Recent in vitro work and a short clinical study suggest that adding a bonding agent layer between sealant and saliva-contaminated enamel allows for adequate bond strength and retention of resin sealants and may improve success of all sealant applications. This five-year clinical study scored 617 occlusal and 441 buccal/lingual molar sealants, with use of a split-mouth design, with half receiving sealant alone and half bonding agent plus sealant. Treatment effects and potential risk factors for sealant ...

Transportation decisions have health consequences that are often not incorporated into policy-making processes. Health Impact Assessment (HIA) is a process that can be used to evaluate health effects of transportation policy. We present a rapid HIA, conducted over eight weeks, evaluating health and economic effects of proposed fare increases and service cuts to Boston, Massachusetts’ public transportation system. We used transportation modeling in concert with tools allowing for quantification and ...

Abstract: Previously, opioid peptide analogues, β‐endorphin, and synthetic opiates were found to inhibit DNA synthesis in 7‐day fetal rat brain cell aggregates via κ‐and μ‐opioid receptors. Here dynorphins and other endogenous opioid peptides were investigated for their effect on DNA synthesis in rat and guinea pig brain cell aggregates. At 1 µ M , all dynorphins tested and β‐endorphin inhibited [ 3 H]thymidine incorporation into DNA by 20–38% in 7‐day rat brain cell aggregates. The putative ε‐antagonist ...

Abstract Treatment of rat C6 glioma cells with the tricyclic antidepressant desipramine induces opioid binding. Here the distribution of these opioid‐binding sites on C6 cell membranes and a functional property were investigated. Immunohistochemical examination of C6 cells was performed using a monoclonal anti‐idiotypic antibody to opioid receptors (Ab2AOR). Ab2AOR uniformly labeled >97% of the cells exposed to desipramine over their entire surface. The opioid‐receptor antagonist naltrexone completely ...

ABSTRACT High concentrations of fluoride in the body may cause toxic effects. Here, we investigated the effects of fluoride on the structure, function, and proteome of a cortical collecting duct epithelium in vitro . Kidney tubule cells (M‐1) were chosen because the concentration of fluoride in the kidney is 4–5‐fold higher than that in plasma. Mouse M‐1 cell monolayers were incubated in fluoride‐containing media, and the amiloride‐sensitive short‐circuit current and transepithelial resistance were ...

Abstract Fluoride exposure is widespread, with drinking water commonly containing natural and artificially added sources of the ion. Ingested fluoride undergoes absorption across the gastric and intestinal epithelia. Previous studies have reported adverse gastrointestinal effects with high levels of fluoride exposure. Here, we examined the effects of fluoride on the transepithelial ion transport and resistance of three intestinal epithelia. We used the Caco‐2 cell line as a model of human intestinal ...

Immune-mediated gene therapy using adenovirus expressing Flt3 ligand and thymidine kinase followed by ganciclovir administration (Flt3/TK) effectively elicits tumor regression in preclinical glioma models. Herein, we assessed new strategies to optimize Flt3L/TK therapeutic efficacy in a refractory RG2 orthotopic glioblastoma model. Specifically, we aimed to optimize the therapeutic efficacy of Flt3L/TK treatment in the RG2 model by overexpressing the following genes within the brain tumor microenvironment: ...

Abstract: Thymidine incorporation into DNA was inhibited dose‐dependently by β‐endorphin in rat fetal brain cell aggregate cultures. The inhibition was reversed partially by μ (cyclic D‐Phe‐Cys‐Tyr‐D‐Trp‐Orn‐Thr‐Pen‐Thr amide) or k (norbinaltorphimine) antagonists. Complete blockade of the β‐endorphin inhibitory effect was achieved only on concomitant exposure to both antagonists. Eadie–Hofstee analysis revealed that β‐endorphin inhibited thymidine incorporation noncompetitively. In the presence of ...

Neonatal cardiomyocytes are instrumental for disease modeling, but the effects of different cell extraction methods on basic cell biological processes remain poorly understood. We assessed the influence of two popular methods to extract rat neonatal cardiomyocytes, Pre‐plating (PP), and Percoll (PC) on cell structure, metabolism, and function. Cardiomyocytes obtained from PP showed higher gene expression for troponins, titin, and potassium and sodium channels compared to PC. Also, PP cells displayed ...

The successful implementation of clean cooking programs can considerably impact health, CO2 emissions, and household safety. However, this success will depend on the extent to which clean cooking programs target the right populations, and account for potential adoption barriers. To identify who these populations are, the present work relies on comparable household datasets from Guatemala, Peru, and Ecuador, and econometric analysis. Findings show that the characteristics of populations that clean ...

Abstract Although ultra-processed foods represent more than half of the total energy consumed by the UK population, little is known about the trend in food consumption considering the degree of food processing. We evaluated the trends of the dietary share of foods categorised according to the NOVA classification in a historical series (2018–2019) among the UK population. Data were acquired from the NDNS, a survey that collects diet information through a 4-d food record. We used adjusted linear regression ...

Systemic lupus erythematosus (SLE) is characterized by increased cardiovascular risk in adult-onset and childhood-onset SLE. Type I interferons (IFNs) appear to play a prominent role in premature vascular damage in adult-onset SLE, at least in part, by inducing impairments in the phenotype and function of endothelial progenitor cells (EPCs), thereby hampering vascular repair. It is not clear whether EPC dysfunction is present in childhood-onset SLE in association with a type I IFN signature. The phenotype ...

In order to explain recent experimental data, electric-field-gradient (EFG) calculations are performed for amorphous ${\mathrm{Zr}}_{70}$${\mathrm{Cu}}_{30}$ and its closer crystalline compound c-${\mathrm{Zr}}_{2}$Cu. To be able to calculate the EFG of non-axially-symmetric systems, we set up a scheme of approximations within a linear combination of atomic orbitals description of d electrons. The model used for the atomic structure of the amorphous alloy is a distribution of hard spheres relaxed by ...

We study the electronic structure of Cu-O clusters that appear in the high TC superconductors by the iterative extended Hückel method. This allows us to estimate the Hubbard parameter U for intrasite ḏ electron repulsion, and the electric field gradient at different sites of the crystal. The results are consistent with NQR and Mössbauer spectra, in this case if a small amount of Fe is introduced in YBa2Cu3O7. This supports the large covalency, small charge transfer and large ḏ level occupancy ...
