The growth of transformed mouse fibroblasts (3T6 cells) in medium containing 5% fetal bovine serum was inhibited after treatment with concentrations greater than 50 microM ATP, ADP, or AMP. Adenosine, the common catabolite of the nucleotides, had no effect on cell growth at concentrations below 1 mM. However, the following results indicate that the toxicity of ATP, ADP, and AMP is mediated by serum- and cell-associated hydrolysis of the nucleotides to adenosine. 1) ADP and AMP, but not ATP, were ...

As in previous years, the consensus group to consider the use of biological agents in the treatment of rheumatic diseases met during the 13th Annual Workshop on Advances in Targeted Therapies in April, 2011. The group consisted of rheumatologists from a number of universities among the continents of Europe, North America, South America, Australia and Asia. Pharmaceutical industry support was obtained from a number of companies for the annual workshop itself, but these companies had no part in the ...

Both testosterone and oxytocin influence an individual's accuracy in inferring another's feelings and emotions. Fetal testosterone, and the second-to-forth digit ratio (2D:4D) as its proxy, plays a role in social cognitive development, often by attenuating socio-affective skill. Conversely, oxytocin generally facilitates socio-affiliative and empathic cognition and behavior. A common polymorphism in the oxytocin receptor gene, OXTR rs53576, has been repeatedly linked with psychosocial competence, ...

Prevotella copri (P copri), a gut commensal, has been reported to be an immune-relevant organism in individuals with rheumatoid arthritis (RA). This study sought to evaluate anti-P copri (anti-Pc) antibody responses in our participant cohorts and to determine when in the natural history of RA such responses develop.

This chapter presents the procedures that increase the plasma membrane permeability of mammalian cells to normally impermeant compounds. Permeabilization methods described include treatment of cells with adenosine 5'-triphosphate (ATP), ionophores, Sendai virus, detergents, and Staphylococcus aureus α-toxin. Physical methods for altering the normal permeability barrier of cultured cells, including scrapeloading, treatment with hypertonic medium, and transient exposure to an electric field, are discussed. ...

The management of advanced laryngeal cancer is evolving, with increasing use of chemoradiation as initial treatment. Recent reports confirm a decline in total laryngectomies (TLs) in the United States. A study was undertaken to evaluate national and regional trends in TLs performed over the most recent decade for which data were available and to use multivariate analysis to characterize these trends in more detail.Population-based cohort study.Healthcare Cost and Utilization Project-Nationwide Inpatient ...

The COVID-19 pandemic has been especially challenging for patients with rheumatic diseases because immune-mediated diseases as well as their treatment could adversely impact susceptibility to or severity of a viral infection.1 2 A recent study from New York City on 86 patients with COVID-19 infection and a history of immune-mediated disease seemed to show that the use of methotrexate, oral glucocorticoids or hydroxychloroquine increased the risk for hospitalisation,3 although the authors still concluded ...

To the Editor: Although many genes have polymorphisms, major histocompatibility complex genes are the most polymorphic. Many assume that the diversity of HLA increases the likelihood that a species can survive pandemics. Indeed, evidence suggests that HLA-B27 is protective for HIV1, hepatitis C2, and possibly influenza3. We recently reported results of a Web-based survey involving patients with spondyloarthritis (SpA)4. We now report an additional analysis of these data obtained between April 10, ...

These configurations used the same physical parameterizations and the same input dataset for the initial and boundary conditions, differing primarily in their spatial resolution. Abstract Herein, a summary of the authors’ experiences with 36-h real-time explicit (4 km) convective forecasts with the Advanced Research Weather Research and Forecasting Model (WRF-ARW) during the 2003–05 spring and summer seasons is presented. These forecasts are compared to guidance obtained from the 12-km operational Eta Model, which employed convective parameterization (e.g., Betts–Miller–Janjić). The results suggest significant value added for the high-resolution forecasts in representing the ...

Abstract A 1‐year‐old, 5‐ to 6‐week pregnant cat was presented with a history of aborting 3 kittens the previous night. During the examination, the cat began to seizure and died. At necropsy, formalin‐fixed tissues and blood smears prepared from an EDTA blood sample collected via cardiac puncture were submitted to a diagnostic laboratory. Examination of the blood smears revealed numerous large (15–75 μm) clumped macrophages containing dark blue intracytoplasmic organisms consistent with Cytauxzoon ...

Antibodies against the 19 kDa C-terminal fragment of merozoite surface protein 1 (MSP1(19)) are a major component of the invasion-inhibitory response in individuals immune to malaria. Abstract Background Recent studies indicate an increase in tumor progression and recurrence in head and neck squamous cell carcinomas (HNSCC) of cancer patients taking recombinant human erythropoietin (rhEpo) for anemia. This study was undertaken to investigate the potential role of rhEpo in invasion, proliferation, and cisplatin-induced cell death in HNSCC cell lines. Methods The following experiments were performed with two HNSCC cell lines, UMSCC-10B and UMSCC-22B. Presence of EpoR in both cell ...

Although clinically evident metastases of nonthyroid malignancies (NTMs) to the thyroid gland are uncommon, it is important to suspect them in patients who present with a new thyroid mass and a history, however far back, of prior malignancy. In fact, metastases from NTMs to the thyroid gland have been reported in 1.4%-3% of all patients who have surgery for suspected cancer in the thyroid gland. Here we review the literature over the last decade regarding this topic.Based on recent literature, the ...

The ability of nicotine to enhance the malignancy of cancer cells is known; however, the possibility that nicotine could regulate a cancer stem cell phenotype remains to be well-established. In this study we sought to determine whether long-term exposure to nicotine could promote cancer stem cell-like properties in two head and neck squamous cell carcinoma cell lines, UMSCC-10B and HN-1. Nicotine treatment induced epithelial-to-mesenchymal transition (EMT) in both cell lines by repressing E-cadherin ...

Objectives Previously, we found that omega-3 fatty acids (n-3 FAs) were inversely associated with anti-cyclic citrullinated peptide (anti-CCP) positivity in participants at risk for future rheumatoid arthritis (RA). Methods The Studies of the Etiology of RA (SERA) cohort includes RA-free participants who are at increased risk for RA. ...

Salivary glands sustain collateral damage following radiotherapy (RT) to treat cancers of the head and neck, leading to complications, including mucositis, xerostomia and hyposalivation. Despite salivary gland-sparing techniques and modified dosing strategies, long-term hypofunction remains a significant problem. Current therapeutic interventions provide temporary symptom relief, but do not address irreversible glandular damage. In this review, we summarize the current understanding of mechanisms ...
