The effect of light on nicotine production in cultured tobacco callus tissues was investi-gated. Illumination strikingly inhibited the nicotine production even though the growth of the tissues was slightly stimulated by the light. Callus tissues were induced from stem and root segments of Rauwolfa serpentina. Growth and alkaloid production of the callus tissues were examined under various culture conditions. A homogeneous transparent gel was formed when an organic solution of cellulose acetate was in contact with a metal alkoxide solution. The gel is insoluble in common solvents and electrolyte solutions. It seems that the reaction is due to coordination bonding between hydroxy groups on the pyranose ring and the metal. Enzyme could be immobilized in the gel fibres. The immobilized enzyme is stable for a long period and can be used in a packed column.

A tobacco callus strain, OMT-53, was selected from many cultures as a desirable strain having high nicotine producing capacity. Several culture conditions were examined, aiming to get higher nicotine production with the callus strain, OMT-53. It was revealed that the nicotine production was remarkably enhanced when the callus tissues were cultured at a limited concentration of α-NAA in culture medium. Resonance Raman spectra have been measured to characterize the heme environment in aldoxime dehydratase (OxdA), a novel hemoprotein, which catalyzes the dehydration of aldoxime into nitrile. The spectra showed that the ferric heme in the enzyme is six‐coordinate low spin, whereas the ferrous heme is five‐coordinate high spin.
