Background Biliary obstruction leads to an increase in biliary pressure within the biliary system, which induces the morphologic adaptation of the biliary tree. Purpose To observe and to quantify the morphologic characteristics of the adaptation in a bile duct ligation rat model and verify it in patients with biliary atresia in a three-dimensional (3D) manner using x-ray phase-contrast CT. Materials and Methods A bile duct ligation model was induced in 40 male Sprague-Dawley rats, which were divided ...

This chapter discusses hepatic stem cells (HpSCs) and provides protocols on HpSCs, especially human hepatic stem cells (hHpSCs). It also includes development of a serum-free, hormonally defined medium (HDM), preparation of tissue extracts enriched in extracellular matrix, and methods to design biodegradable, polylactide scaffoldings or microcarriers in ways appropriate for progenitors and use of bioreactors. There has been recognition that the epithelial–mesenchymal relationship is lineage dependent. ...

Macrophages affect the magnitude and duration of inflammatory response in a functionally heterogeneous manner. The phenotype of macrophages is maintained through a reversible homeostatic mechanism. A number of determinants that modulate macrophage plasticity have been identified, although the precise mechanisms are not fully understood. In this study, we report that stimulation of isolated human blood monocytes and mouse bone marrow-derived macrophages with human serum amyloid A (SAA), a major acute- ...

Microvascular changes in liver fibrosis are considered as an important pathophysiological characteristic. Now, there is a lack of high-resolution three-dimensional (3D) imaging methods for observing the microvasculature throughout the entire livers. This study aims to investigate the 3D microvascular changes in the whole fibrotic livers via X-ray phase-contrast computed tomography (PCCT) and explore the correlations between portal pressures and microvascular changes in liver fibrosis progression. ...

Atherosclerosis is a primary cause of heart disease and stroke; it is the underlying cause of about 50% of all deaths in Western countries. It is known that early detection of atherosclerotic lesions would significantly reduce the risk of mortality. The objective of this study was to develop a radioimaging method for early detection of atherosclerotic plaques. A novel polyiodinated cholesterol analog, cholesteryl 1,3-diiopanoate glyceryl ether (C2I, patent pending), was synthesized and radiolabeled ...

Introduction:The use of ex vivo expansion of hematopoietic progenitor cells has been suggested as a possible means to accelerate the speed of engraftment in cord blood (CB) transplantation. Macrophage inflammatory protein-1 alpha (MIP-1a) has been shown to have a role in the control of myeloid stem and progenitor cell proliferation. So the aim of this study was to evaluate the effects of MIP-1a on ex vivo expansion of hematopoietic progenitor cells in different culture media for the propose of cord ...

Purpose Examine the feasibility of characterizing the regulation of renal oxygenation using high‐temporal‐resolution monitoring of the response to a step‐like oxygenation stimulus. Methods For mapping, multi‐echo gradient‐echo imaging was used (temporal resolution = 9 seconds). A step‐like renal oxygenation challenge was applied involving sequential exposure to hyperoxia (100% O 2 ), hypoxia (10% O 2 + 90% N 2 ), and hyperoxia (100% O 2 ). In vivo experiments were performed in healthy rats (N = 10) and in rats with ...
