Other than efficacy of interaction with the molecular target, metabolic stability is the primary factor responsible for the failure or success of a compound in the drug development pipeline. The ideal drug candidate should be stable enough to reach its therapeutic site of action. Despite many recent excellent achievements in the field of computational methods supporting drug metabolism studies, a well-recognized procedure to model and predict metabolic stability quantitatively is still lacking. ...

Diverse death stimuli including anticancer drugs trigger apoptosis by inducing the translocation of cytochrome c from the outer mitochondrial compartment into the cytosol. Once released, cytochrome c cooperates with apoptotic protease-activating factor-1 and deoxyadenosine triphosphate in caspase-9 activation and initiation of the apoptotic protease cascade. The results of this study show that on death induction by chemotherapeutic drugs, staurosporine and triggering of the death receptor CD95, ...

This paper provides a detailed analysis of the Polish Bank Conciliation Agreement (BCA) and relations between state owned enterprises and banks during transition. The study is conducted from the perspective of the enterprises implementing BCAs with debt /equity swaps and regular BCAs using data from surveys conducted by the authors during 1994 to 1995. Results of the analysis suggest that bank conciliation agreements did not protect creditors and sometimes supported bad firms at the cost of good ones. ...

Poland was the first country in Central Eastern Europe' where radical market-oriented reforms were launched, and it was also the country in which positive results of the new economic policy first emerged. GDP started rising as early as 1992, and subsequent growth remains healthy at a rate in excess of 4-5 percent a year. As a result, the Polish economy, despite numerous problems and unresolved issues, is more and more frequently quoted as an example of successful transition from the central plan ...

Archaeological sites at Affad (Sudan) are the only ones in northeastern Africa providing ostological remains of both African aurochs (Bos primigenius), dated to 50 kya, and domestic cattle, dated to 7–6 kya. The evidence enables studies of behavioral diversity between taurids. Strontium isotope analyses of the tooth enamel of both Pleistocene and Holocene ruminants suggest the local origin of these animals. The archaeozoological analysis reveals the temporal variability of environmental conditions ...

Induction of apoptosis is a hallmark of cytostatic drug and radiation-induced cell death in human lymphocytes and lymphoma cells. However, the mechanisms leading to apoptosis are not well understood. We provide evidence that ionizing radiation induces a rapid activation of caspase-8 (FLICE) followed by apoptosis independently of CD95 ligand/receptor interaction. The radiation induced cleavage pattern of procaspase-8 into mature caspase-8 resembled that following CD95 crosslinking and resulted in ...

Metabolic stability tests are one of the fundamental steps at the preclinical stages of new drug development. Microsomes, used as a typical enzymatic model of liver biotransformation, can be a challenging matrix for analytical scientists due to a high concentration of cellular proteins and membrane lipids. In the work, we propose a new procedure integrating biotransformation reaction with SPME-like protocol for sample clean-up. It is beneficial to increase the overall quality of results in contrary ...

The study enabled obtaining a number of new derivatives of 4-aryl-pyrido[1,2-c]pyrimidine 9.1–9.27 having conformationally restricted tryptamine moiety. In vitro studies (RBA) have shown that derivatives 9.1, 9.2, 9.4, 9.7, 9.9, 9.14 and 9.27 exhibit high affinity to molecular targets 5-HT1A receptor and SERT protein. In general, compounds with an unsubstituted or a para-substituted benzene ring of the pyrido[1,2-c]pyrimidine residue in the terminal part were characterized by higher binding ability, ...

Molecular docking has often been used before to calculate in silico affinity of drugs towards their molecular target, but not to estimate leading CYP isoform responsible for metabolism of studied compounds. The aim of this study is to present molecular docking as a valid alternative for costly in vitro studies resulting in estimation of leading CYP isoform. In vitro part was based on incubations of studied compounds with isolated CYP3A4 isoform followed by LC-MS analysis. The in silico stage consisted ...

Extended studies in the 4-aryl-pyrido[1,2-c]pyrimidine group resulted in 27 new compounds (10.1-10.27), 5,6,7,8-tetrahydropyrido[1,2-c]pyrimidine derivatives. In vitro tests (RBA) were carried out for 10.1-10.27 compounds in order to determine their affinity to 5-HT1A receptor and SERT protein. 10.1-10.3, 10.6, 10.7, 10.16 and 10.27 compounds had high binding ability to both molecular targets (5-HT1A Ki = 8–87 nM; SERT Ki = 8–52 nM). For these compounds (10.1-10.3, 10.6, 10.7, 10.16, 10.27) further in vitro, ...

Two series of novel 4-aryl-2H-pyrido[1,2-c]pyrimidine (6a–i) and 4-aryl-5,6,7,8-tetrahydropyrido[1,2-c]pyrimidine (7a–i) derivatives were synthesized. The chemical structures of the new compounds were confirmed by 1H and 13C NMR spectroscopy and ESI-HRMS spectrometry. The affinities of all compounds for the 5-HT1A receptor and serotonin transporter protein (SERT) were determined by in vitro radioligand binding assays. The test compounds demonstrated very high binding affinities for the 5-HT1A receptor ...

The uppermost Eifelian Kostyukovichi Formation corresponds to a distinct marine transgressive-regressive cycle in predominantly continental to restricted-marine Middle Devonian succession of the intracratonic Belarusian Basin in a near-equatorial area of SE Euramerica. The presence of the conodont ensensis Zone points to the correlation with the global Kačák Episode whose onset coincided with the eustatic If transgression. The regions covered by the United Nations Economic Commission for Europe (UNECE)—the whole European continent, North America and Central Asia— are characterized by a tremendous diversity in levels of economic development. While most countries of Western Europe and North America have levels of gross domestic product (GDP) per capita well above $20,000, for Eastern Europe, the Caucasus and Central Asia (EECCA) and South Eastern Europe (SEE), the level is below $10,000. Because they have failed to address the fundamental economic imbalances within Europe obscured by the single currency, each effort by European leaders so far to resolve the euro crisis has only deepened it. Without a decisive move toward fiscal and political union, accompanied by policies that push productivity and competitiveness toward convergence while closing the democratic deficit, the Eurozone will disintegrate. To discuss the way forward, the Nicolas Berggruen Institute's Council on the Future ...
