We examined exendin(9-39), an antagonist of glucagon-like peptide-1 (GLP-1) receptor (GLP-1R), as a potential probe for imaging of pancreatic β-cells. To evaluate in vitro receptor specificity, binding assay was performed using dispersed mouse islet cells. Binding assay showed competitive inhibition of [125I]BH-exendin(9-39) binding by non-radioactive exendin(9-39). To assess in vivo selectivity, the biodistribution was evaluated by intravenous administration of [125I]BH-exendin(9-39) to mice. Radioactivity ...

Hypoxia-inducible factor-1 (HIF-1) plays an important role in malignant tumor progression and in the development of resistance to radiotherapy. We designed a novel fusion protein (PTD-ODD-SAV [POS]) consisting of a protein transduction domain (PTD), streptavidin (SAV), and a portion of the oxygen-dependent degradation domain (ODD) of HIF-1α that confers the same oxygen-dependent regulation as HIF-1α on POS. (3- 123/125 I-iodobenzoyl)norbiotinamide ( 123/125 I-IBB) was conjugated to the SAV moiety of POS ...

Acute burn injury is usually associated with pain in the injured and nearby areas. However, we have recently reported that a thoracic scald induces hindpaw hyperalgesia during the healing stage in rats. The present study investigated the cause of the remotely occurring hyperalgesia. Behavioral testing using the von Frey test revealed that rats developed hyperalgesia in the neck and flank as well as the hindpaw 2–3 weeks after injury. The concentration of nerve growth factor (NGF) in the skin of the ...

Radical scavenging activities of 34 natural antioxidants were investigated from an electrochemical viewpoint. While the correlation of the oxidation potentials with their DPPH radical scavenging activities (represented by EC50) was not high (the correlation coefficient, r=0.73), the number of electrons n required for oxidation of an antioxidant, being obtained by continuous flow-column electrolysis with a slower flow rate (0.05 ml min−1), did show a good correlation with EC50 (1/EC50=1.67n+0.50 with ...

The reactions of N2(a′ 1Σu−, v=0) with H2, CH4, and their isotopic variants were examined. N2(a′, v=0) was produced by energy transfer from N2(a 1Πg, v=0), while N2(a, v=0) was produced by two-photon excitation of ground state N2. The rate constant for the deactivation of N2(a′,v=0) can be determined by measuring the decay profiles of N2(a, v=0) under the conditions that equilibration between N2(a, v=0) and N2(a′, v=0) can be assumed. The detection of N2(a, v=0) was accomplished by a laser-induced ...

The oxidation mechanism of caffeic acid (CAF) has been studied by means of cyclic voltammetry with the plastic formed carbon or glassy carbon electrode. CAF gives a well-developed two-electron reversible wave in acidic media, whereas it shows an irreversible behavior, i.e., a decrease of the rereduction peak, in less acidic media, suggesting that the oxidation of CAF follows an irreversible chemical reaction(s). Digital simulation analyses based on different oxidation mechanisms have been performed ...

Background: α1-AT is a 52-kDa acute-phase protein and a typical serine proteinase inhibitor, which is present in human serum. In vivo, the inhibitor prevents tissue damage by inactivating proteinases, such as elastase, that are released from activated neutrophils in the presence of inflammation. Methods: We obtained a monoclonal antibody against oxidized α1-AT(3F4) using chloramine T-oxidized α1-AT as the antigen. Results: This antibody did not react with either the native α1-AT or the elastase– ...

Vanilloid receptor 1 (VR1) is essential to the development of inflammatory hyperalgesia. We investigated whether inflammation can increase in VR1 positive neuronal profiles in rat DRG neurons using histochemical methods. We also used size frequency analysis and double staining with several neuronal markers to investigate whether or not inflammation alters VR1 expression. Inflammation induced a 1.5-fold increase in percentage of VR1-like immunoreactivity (LI) positive profiles per total neuronal profiles, ...

We have developed an electron-tracking Compton camera (ETCC) for medical use. Our ETCC has a wide energy dynamic range (200–1300 keV) and wide field of view (3 sr), and thus has potential for advanced medical use. To evaluate the ETCC, we imaged the head (brain) and bladder of mice that had been administered with F-18-FDG. We also imaged the head and thyroid gland of mice using double tracers of F-18-FDG and I-131 ions.

γ-Secretase plays an important role in the development of Alzheimer disease (AD). γ-Secretase activity is enriched in autophagic vacuoles and it augments amyloid-β (Aβ) synthesis. Autophagy-lysosomal dysfunction has been implicated in AD, but whether γ-secretase activity is affected by autophagy remains unclear. Here we report that γ-secretase activity is enhanced in basal autophagy-disturbed cells through the α subunit of eukaryotic translation initiation factor 2 (eIF2α) kinase, general control nonderepressible ...

Nicotinic acetylcholine receptors (nAChRs) are of great interest because they are implicated in various brain functions. They also are thought to play an important role in nicotine addiction of smokers. Chronic (-)-nicotine, a nAChR agonist, treatment in mice and rats elicits a dose-dependent increase in nAChRs in the brain. Upregulation of nAChRs in postmortem human brains of smokers has also been reported. However, changes in nAChRs after cigarette smoking cessation in humans are poorly understood. ...

In this study, we investigated the effect of vesicular zinc on ischemic neuronal injury. In cultured neurons, addition of a low concentration (under 100 μM) of zinc inhibited both glutamate-induced calcium influx and neuronal death. In contrast, a higher concentration (over 150 μM) of zinc decreased neuronal viability, although calcium influx was inhibited. These results indicate that zinc exhibits biphasic effects depending on its concentration. Furthermore, in cultured neurons, co-addition of glutamate ...
