Despite a preponderance of evidence highlighting the involvement of dopamine in the regulation of neurogenesis in the adult mammalian brain, the mechanism by which this occurs remains unknown. Previous results demonstrated that dopaminergic nigrostriatal projections regulate neural precursor cell

A new technique for developing k‐distributions applied to longwave radiation parameterization has been presented in a preceding paper. Now we discuss an extension of this technique to the shortwave spectral range. A fast k‐distribution model (FKDM) for gaseous absorption calculations suitable for use in weather and climate prediction is described. FKDM has been created using 15 k‐distribution terms only, less than in other comparable codes. The molecular species represented in the model are H 2 O, ...

Halogenation of the aporphine alkaloid boldine at the 3-position leads to increased affinity for rat brain D1-like dopaminergic receptors with some selectivity over D2-like receptors. A series of 3-halogenated and 3,8-dihalogenated (halogen = Cl, Br or I) derivatives of predicentrine (9-O-methylboldine) and glaucine (2,9-di-O-methylboldine) were prepared and assayed for binding at D1 and D2 sites. Halogenation of predicentrine led to strong increases in affinity for D1-like receptors, while the affinities ...

Abstract Dopamine secreted from hypophysial hypothalamic neurons is a principal inhibitory regulator of pituitary hormone secretion. Mice with a disrupted D2 dopamine receptor gene had chronic hyperprolactinemia and developed anterior lobe lactotroph hyperplasia without evidence of adenomatous transformation. Unexpectedly, the mutant mice had no hyperplasia of the intermediate lobe melanotrophs. Aged female D2 receptor −/− mice developed uterine adenomyosis in response to prolonged prolactin exposure. These ...

We present a detailed and quantitative comparison of the quantum mechanical (QM) and quasiclassical (QCT) descriptions of the stereodynamics of elementary chemical reactions. Analogous formulations of the QM and QCT k–k′–j′ vector correlation in atom–diatom reactions have been derived and shown to be equivalent in the correspondence principle limit. The comparison between the results obtained from the application of the QM and QCT methodologies to the H+D2(v=0, j=0)→HD(v′,j′)+D reaction at a collision ...

Behavioral sensitization to psychostimulants manifests as an increased locomotor response with repeated administration. Dopamine systems are accepted to play a fundamental role in sensitization, but the role of specific dopamine receptor subtypes has not been completely defined. This study used the combination of dopamine D2 receptor-deficient mice and a D1-like antagonist to examine dopamine D1 and D2 receptor involvement in acute and sensitized locomotor responses to methamphetamine. Absence of ...

We present results of quantum calculations we have performed on the title reaction in order to study its stereodynamics at collision energies of 0.54 and 1.29 eV. Our theoretical model is based on a representation where directional properties are expressed in terms of real rotational polarization moments instead of magnetic quantum numbers. We analyze the physical meaning of rotational polarization moments and show that, when defined as in the present work, these quantities directly describe the ...

The purpose of this paper is to analyze and interpret the onset of some well‐characterized transient inertio‐gravity waves, observed during the PYREX (Pyrénées Experiment) campaign. The data used are the high‐resolution soundings, launched every 6 hours on both sides of the ridge. In agreement with other observations made during the field campaign, certain among those soundings often show steady mountain waves. It appears that those waves are often transients and are followed by well‐characterized ...

The benefits of adjuvant treatment in the context of a D2 lymph node dissection are controversial. The aim was to investigate the effects of postoperative adjuvant treatment on the survival of patients with a curative resection for gastric cancer and a D2 lymph node dissection. We performed a retrospective cohort study. Patients operated from 1996 to 2013 were selected. We compared long term survival of patients treated with surgery alone and those with surgery plus postoperative adjuvant treatment. ...

Rationale. Dopamine (DA) D2-like antagonists block several effects of cocaine, including its locomotor stimulant and interoceptive discriminative-stimulus effects. Because these compounds generally lack selectivity among the D2-like DA receptors, the specific roles of the subtypes remain unclear. Objectives. DA D2 receptor knockout (DA D2R KO), heterozygous (HET), and wild-type (WT) mice were used to study the role of D2 DA receptors in the effects of cocaine. Some effects of the relatively selective ...

The rewarding effects of opiates are thought to be mediated through dopaminergic mechanisms in the ventral tegmental area, dopamine-independent mechanisms in the nucleus accumbens, or both. The purpose of the present study was to explore the contribution of dopamine to opiate-reinforced behavior using D2 receptor knock-out mice.Wild-type, heterozygous, and D2 knock-out mice were first trained to lever press for water reinforcement and then implanted with intravenous catheters. The ability of intravenously ...

Rationale: Dopamine D2 receptors are postulated to play an important role in modulating the reinforcing effects of abused drugs including ethanol. Objectives: This experiment examined operant ethanol self-administration in dopamine D2 receptor knockout (KO) mice and wild-type (WT) mice using a continuous access procedure. Methods: Adult male KO and WT mice were trained in 30-min sessions to perform a lever press response for access to 10% v/v ethanol. After training, the mice were placed in test chambers ...

Phenotypes were assessed topographically in mice lacking functional D2 dopamine receptors [‘knockouts’], using an ethologically based approach to assess all behaviours in the natural repertoire. D2-null mice evidenced an ethogram characterised initially by modest reductions in locomotion and shifts in rearing topographies. Subsequently, topographies of behaviour habituated similarly for wildtypes and ‘knockouts’. Following challenge with the D2-like agonist RU 24213, both inhibition of rearing at a ...

Typical neuroleptic drugs elicit their antipsychotic effects mainly by acting as antagonists at dopamine D2 receptors. Much of this activity is thought to occur in the cerebral cortex, where D2 receptors are found largely in inhibitory GABAergic neurons. Here we confirm this localization at the electron microscopic level, but additionally show that a subset of cortical interneurons with low or undetectable expression of D2 receptor isoforms are surrounded by astrocytic processes that strongly express ...

Pharmacological blockade studies have supported a role of the dopamine system in ethanol reward for many years, but receptor subtype specificity has been difficult to establish. Recently, genetically engineered mice lacking functional dopamine D2 receptors have been shown to drink less ethanol in a two-bottle choice task. To determine whether reduced ethanol intake reflects a reduction in ethanol reward, D2 receptor-deficient [knockout (KO)] mice were compared to heterozygous (HET) and wild-type (WT; ...

Mice receiving reserpine (1 mg/kg/day) during 5 days develop behavioral supersensitivity. To study the possible molecular correlates of these adaptive changes we compared binding parameters of D1 and D2 receptors and adenylate cyclase activity in striata from normal and reserpinized mice. Saturation curves using [3H]SCH 23390 showed no changes in maximum binding capacity (Bmax) or Kd of striatal D1 receptors taken from control or 5 days reserpine-treated mice. However, [3H]spiperone saturation curves ...

The rewarding effects of lateral hypothalamic brain stimulation, various natural rewards, and several drugs of abuse are attenuated by D1 or D2 dopamine receptor (D1R or D2R) antagonists. Much of the evidence for dopaminergic involvement in rewards is based on pharmacological agents with limited or "relative" selectivity for dopamine receptor subtypes. Genetically engineered animal models provide a complementary approach to pharmacological investigations. In the present study, we explored the contribution ...

Dopamine (DA) has, as of late, become singled out from the profusion of other neurotransmitters as what could be called a key substance, in the regulation of the sleep–wake states. We have hypothesized that dopaminergic D2 receptor blockage induced by haloperidol could generate a reduction or even an ablation of rapid eye movement (REM) sleep. Otherwise, the use of the selective D2 agonist, piribedil, could potentiate REM sleep. Electrophysiological findings demonstrate that D2 blockage produced a ...

Recently, the importance of the dopaminergic D2 receptor (D2R) subtype in normal body growth and neonatal GH secretion has been highlighted. Disruption of D2R alters the GHRH–GH–IGF-I axis and impairs body growth in adult male mice. The D2R knockout (KO) dwarf mouse has not been well characterized; we therefore sought to determine somatotrope function in the adult pituitary. Using immunohistochemistry and confocal microscopy, we found a significant decrease in the somatotrope population in pituitaries ...

Vascular endothelial growth factor (VEGF)-A is an important angiogenic cytokine in cancer and pathological angiogenesis and has been related to the antiangiogenic activity of dopamine in endothelial cells. We investigated VEGF expression, localization, and function in pituitary hyperplasia of dopamine D2 receptor (D2R)-knockout female mice. Pituitaries from knockout mice showed increased protein and mRNA VEGF-A expression when compared with wild-type mice. In wild-type mice, prolonged treatment with ...
