Significance Coronary artery disease (CAD) is a major cause of death and disability. Genetically modified SR-BI/apoE double KO (dKO) mice spontaneously exhibit many features of human CAD, including hypercholesterolemia, clogged arteries, myocardial infarction (MI) (or heart attack), heart failure, and premature death. We identified many changes in gene expression in dKO hearts [e.g., increases in apolipoprotein D (apoD)] during CAD development and compared them to those occurring after surgically induced ...

Huntington's Disease-Like 2 (HDL2), caused by a CTG/CAG expansion in JPH3 on chromosome 16q24, is the most common Huntington's Disease (HD) phenocopy in populations with African ancestry. Qualitatively, brain MRIs of HDL2 patients have been indistinguishable from HD. To determine brain regions most affected in HDL2 a cross-sectional study using MRI brain volumetry was undertaken to compare the brains of nine HDL2, 11 HD and nine age matched control participants. Participants were ascertained from ...

Studies to identify predictive biomarkers can be carried out in isogenic cancer cell lines, which enable interrogation of the effect of a specific mutation. We assessed the effects of four drugs, the PI3K–mammalian target of rapamycin inhibitor dactolisib, the PI3K inhibitor pictrelisib, and the MEK (MAPK/ERK Kinase) inhibitors PD 0325901 and selumetinib, in isogenic DLD1 parental, KRAS+/−, KRASG13D/−, PIK3CA+/− and PIK3CAE545K/− colorectal carcinoma cell lines. Importantly, we found substantial ...

Abstract The African wild dog is endemic to sub‐Saharan Africa and belongs to the family Canidae which includes domestic dogs and their closest relatives (i.e., wolves, coyotes, jackals, dingoes, and foxes). The African wild dog is known for its highly social behavior, co‐ordinated pack predation, and striking vocal repertoire, but little is known about its brain and whether it differs in any significant way from that of other canids. We employed gross anatomical observation, magnetic resonance imaging, ...

Over 15 million patients with epilepsy worldwide do not respond to drugs. Successful surgical treatment requires complete removal or disconnection of the seizure onset zone (SOZ), brain region(s) where seizures originate. Unfortunately, surgical success rates vary between 30 and 70% because no clinically validated biological marker of the SOZ exists. We develop and retrospectively validate a new electroencephalogram (EEG) marker—neural fragility—in a retrospective analysis of 91 patients by using ...

Background Huntington's Disease-Like 2 (HDL2) is one of the most common Huntington's Disease (HD) phenocopies. Clinical similarities between these genetically different diseases are remarkable and the MRIs between them have been indistinguishable. Objective To compare MRIs between HDL2 and HD against unaffected controls using semiautomated volumetric analysis and morphometry. Methods Participants with a mixed or African ancestry were enrolled from an area with the world's highest frequency of HDL2. All MRIs ...
