The HIV-1 Nef protein was analyzed for apoptotic structural motifs that interact with the CXCR4 receptor and induce apoptosis in CD4(+) lymphocytes. Two apoptotic motifs were identified. One centered on Nef amino acids (aa) 50 to 60, with the overlapping 20-mer peptides retaining about 82% of the activity of the full Nef protein. The second centered on aa 170 to 180, with the overlapping 20-mer peptides retaining about 30% of the activity of the full protein. Significant apoptotic abilities were ...

Abstract Understanding energy transfer among hydroporphyrins is of fundamental interest and essential for a wide variety of photochemical applications. Toward this goal, a synthetic free base ethynylphenylchlorin has been coupled with a synthetic free base bromobacteriochlorin to give a phenylethyne‐linked chlorin–bacteriochlorin dyad ( FbC‐pe‐FbB ). The chlorin and bacteriochlorin are each stable toward adventitious oxidation because of the presence of a geminal dimethyl group in each reduced pyrrole ...

Alpha-particle spectrometry is a powerful analytical tool for nuclear forensics and environmental monitoring. Superconducting transition-edge sensor microcalorimeters have been shown to yield unsurpassed energy resolution for alpha spectrometry. With nearly an order of magnitude better energy resolution (1.06 keV FWHM at 5.3 MeV) than the current state-of-the-art silicon detectors (8-10 keV at 5.3 MeV), it is possible to measure samples containing multiple radioisotopes that would require expensive ...

Bicontinuous microemulsions made from oil, water, and surfactants were examined as substitutes for organic solvents in carbon−carbon bond-forming reactions. Conjugated additions of primary alkyl iodides 3a−c to 2-cyclohexen-1-one (4) to give 3-alkyl cyclohexanones 5a−c and cyclization of 2-(4-bromobutyl)-2-cyclohexen-1-one (9) to 1-decalone (10) were mediated by the Co(I)L complex vitamin B12s generated at carbon cloth electrodes under mild conditions. The first one-pot deoxycyanamidation of alcohols has been developed using N-cyano-N-phenyl-p-methylbenzenesulfonamide (NCTS) as both a sulfonyl transfer reagent and a cyanamide source, accessing a diverse range of tertiary cyanamides in excellent isolated yields. This approach exploits the underdeveloped desulfonylative (N–S bond cleavage) reactivity pathway of NCTS, which is more commonly employed for electrophilic C- and N-cyanation processes.

Abstract The importance of Müller glia for retinal homeostasis suggests that they may have vulnerabilities that lead to retinal disease. Here, we studied the effect of selectively knocking down key metabolic genes in Müller glia on photoreceptor health. Immunostaining indicated that murine Müller glia expressed insulin receptor (IR), hexokinase 2 (HK2) and phosphoglycerate dehydrogenase (PHGDH) but very little pyruvate dehydrogenase E1 alpha 1 (PDH‐E1α) and lactate dehydrogenase A (LDH‐A). An integrated system-in-package-based 60-GHz 4 $\,\times\,$ 4 antenna array with CMOS on–off keying (OOK) transmitter and low-noise amplifier (LNA) is investigated. The 4 $\,\times\,$ 4 circular polarized array exhibits a wide impedance bandwidth $({\rm VSWR}< 2)$ and 3-dB axial ratio bandwidth of over 8 GHz using a strip line sequential rotation feeding scheme. It has a beam-shaped pattern with a 3-dB beamwidth of 20 $^{\circ}$ and a peak gain of 16.8 dBi. The modulators in 90-nm CMOS includes 60-GHz oscillators ...

Four randomized, double-blind, placebo-controlled, 4-period drug-drug interaction studies were conducted in healthy subjects to evaluate the pharmacokinetic and pharmacodynamic (PD) interactions between mirogabalin and commonly used central nervous system depressants. Mirogabalin or placebo was administered alone or with single-dose lorazepam, zolpidem, tramadol, ethanol, or interacting drug placebo. Safety was assessed and serial samples for pharmacokinetic parameters were collected for up to 48 ...

Lasso peptides are a family of ribosomally synthesized and post-translationally modified peptides (RiPPs) defined by their threaded-ring topology. The N-terminus of the peptide forms an isopeptide bond with an aspartate or glutamate side chain to create a 7–9 amino acid (aa) macrocyclic ring through which the rest of the peptide is threaded. The result is a highly constrained three-dimensional structure. Even though they share a threaded-ring feature, characterized lasso peptides vary greatly in sequence ...

Further optimization efforts on the previously disclosed PTP-1B inhibitor 1 a led to the discovery of a new series of potent compounds, for example derivative 33, which targeted the second phosphotyrosine binding pocket near the catalytic site. In the new series, the N-sulfonylpiperidine group of 1 a was replaced with a smaller substituent, capable of forming a new hydrogen bond interaction with arginine 24. The series reported here maintains the binding mode of 1 a and has the added advantages ...
