ABSTRACT The aim of this work was to catalog the clinical and ecoepidemiological characteristics of melioidosis in Brazil. The clinical-epidemiological features of melioidosis in Ceará are similar to those in other regions where the disease is endemic. The present study described clinical and epidemiological aspects of three cases of feline histoplasmosis and compared them to previously described cases. A detailed mycological identification and antifungal susceptibility profile of each isolate are presented. ABSTRACT This study evaluated the in vitro effect of sulfamethoxazole-trimethoprim against Histoplasma capsulatum var. capsulatum isolated from HIV-positive patients. The drugs were tested by microdilution testing in accordance with the CLSI guidelines. Herein, different microorganisms were tested in the enantioselective reduction of ethyl 2-oxo-4-phenylbutyrate in aqueous medium for the preparation of ethyl (R)-2-hydroxy-4-phenylbutyrate, a key intermediate in the production of angiotensin converting enzyme (ACE) inhibitors. Several microorganism strains were used to reduce 2,2,2-trifluoroacetophenone (1) and 4′-Br-2,2,2-trifluoroacetophenone (3). Immobilized cells of Geotrichum candidum in calcium alginate led to conversion and enantiomeric excess higher than 99%. By using immobilized G. candidum cells under continuous flow conditions, the same conversion and enantiomeric excess were achieved in 90 min of residence time.

Microorganisms were used to reduce ethyl 4-chloroacetoacetate (CAAE) to ethyl (S)-4-chloro-3-hydroxybutanoate [(S)-CHBE]. Mucor ramannianus provided 98% conversion with 84% ee. Free cells of Kluyveromyces marxianus led to 95% conversion with 81% ee. After a fractionary factorial design to study the reaction conditions, calcium alginate immobilized cells of K. marxianus furnished the product with 99% conversion with 91% ee.

Seven wild-type microorganism strains were used to reduce ethyl 3-oxohexanoate to ethyl (R)-3-hydroxyhexanoate. Free cells of Kluyveromyces marxianus and Aspergillus niger led to higher than 99% of conversion with higher than 99% ee. After immobilization in calcium alginate spheres, cells of K. marxianus exhibited high enantioselectivity (>99% ee) and conversion level (99%) within 24 h even if substrate was added at concentration of 10 g/L (62 mM).

This study analyzed the RYP1 gene as a target for the molecular diagnosis of histoplasmosis. This assay detected fungal DNA in 13/13 blood samples from HIV/AIDS-patients with histoplasmosis. Therefore, the detection of RYP1 gene in whole blood sample is a quick and sensitive test to diagnose histoplasmosis.
