Human colorectal cancers (CRCs) display a large number of genetic and epigenetic alterations, some of which are causally involved in tumorigenesis (drivers) and others that have little functional impact (passengers). To help distinguish between these two classes of alterations, we used a transposon-based genetic screen in mice to identify candidate genes for CRC. Mice harboring mutagenic Sleeping Beauty (SB) transposons were crossed with mice expressing SB transposase in gastrointestinal tract epithelium. ...

Abstract CRISPR‐Cas9 gene editing technology provides a method to generate loss‐of‐function studies to investigate, in vivo, the specific role of specific genes in regulation of reproduction. With proper design and selection of guide RNAs (gRNA) designed to specifically target genes, CRISPR‐Cas9 gene editing allows investigation of factors proposed to regulate biological pathways involved with establishment and maintenance of pregnancy. The advantages and disadvantages of using the current gene editing ...

Importance Stroke risk varies by systolic blood pressure (SBP), race, and ethnicity. The association between cumulative mean SBP and incident stroke type is unclear, and whether this association differs by race and ethnicity remains unknown. Objective To examine the association between cumulative mean SBP and first incident stroke among 3 major stroke types—ischemic stroke (IS), intracerebral hemorrhage (ICH), and subarachnoid hemorrhage (SAH)—and explore how these associations vary by race and ethnicity. ...

Aromatase catalyzes the synthesis of estrogens and has been shown to have an important role during the establishment of pregnancy in the pig. This study confirmed the differential expression of the three aromatase isoforms.Although three porcine aromatase isoforms have been identified, their gene expression profiles in reproduction are still poorly understood. Here, we identified by Sanger sequencing unique nucleotide signatures for the three paralogous copies of Cyp19 and analyzed by RT-PCR the ...

The Prostate Imaging and Data Reporting System version 2 (PI-RADS v2) aims to simplify performance, interpretation, and reporting of prostate magnetic resonance imaging (MRI).1 PI-RADS v2 introduced probability scores (assessment categories, Table 1), which indicate the likelihood of clinically significant cancer (Gleason score ≥7)2 based upon MRI findings. PI-RADS v2 has been validated as accurate for detection of cancers and improves interobserver agreement. 3 Despite this, in our experience, use of ...

Conceptus estrogens and prostaglandins have long been considered the primary signals for maternal recognition of pregnancy (MRP) in the pig. However, loss-of-function studies targeting conceptus aromatase genes (CYP19A1 and CYP19A2) and prostaglandin-endoperoxide synthase 2 (PTGS2) indicated that conceptuses can not only signal MRP without estrogens or prostaglandins but can maintain early pregnancy. However, complete loss of estrogen production leads to abortion after day 25 of gestation. Although ...

Abstract TLR agonists have been used in treating diseases including infections, cancer and as vaccine adjuvants. Aldara™ (imiquimod 5% cream) is the first TLR7 agonist approved in the US, and is indicated for the treatment of genital warts caused by human papillomavirus (HPV). Aldara was not well-tolerated when delivered vaginally; therefore a 2nd generation analog, 851, that retained the ability to stimulate cytokine production upon vaginal application was selected for development to treat high-risk ...

Introduction: Stroke risk varies by systolic blood pressure (SBP) and race/ethnicity. Whether the relationship between cumulative mean SBP and first-time incident stroke type differs by racial/ethnic group is unknown. This study examines the relationship between cumulative mean SBP and first-time incident stroke among 3 major stroke types: ischemic stroke (IS), intracerebral hemorrhage (ICH), subarachnoid hemorrhage (SAH) and how race/ethnicity influences these relationships, using a pooled cohort analysis. ...
