novel PV-localized proteins ...

Abstract Intraperitoneal injection of 10 µg of egg albumin (EA) or keyhole limpet hemocyanin plus alum elicited a low titer reagin response in most male Sprague-Dawley rats. Infection with Nippostrongylus brasiliensis larvae served to potentiate the reagin response in many animals. The potentiated response yielded impressive reagin titers, at least in rats injected with EA, but was of short duration. The potentiated response appeared to be restricted to antibodies of the IgE class and did not involve ...
