The application of high-resolution melting (HRM) analysis of DNA is reported for scanning and genotyping Olea europaea germplasm. To test the sensitivity of the method, a functional gene marker, phytochrome A (phyA), was used, since this gene is correlated with important traits for the ecology of the species. We have designed a set of oligos able to produce amplicons of 307 bp to scan for the presence of single polymorphic mutations in a specific phyA fragment encompassing the chromophore attachment ...

Pregnancy is associated with an increased risk of venous thromboembolism, which probably varies according to the presence of single or multiple thrombophilic defects. Acquired or inherited thrombophilia is moreover associated with adverse outcomes in pregnancy. For this reason, in the past, pregnant women at risk of venous thromboembolism or pregnancyes have been treated with oral anticoagulants or unfractionated heparin. Both of them are associated with fetal or maternal side effects. Low-molecular- ...

The effect of salt stress on leaf morphology and functionality was studied in three Populus alba genotypes differing in tolerance to salinity: 6K3 (sensitive), 2AS11 (moderately tolerant), and 14P11 (tolerant). Plants were subjected to an intense and progressive salt stress from 50 to 250 mM NaCl by 50 mM steps at 10-day intervals. The micromorphological results highlighted phenotypic variation among the three genotypes already in control plants, with the genotype 14P11 having significantly smaller ...

Light is a fundamental environmental cue in the life of plants, playing a crucial role, directly or indirectly, in the regulation of plant development and growth. It represents the signal to be elaborated in the photoperception processes by the photoreceptors. Plants have evolved exquisite sensory systems for monitoring their environment and initiating appropriate strategies in their lifetime. In order to optimise the acquisition of light energy for photosynthesis, plants have developed a series ...

Hemophilia A is an X-linked bleeding disorder caused by widespread mutations in the human coagulation factor 8 gene. We have searched for mutations in factor 8 gene DNAs from 40 unrelated Italian patients with hemophilia A. All patients came from the same region (Calabria) and were followed-up at the same hemophilia center. Of the 40 patients, 20 (50%) had severe hemophilia A, 19 (47.5%) had moderate hemophilia A, and one (2.5%) had mild hemophilia A. All patients were first screened for the common ...

This paper presents the results of the interaction of red light (R) with blue light (B), applied to shoots of M9 apple (Malus pumila paradisiaca Schmid) rootstock, on the regulation of stem growth, apical dominance and branching. These results are compared with the active form of phytochromes (PHYs) under monochromatic and dichromatic light treatments. The inhibition of internode elongation was determined by PHY photoequilibrium, with the additional effect of B, by means of a separate photoreceptor. ...

The effect of light quality on in vitro rooting of cherry rootstock "Colt" was studied to evaluate adventitious root development, ex vitro acclimatization and plant growth performance. Adventitious rooting was dependent on the light quality to which the microcutting were exposed. Highest number of roots per microcuttings was recorded under dichromatic light (blue + red). This response could be ascribed to the control of a strong synergistic interaction between phytochromes and blue light photoreceptors. ...

Salicylic acid (SA), ethylene (ET), and wounding are all known to influence plant defense response. Experiments attempting to determine SA's relation to ET biosynthesis and defense gene expression have shown conflicting results. To confront this, we developed an in vitro model system to investigate how SA affects ET biosynthesis, hydrogen peroxide (H2O2) production and endochitinase gene expression in the European chestnut. ET measurements of in vitro shoots indicated a critical time point for SA exogenous ...

SummaryThe genotyping of cultivated varieties using molecular markers is a crucial aim of modern plant breeding programmes, germplasm collection management and product traceability. Methods for DNA extraction from olive oil are now available, and the DNA extracted is suitable for PCR applications. The aim of this work was to identify simple and reliable PCR markers useful for cultivar identification and oil traceability. We chose to analyse 13 cultivars and one feral accession of Olea europaea for ...

The interaction between shoot proliferation and regulative action of light quality was investigated in terms of cluster growth and development. In the commercial laboratory, the shoot multiplication procedure is normally conducted by growing clusters of shoots during several sub-cultures. Thus in this work the regulative action of light quality was evaluated by analysing the shoot cluster proliferation rate, the evolution of axillary bud differentiation, the axillary shoot development on both initial ...

Self-incompatibility is one of the most important barriers adopted by many flowering plants to prevent inbreeding, thus maintaining diversity within species. The elucidation of this reproductive constraint is crucial in olive (Olea europaea L.) because it may help to improve breeding strategies and orchard management. The available literature reports that a gametophytic self-incompatibility (GSI) system might exist in olive, even though neither cytological nor molecular data were provided. A molecular ...

Plants require access to free water for nutrient uptake, but excess water surrounding the roots can be injurious or even lethal because it blocks the transfer of free oxygen between the soil and the atmosphere. Genetic improvement efforts in this study were focused on the increased tolerance in roots to waterlogging. Among a pool of clones generated in vitro from leaf explants of rootstock Mr.S.2/5 of Prunus cerasifera L., the S.4 clone was flood tolerant whereas the S.1 clone was sensitive. The ...

Abstract Early development of immunity after hepatitis B vaccination is particularly important for patients such as hemophiliacs, at high risk for acquiring hepatitis B from potentially infectious plasmaderived concentrates. The purpose of this study was to evaluate whether or not protective antibody titers could be achieved quickly and maintained in hemophiliacs by an accelerated vaccination schedule. A yeast‐recombinant hepatitis B vaccine (Engerix B, SKF Ritt) was given subcutaneously in the deltoid ...

The olive tree grows typically in the Mediterranean Basin. It probably originated in Asia Minor and was put under cultivation in countries of the seacoast of the eastern Mediterranean between 1200–1700 B. C., depending on the regions. In the beginning, olive was more likely to be cultivated to be eaten as fruit, which contains 15 to 25% fat and was easy to be stored. Olives therefore constituted an important component of the human diet.

Abstract Many plants are incapable of flowering in inductive daylengths during the early juvenile vegetative phase (JVP). Arabidopsis mutants with reduced expression of TEMPRANILLO ( TEM ), a repressor of FLOWERING LOCUS T ( FT ) had a shorter JVP than wild-type plants. Reciprocal changes in mRNA expression of TEM and FT were observed in both Arabidopsis and antirrhinum, which correlated with the length of the JVP. FT expression was induced just prior to the end of the JVP and levels of TEM1 mRNA declined ...
