The adoption of digital pathology offers benefits over labor-intensive, time-consuming, and error-prone manual processes. However, because most workflow and laboratory transactions are centered around the anatomical pathology laboratory information system (APLIS), adoption of digital pathology ideally requires integration with the APLIS. A digital pathology system (DPS) integrated with the APLIS was recently implemented at our institution for diagnostic use. We demonstrate how such integration supports ...

Abstract BACKGROUND Circulating free DNA sequencing (cfDNA-Seq) can portray cancer genome landscapes, but highly sensitive and specific technologies are necessary to accurately detect mutations with often low variant frequencies. METHODS We developed a customizable hybrid-capture cfDNA-Seq technology using off-the-shelf molecular barcodes and a novel duplex DNA molecule identification tool for enhanced error correction. RESULTS Modeling based on cfDNA yields from 58 patients showed that this technology, ...

This paper presents several new algorithms for the regularized reconstruction of a surface from its measured gradient field. By taking a matrix-algebraic approach, we establish general framework for the regularized reconstruction problem based on the Sylvester Matrix Equation. Specifically, Spectral Regularization via Generalized Fourier Series (e.g., Discrete Cosine Functions, Gram Polynomials, Haar Functions, etc.), Tikhonov regularization, Constrained Regularization by imposing boundary conditions, ...

This paper addresses the issue of regularization in the surface reconstruction from gradients problem in Industrial Photometric Stereo. Regularization of the solution is a necessary step in an industrial environment, where algorithms must cope with non-Gaussian noise, such as outliers, or non-Lambertian textures such as corrosion. Introducing Tikhonov regularization into the global least squares solution suppresses the influence of outliers in the reconstruction. Viable methods should both minimize ...

Applying neurotrophins to the round window immediately after a single noise exposure will prevent noise-induced hidden hearing loss.Loud noise can eliminate neural connections between inner hair cells and their afferent neurons (thereby diminishing sound perception) without causing a detectable change on audiogram. This phenomenon is termed hidden hearing loss.Guinea pigs were exposed for 2 hours to 4 to 8 kHz noise at either 95 or 105 dB SPL. Immediately afterward a 4 μl bolus of neurotrophins (brain- ...

A new method to fit specific types of conics to scattered data points is introduced. Direct, specific fitting of ellipses and hyperbolae is achieved by imposing a quadratic constraint on the conic coefficients, whereby an improved partitioning of the design matrix is devised so as to improve computational efficiency and numerical stability by eliminating redundant aspects of the fitting procedure. Fitting of parabolas is achieved by determining an orthogonal basis vector set in the Grassmannian ...

Degenerative lumbar spondylolisthesis (DS) patients are treated with instrumented fusion, following EBM guidelines, and typically have excellent clinical outcomes. However, not all lumbar fusion procedures adhere to EBM guidelines, typically due to a lack of prospective data. This retrospective study compared outcomes of DS lumbar fusion patients treated according to EBM guidelines (EBM concordant) to lumbar fused patients with procedures that did not have clear EBM literature that supported this treatment, ...

Aggregation of tau protein in the brain is associated with a class of neurodegenerative diseases known as tauopathies. FK506 binding protein 51 kDa (FKBP51, encoded by FKBP5) forms a mature chaperone complex with Hsp90 that prevents tau degradation. In this study, we have shown that tau levels are reduced throughout the brains of Fkbp5-/- mice. Recombinant FKBP51 and Hsp90 synergized to block tau clearance through the proteasome, resulting in tau oligomerization. Overexpression of FKBP51 in a tau ...
