Mycobacterium tuberculosis (Mtb) evades the immune response by impairing the functions of different antigen‐presenting cells. We have recently shown that Mtb hijacks differentiation of monocytes into dendritic cells (DCs). To further characterize the mechanisms underlying this process, we investigated the consequences of inducing dendritic cell differentiation using interferon‐α and granulocyte‐macrophage colony‐stimulating factor in the presence of supernatants (SNs) obtained from monocyte cultures ...

Abstract TLRs interact with a growing list of pathogen-derived products and these interactions drive the activation of innate and adaptive immune responses. Dendritic cells (DC) play a key role in these events expressing a heterogeneous repertoire of TLRs. We have previously demonstrated the production of type I IFNs in DC following bacterial infections and TLR triggering. In this study, we sought to characterize the transcriptome specifically induced in human DC by IFN-β production stimulated upon ...

Abstract In human monocyte-derived dendritic cells (DC), infection with Mycobacterium tuberculosis and viruses or stimulation with Toll-like receptor type 3 and 4 agonists causes the release of type I interferon (IFN). Here, we describe that the IFN-β released upon stimulation with lipopolysaccharide (LPS) or polyinosinic:polycytidylic acid (poly I:C) is responsible for a rapid and sustained signal transducer and activator of transcription 1 and 2 activation and expression of IFN-stimulated genes, such ...

The Mycobacterium tuberculosis genome encodes 13 sigma factors. We have previously shown that mutations in some of these transcriptional activators render M. tuberculosis sensitive to various environmental stresses and can attenuate the virulence phenotype. In this work, we focused on extracytoplasmic factor sigmaE and studied the effects induced by the deletion of its structural gene (sigE) in the infection of human monocyte-derived dendritic cells (MDDC). We found that the wild-type M. tuberculosis ...

Background. Studies on Hispanics/Latinos with diabetes have largely focused on Mexican-American populations. This qualitative study explored psychosocial issues that affect diabetes self-management for Hispanic men and women of primarily Caribbean ancestry. Methods. Thirty-seven adults with diabetes in Bronx, N.Y., were recruited to seven focus groups, which were conducted in Spanish and English, audiotaped, transcribed, and subjected to qualitative analysis. Results. New themes emerged that are ...

Aspergillus fumigatus is the most prevalent airborne fungal pathogen and causes fatal invasive aspergillosis in immunocompromised patients. Given the essential role of dendritic cells (DC) in initiating and regulating immune responses, we investigated the impact of A. fumigatus conidial infection on human DC. A. fumigatus conidia were rapidly internalized and induced the release of tumor necrosis factor alpha within the first 8 h. After A. fumigatus infection, the majority of DC underwent full maturation, ...

Abstract In humans, the type I interferon (IFN) family consists of 13 IFN‐α subtypes, IFN‐β and IFN‐o the newly discovered IFN‐like family consists of IFN‐λ1, ‐λ2 and ‐λ3. We have investigated the expression of type I and λ IFN genes following virus infections or Toll‐like receptor (TLR) triggering in monocyte‐derived DC (MDDC) and plasmacytoid DC (pDC). We found thatall IFN‐α, ‐β, ‐o and ‐λ subtypes are expressed in influenza‐virus‐infected MDDC or pDC. Conversely, differential type I IFN gene transcription ...

The roles of plasmacytoid dendritic cells (pDCs) and their response to interferon (IFN)-β therapy in multiple sclerosis (MS) patients are poorly understood. We identified pDC accumulation in white matter lesions and leptomeninges of MS brains and abundant expression of the Type I IFN-induced protein MxA, mainly in perivascular CD3+ lymphocytes in lesions, indicating Type I IFN production by activated pDCs. The pDC chemoattractant chemerin was detected in intralesional cerebrovascular endothelial cells, ...

Detection of pulmonary artery aneurysms (PAA) in pulmonary arterial hypertension (PAH) is increasing. We sought to determine the frequency of PAA in a PAH cohort, variables related to its development and its prognostic impact. We conducted a retrospective analysis of PAH patients who underwent a computed tomography or magnetic resonance. PAA was defined as a pulmonary artery >40 mm. Baseline, echocardiographic, and hemodynamic findings at PAH diagnosis were compared. Freedom from death or lung transplant ...

Nutritional secondary hyperparathyroidism (NSH) is uncommon in horses. Aim of this report is evaluate an alternative therapeutic approach and provide more information on the diagnostic procedures, through detailed microscopic findings of the bone lesions pony with NSH. Administration of tiludronate along with a balanced diet can be used in a treatment protocol for ponies with NSH. This case report suggests that tiludronate can be used to ameliorate the clinical signs of NSH.

Abstract The Etruscan site of Pyrgi extended over 10 ha along the Tyrrhenian coastline. Fieldwork has so far brought to light the sanctuary and a ceremonial complex at the southern edge of the settlement, whereas additional evidence is provided by a section cut by sea erosion across the site. The archaeological layer is about 2 m thick and includes dry masonry buildings arranged into lots; the most superficial features are disturbed by deep agricultural activities and the top layer includes scattered ...

Because of the aging population, the incidence and prevalence of dementia has increased. The most common form of dementia is Alzheimer's disease. The present review focuses on the progression and prognosis of this disease, although many of the issues discussed can be extrapolated to other types of dementia. Despite public education efforts in recent years, dementia continues to have a tremendous impact on the patient, the primary caregiver and/or family and society in general. A diagnosis of dementia ...

Growing evidences put B lymphocytes on a central stage in multiple sclerosis (MS) immunopathology. While investigating the effects of interferon‐β (IFN‐β) therapy, one of the most used first‐line disease‐modifying drugs for the treatment of relapsing‐remitting MS, in circulating B‐cell sub‐populations, we found a specific and marked decrease of CD27 + memory B cells. Interestingly, memory B cells are considered a population with a great disease‐driving relevance in MS and resulted to be also target ...
