Few instances of a single act of legislation have shifted industrial policy in the pharmaceutical industry like the Orphan Drugs Act did when it was signed in the USA in 1983. Susceptibility to haemolytic anaemia after ingestion of fava beans has been investigated extensively in the Mediterranean, but favism has also been observed in other countries' including England,2 where variants of glucose-6-phosphate dehydrogenase other than the Mediterranean variant have been responsible. In general, all patients have very low glucose-6-phosphate dehydrogenase activity in red cells (<5% of normal activity). Favism is widely believed not to occur in subjects with the glucose-6-phosphate ...

A SINGLE base-pair mutation (βs) in codon 6 of the human β-globin gene, causing a single amino-acid substitution, is the cause of sickle cell anaemia1. The mutant haemoglobin molecule, HbS, polymerizes when deoxygenated and causes deformation of the erythrocytes to a characteristic 'sickled' shape. Sickling of cells in small vessels causes painful crises and other life-threatening complications2,3. Although the molecular basis for sickle cell anaemia has been known for 30 years, no definitive treatment ...

Summary We have studied the arrangement of the α, β and γT cell receptor (TCR) genes in 27 patients with T cell lymphoproliferative disorders. Nine patients had acute lymphoblastic leukaemia (T‐ALL), nine patients had prolympho‐cytic leukaemia (PLL), six patients presented with a T‐CLL/T‐lymphocytosis syndrome, two patients had Sezary syndrome (SS) and one patient had HTLV‐I positive T‐cell leukaemia/ lymphoma (ATLL). α TCR gene rearrangement could be demonstrated by the use of three available probes ...

We have established 3 new EBV-negative cell lines, designated Sc-1, Ri-1 and Ci-1, from patients with B-cell lymphoma/leukemia. We characterized them by cytogenetics and by study of surface membrane antigens with a panel of monoclonal antibodies (MAbs), surface and cytoplasmic immunoglobulin (Ig) expression and Ig heavy- and light-chain genes. All 3 lines had a 14q+ abnormality. Ri-1 also had translocations involving chromosomes 2, 8 and 18. Ci-1 also had abnormalities involving chromosomes 2, 8 and ...

Abstract Leukaemic cells from 2 patients with B‐prolymphocytic leukaemia were immortalized in vitro by means of Epstein‐Barr virus and phorbol‐ester TPA. The resulting cell lines, named JVM‐2 and JVM‐3, have been growing continuously in liquid culture for more than one year. JVM‐2 is characterized cytogenetically by t(11;14)(q13;q32), and JVM‐3 by trisomy 12. The immunoglobulin (Ig) heavy‐and light‐chain genes showed the same pattern of rearrangement in both lines as in the original prolymphocytes ...

In 1977 the Red Cell Enzyme Expert Panel of the International Committee for Standardization in Haematology (ICSH) issued recommended methods for red cell enzyme analysis (Beutler et al. 1977).These methods have made it possible to measure red cell enzyme activity with reliable and convenient techniques, and to compare results obtained in different laboratories. After the publication of the ICSH recommended methods, pyrimidine 5'-nucleotidase (P5N) deficiency came to our attention. This enzyme was first ...

Abstract Two distinct types of T-cell receptors (TCR), designated alpha beta and gamma delta, have been identified on the surface of T cells. In the adult, T cells bearing the gamma delta TCR are a minority and they have the phenotype CD3+, CD4-, CD8-/+. By using appropriate probes, rearrangements of the TCR alpha, beta, and gamma genes have been extensively investigated in a variety of lymphoproliferative disorders. Because the TCR delta gene has been cloned only recently, no comparable information ...

Using the dominant control region (DCR) sequences that flank the beta-globin gene locus, we have been able to achieve high-level expression of the human alpha-globin gene in transgenic mice. Expression in fetal liver and blood is copy number dependent and at levels comparable to that of the endogenous mouse alpha-globin genes. Transgenic fetuses with high-copy numbers of the transgene suffer severe anemia and die before birth. Using a construct with both the human alpha- and beta-globin genes and ...

The Terai region of Nepal has been known to be heavily malarious since remote times, and it has, therefore, been regarded as uninhabitable by most Nepalese people. The Tharu people, who have been living in the Terai for centuries, were reputed to have an innate resistance to malaria. Following successful control of malaria by the Nepal Malaria Eradication Organization (NMEO), a large and heterogeneous non-Tharu population now inhabits the Terai along with Tharus. By analysing NMEO records, we have ...

Lymphomatoid granulomatosis and lethal midline granuloma are both characterized histologically by atypical pleomorphic angiocentric infiltrates. Whether these conditions are malignant lymphoproliferative disorders remains controversial. Here we report the results of studies carried out in a patient with coeliac disease, who developed recurrent self-healing subcutaneous nodules with the histological changes of lymphomatoid granulomatosis and an invasive nasal tumour with the histological features ...
