Abstract Inosine-5′-monophosphate dehydrogenase (IMPDH) plays key roles in purine nucleotide metabolism and cell proliferation. Although IMPDH is a widely studied therapeutic target, there is limited information about its physiological regulation. Using Ashbya gossypii as a model, we describe the molecular mechanism and the structural basis for the allosteric regulation of IMPDH by guanine nucleotides. We report that GTP and GDP bind to the regulatory Bateman domain, inducing octamers with compromised ...

Native tubulin αβ dimers and microtubules have been subjected to limited proteolysis with trypsin, chymotrypsin, elastase, clostripain, proteinase lysine-C, thermolysin, protease V8, papain, subtilisin, proteinase K, proteinase aspartic-N, and bromelain. Eighty nicking points have been mapped onto the α- and β-tubulin sequences with the aid of site-directed antibodies, of which 18 sites have been exactly determined by N-terminal sequencing, and the probable position of 6 others deduced from protease ...

Inosine-5'-monophosphate dehydrogenase (IMPDH) is an essential enzyme for nucleotide metabolism and cell proliferation. Despite IMPDH is the target of drugs with antiviral, immunosuppressive and antitumor activities, its physiological mechanisms of regulation remain largely unknown. Using the enzyme from the industrial fungus Ashbya gossypii, we demonstrate that the binding of adenine and guanine nucleotides to the canonical nucleotide binding sites of the regulatory Bateman domain induces different ...

Ferredoxin-dependent thioredoxin reductase was identified 35 y ago in the fermentative bacterium Clostridium pasteurianum [Hammel KE, Cornwell KL, Buchanan BB (1983) Proc Natl Acad Sci USA 80:3681–3685]. The enzyme, a flavoprotein, was strictly dependent on ferredoxin as reductant and was inactive with either NADPH or NADH. This early work has not been further pursued. We have recently reinvestigated the problem and confirmed that the enzyme, here designated ferredoxin-dependent flavin thioredoxin ...

In this work, several of the most popular and state of the art classification methods are compared as pattern recognition tools for classification of resistance spot welding joints. Instead of using the result of a non-destructive testing technique as input variables, classifiers are trained directly with the relevant welding parameters, i.e. welding current, welding time and the type of electrode (electrode material and treatment). The algorithms are compared in terms of accuracy and area under ...

Access to employment for people with intellectual disability (ID) has become a social priority. The aim of the present study is to try to determine which variables [sociodemographic variables, intelligence quotient (IQ), presence or absence of a psychiatric disorder, functioning, self-determination, and behavioural problems] could most reliably account for access to remunerated employment of people with ID.Two groups of people with ID participated in this study: (1) 69 workers in a sheltered-employment ...

A main problem in autonomous vehicles in general, and in unmanned aerial vehicles (UAVs) in particular, is the determination of the attitude angles. A novel method to estimate these angles using off-the-shelf components is presented. This paper introduces an attitude heading reference system (AHRS) based on the unscented Kalman filter (UKF) using the three-axis attitude determination (TRIAD) algorithm as the observation model. The performance of the method is assessed through simulations and compared ...

Secretory vesicles of chromaffin cells are acidic organelles that maintain an increasing pH gradient towards the cytosol (5.5 vs. 7.3) that is mediated by V-ATPase activity. This gradient is primarily responsible for the accumulation of large concentrations of amines and Ca2+, although the mechanisms mediating Ca2+ uptake and release from granules, and the physiological relevance of these processes, remain unclear. The presence of a vesicular matrix appears to create a bi-compartmentalised medium ...

Classification and regression tree (CART) and random forest techniques were proposed as pattern recognition tools for classification of ultrasonic oscillograms of resistance spot welding (RSW) joints. The results showed that CART models produced an acceptable error rate with high interpretability. These features may be used to understand and control the decision processes, instruct other human operators, compare margins of safety or modify them depending on the criticality of the industrial process. ...

This paper presents the design, fabrication, and characterization of a planar broadband chessboard structure to reduce the radar cross-section (RCS) of an object. The chessboard -like configuration is formed by combining two artificial magnetic conductor (AMC) cells. The bandwidth limitations intrinsic to AMC structures are overcome in this work by properly selecting the phase slope versus frequency of both AMC structures. A 180 ° phase difference has been obtained over more than 40% frequency bandwidth ...

In this paper, we give a succinct introduction to some basic concepts imported from the fields of Machine and Statistical Learning that can be useful in the analysis of complex agent-based models (ABM). The paper presents some guidelines in the design of experiments. It then focuses on considering an ABM simulation as a computational experiment relating parameters with a response variable of interest, i.e. a statistic obtained from the simulation. This perspective gives the opportunity of using a ...

Abstract Some Cretaceous dinosaur taxa with a broad enough record on the continents of the northern hemisphere (Laurasia) or in the southern continents (Gondwana) have been interpreted as Laurasian or Gondwanan in origin. The occasional presence of these taxa outside Laurasia or Gondwana respectively has frequently been explained in terms of dispersal from their place of origin by means of land bridges that are indeterminate in location and character. One example of such a dispersal event is provided ...

The vertebrate-bearing beds of the Laño quarry (Condado de Treviño) are among the most relevant sites from the Late Cretaceous of Europe. Geologically, Laño and the adjacent region are set on the southern limb of the South-Cantabrian Synclinorium (SE Basque-Cantabrian Region, northern Iberian Peninsula). The Laño sites were discovered in 1984; thousands of bones and teeth, including microfossils, have been collected during the prospection in the field and excavation campaigns. Abstract The effect of the LnOx support has been studied for Ni‐based CO 2 methanation catalysts. 10 wt.% nickel catalysts with LaOx, CeO 2 and PrOx supports have been prepared, characterized by N 2 adsorption, XRD, XRF, TG‐MS (N 2 ‐TPD and H 2 ‐TPR) and XPS, and have been tested for CO 2 methanation. The catalytic activity follows the trend Ni/CeO 2 >Ni/PrOx≫Ni/LaOx, all catalysts being very selective towards CH 4 formation. The activity depends both on the nature of the catalytic active sites and ...

The use of fossil fuels to satisfy the growing energy demand results in the emission of a huge amount of CO2 to the atmosphere. One alternative to overcome this environmental issue is the CO2 valorization through the storage and in situ hydrogenation to CH4. In this work, Ru-CaO/Al2O3 and Ru-Na2CO3/Al2O3 dual function materials are synthesized with different adsorbent loadings, namely 5, 10 and 15 wt.%. The prepared catalysts are characterized in terms of surface area by N2 adsorption and desorption, ...
