Abstract Quenching of fluorescence of highly purified dihydrofolate reductase from a methotrexate-resistant subline of the L1210 lymphoma by substrates and inhibitors was used to determine dissociation constants between pH 5 and pH 9. At pH 7.0, the dissociation constant of dihydrofolate from the enzyme-dihydrofolate complex was 1.2 x 10-7 m, 10 times greater than that of NADPH and several times less than that of folate at the same pH. Cells of human origin in suspension culture increased their dihydrofolate reductase activity during logarithmic growth. High concentrations of folic acid caused a further increase in this activity. The effect of folic acid seemed specific since general protein synthesis and the activity of three other enzymes were not altered in its presence. Abstract Dihydrofolate reductase has been purified 1000-fold from a methotrexate-resistant subline of the L1210 lymphoma with 49% yield and purity up to 100%. The procedure involves ammonium sulfate and acid precipitation, gel filtration on Sephadex G-100, and chromatography on hydroxylapatite. Purity has been determined by titrating enzymic activity and protein fluorescence with a stoichiometric inhibitor, methotrexate, and confirmed by electrophoresis and immunodiffusion.
