A chlorinated isocoumarin with an acetylenyl side chain and its 3,4-dihydro derivative, named gymnopalynes A (1) and B (2), were isolated from cultures of a basidiomycete originating from the rain forest of Northern Thailand. The producing organism was identified as a species of Gymnopus (Marasmiaceae). Their structures were elucidated by spectroscopic methods including UV/vis and NMR spectroscopy as well as high-resolution mass spectrometry as 3-(3-chloroprop-1-yn-1-yl)-1H-isochromen-1-one (1) and ...

From stromata (fruiting bodies) of three xylariaceous fungi belonging to the genus Annulohypoxylon (A. leptascum, A. cf. truncatum and an unidentified Annulohypoxylon sp.), truncatone A (1), three unprecedented derivatives named truncatones B–D (2–4) besides the known 4,5,4′,5′-tetrahydroxy-1,1′-binaphthyl (BNT, 5), hypoxylonol C (6) and hypoxylonol F (7) were isolated. Planar structures of the new benzo[j]fluoranthene derivatives 2–4 were determined on the basis of NMR and HRESIMS data. While the relative ...

Bioassay-guided fractionation of antibacterial extracts from cultures of a basidiomycete from Northern Thailand, which represents a new species of the genus Deconica, yielded the terpenoid deconin A (1), whose structure was elucidated by spectral methods (NMR, HRMS) as a cuparenic/mevalonic acid conjugate. The absolute configuration of 1 was determined after saponification and comparison of specific rotations of the resulting cuparenic acid and mevalonolactone with authentic standards and literature ...

A specimen of the Hypoxylon rubiginosum complex featuring unusual bicoloured stromata was collected in northern Thailand and examined by means of classical morphological methodology, complemented by studies of its secondary metabolites using high performance liquid chromatography coupled with diode array and electrospray mass spectrometric detection (HPLC-DAD/MS), and molecular phylogenetic analysis of its ITS and partial beta-tubulin DNA sequences. In addition, the ultrastructure of its ascospores ...

Rationale Although mass spectrometry (MS) is a powerful tool in structural elucidation of unknown flavonoids based on their unique fragmentation patterns, proposing the correct fragmentation mechanism is still a challenge from tandem mass spectrometry data only. In recent years, computational tools such as molecular networking and MS2LDA have played a major role in the identification of structurally related compounds through an in‐depth survey of their fragmentation patterns. Methods Therefore, in ...

To date little is known about the genetic background that drives the production and diversification of secondary metabolites in the Hypoxylaceae . With the recent availability of high-quality genome sequences for 13 representative species and one relative ( Xylaria hypoxylon ) we attempted to survey the diversity of biosynthetic pathways in these organisms to investigate their true potential as secondary metabolite producers. Manual search strategies based on the accumulated knowledge on biosynthesis ...

During the course of a screening for novel anti-infective agents from cultures of tropical Xylariaceae originating from French Guiana and Thailand, pronounced antifungal activity was noted in extracts of cultures of Hypoxylon monticulosum. A bioassay-guided fractionation led to the known metabolite sporothriolide as active principle. In addition, three new derivatives of sporothriolide were isolated, for which we propose the trivial names sporothric acid, isosporothric acid and dihydroisosporothric ...

Tailoring enzymes in cytochalasan biosynthesis are relatively promiscuous. Exploiting this property, we deduced the function of four cryptic cytochrome P450 monooxygenases via heterologous expression of six cytochrome P450-encoding genes, originating from Hypoxylon fragiforme and Pyricularia oryzae, in pyrichalasin H ΔP450 strains. Three cryptic cytochrome P450 enzymes (HffD, HffG, and CYP1) restored pyrichalasin H production in mutant strains, while CYP3 catalyzed a site-selective epoxidation leading ...

The diversity of azaphilones in stromatal extracts of the fungus Hypoxylon fragiforme was investigated and linked to their biosynthetic machineries by using bioinformatics. Nineteen azaphilone-type compounds were isolated and characterized by NMR spectroscopy and mass spectrometry, and their absolute stereoconfigurations were assigned by using Mosher ester analysis and electronic circular dichroism spectroscopy. Four unprecedented bis-azaphilones, named hybridorubrins A-D, were elucidated, in addition ...

CD3-engaging bispecific antibodies (bsAbs) and chimeric antigen receptor (CAR) T cells are potent therapeutic approaches for redirecting patient T cells to recognize and kill tumors. Here we describe a fully human bsAb (REGN5458) that binds to B-cell maturation antigen (BCMA) and CD3, and compare its antitumor activities vs those of anti-BCMA CAR T cells to identify differences in efficacy and mechanism of action. In vitro, BCMAxCD3 bsAb efficiently induced polyclonal T-cell killing of primary human ...

Abstract We report new stromatal azaphilone pigments rubiginosins Z‐X from the ascomycete Hypoxylon rubiginosum , as well as rubiginosins Z and W from H. texense , which were isolated along with known monomeric and dimeric congeners. Structures were elucidated using comprehensive HRMS, NMR, and ECD analysis, revealing azaphilones from both fungi to be exclusively C‐8( S )‐configured. The orsellinic acid (OA)‐carrying rubiginosins A, Z and dimeric rutilins A‐B exhibited cytotoxicity. Rubiginosins X‐W bearing ...

In the present work, highly ordered titanium dioxide (TiO2) nanotube anodes were grown using a rapid anodization process. The photoelectrochemical performances of these electrodes strongly depend on the anodization conditions. Parameters such as electrolyte composition, anodization potential and anodization time are shown to affect the geometrical parameters of TiO2 nanotubes. The optimal anodization parameters are determined by photocurrent measurements, linear sweep voltammetry and electrochemical ...

Coronaviruses have a broad host range and exhibit high zoonotic potential. In this chapter, we describe their genomic organization in terms of encoded proteins and provide an introduction to the peculiar discontinuous transcription mechanism. Further, we present evolutionary conserved genomic RNA secondary structure features, which are involved in the complex replication mechanism. With a focus on computational methods, we review the emergence of SARS-CoV-2 starting with the 2019 strains. In that ...

Longitudinal analyses of the innate immune system, including the earliest time points, are essential to understand the immunopathogenesis and clinical course of coronavirus disease (COVID-19). Here, we performed a detailed characterization of natural killer (NK) cells in 205 patients (403 samples; days 2 to 41 after symptom onset) from four independent cohorts using single-cell transcriptomics and proteomics together with functional studies. Abstract SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2) is a novel virus of the family Coronaviridae. The virus causes the infectious disease COVID-19. The biology of coronaviruses has been studied for many years. However, bioinformatics tools designed explicitly for SARS-CoV-2 have only recently been developed as a rapid reaction to the need for fast detection, understanding and treatment of COVID-19. To control the ongoing COVID-19 pandemic, it is of utmost importance to get insight ...
